        Most of the cholesterol is synthesized by liver in the body while about one of third is taken via dietary. The main functions of cholesterol is to protect membranes in cell surface, avoid the arterial bleeding by hypertension, and prolong the life of erythrocytes, and so on. However, overload of cholesterol leads to arteriosclerosis associated with leading death cause. Lack of physical activity, emotional and environmental stress, and low intake of protein or vitamin E induce the unbalance between HDL- and LDL-cholesterol so become a basis of ischemic disorders in heart, brain and elsewhere in the body. So far, four major classes of medications for hyperlipidemia are HMG-CoA reductase inhibitors (statins), bile acid sequestrants, nicotinic acid, and fibric acids. The statins can lower LDL and levels triglyceride, but may induce myopathy and an elevation of liver enzyme levels. The bile acid sequestrants lower LDL levels and raise HDL levels with no effect on triglyceride levels but side effects of gastrointestinal (GI) distress, constipation, and a decrease in the absorption of other drugs. Nicotinic acid and fibric acids lower LDL and triglyceride levels with showing flushing, hyperglycemia, hyperuricemia, GI distress, and hepatotoxicity dyspepsia, gallstones, myopathy, and unexplained noncardiac death as adverse effects. Above western drugs lower cholesterol by 15 to 30% while all have notable adverse effects. In traditional medicine, hyperlipidemia is regarded as retention of phlegm and fluid disease. Etiology and pathogenesis of hyperlipidemia is basically based on Spleen-Deficiency and Phlegm-Stagnation, accumulation and stasis of -heat, and Qi & blood stagnation induced by Phlegm-damp, water-dampness, and blood stasis. Thereby, strengthening Spleen and dissolving Phlegm, clearing away heat and diuresis, and supplementing Qi and activating blood circulation are commonly used therapeutic methods for hyperlipidemia. The traditional herbal medicine, have been used for patients with CVA, hypertension or hyperlipidemia in Oriental hospital or Oriental clinic. The lock and key theory is used to develop most of western medicine, however many diseases are caused by mixed factors in body-complex system. We expect that Oriental pharmacological theory could be newborn as a novel drug showing high advantage of blood levels of lipidsand QOL of performance without side effects.

        과루인 및 과루인청혈플러스가 인간 탯줄 정맥 내피 세포(Human umbilicl vein endothelial cells, HUVECs)에서 염증인자 발현 억제에 미치는 영향

        Objective: To examine the effects of Trichosanthes kirilowii Maximowicz extract (TE) and Trichosanthes kirilowii Maxi mowicz Cheonghyeol Plus Phellinus linteus Cheonghyeol plus (TCP) on anti-inflammatory factor expression in human umbilical vein endothelial cells (HUVECs). Methods: HUVECs were activated with TNF-α and then treated with TE and TCP. The expression levels were then measured for intracellular genes (KLF2, eNOS, MCP-1, ICAM-1, and VCAM-1), proteins (KLF2, eNOS, MCP-1, ICAM-1, VCAM-1, ERK, and JNK, p38), and extracellular biomarkers (ICAM-1, VCAM-1, and MCP-1). Results: 1. TCP at concentrations of 100 ㎍/mL or greater significantly increased the expression of KLF2 and eNOS intracellular genes and significantly decreased the expression of ICAM-1, VCAM-1, and MCP-1 genes compared to the control group. 2. TCP at concentrations of 100 ㎍/mL or greater significantly increased the expression of KLF2, eNOS proteins compared to the control group, and significantly reduced the expression of VCAM-1, ICAM-1, MCP-1, ERK, and p38 proteins. However, JNK protein phosphorylation showed no significant change compared to the control group. 3. TCP at concentrations of 100 ㎍/mL or more significantly decreased the production of MCP-1, ICAM-1, and VCAM-1 extracellular biomarkers compared to the control group. 4. TE at a concentration of 100 ㎍/mL did not cause any significant change in the expression of intracellular genes or proteins, in the production of the extracellular biomarker MCP-1, or in the amount of JNK protein compared to the control group. Other intracellular genes, proteins, and extracellular biomarker expression showed the same trend as observed with TCP exposure. Conclusion: This study experimentally confirmed that TE and TCP could be effective in preventing or inhibiting various inflammatory vascular diseases due to their anti-inflammatory effects.

        간양상항방(肝陽上亢方)이 고혈압(高血壓)에 미치는 영향

        This study was done to investigate the effect of Ganyangsanghang-bang(GYSHB) on hypertension. After administering GYSHB extract to SHR for 5 weeks, changes in blood pressure, pulse rate, aldosterone and catecholamine levels in plasma were examined, and immunohistochemical changes were observed, and liver function test was done. The following results were obtained; blood presure decreased significantly as well as levels of aldosterone and norepinephrine in SHR. But levels of dopamine were unaffected. No capillary vessel dilation in adrenal cortex was observed. Safety against hepatic toxicity was showed. These results support a role for GYSHB might be usefully applied in treatment of hypertension.

        가미계혈등탕(加味鷄血藤湯)이 고혈압(高血壓)에 미치는 영향(影響)

        Objectives : This study was done to investigate the effect of Gamigehyuldeung-tang on hypertension. Methods : After administering Gamigehyuldeung-tang extract to SHR(Spontaneous Hypertensive Rats) for 5 weeks, changes in blood pressure, pulse rate, aldosterone and catecholamine levels in plasma were examined, and immunohistochemical changes and scanning electron microscopic changes were observed. Results : The following results were obtained; blood presure decreased significantly as well as levels of aldosterone, dopamine and epinephrine in SHR. But levels of norepinephrine were unaffected. No capillary vessel dilation was observed. A decrease in cell damage was seen in microscope investigation. Conclusions : These results support a role for Gamigehyuldeung-tang might be usefully applied in treatment of hypertension.

        db/db mouse에서 인진(茵蔯), 울금(鬱金), 산사(山査) 복합추출물의 항고지혈 및 항산화 효과

        Objective: This study was performed to investigate the effect of IUS (Inulsan, an extract of Artemisiae capillaris (茵蔯), Curcumae longae (鬱金), and Crataegi fructus (山査)) on anti-hyperlipidemia, anti-oxidation, and anti-inflammation.Method: We administered water extracts of Artemisiae capillaris, Curcumae longae, and Crataegi fructus for three weeks to db/db mice (C57BL/Ks), animal models induced with type 2 diabetes mellitus. Mice were divided into three groups: normal (C57BL/6J mice group), control group (db/db mice without administration of IUS) and IUS group (db/db mice treated with IUS). Then we measured total cholesterol, LDL cholesterol, HDL cholesterol, and triglyceride in the serum after the oral administration of IUS.Results: 1. IUS did not show any cytotoxicity in RAW 264.7 cells. 2. IUS decreased AST, ALP, and creatinine levelsand did not show any liver or renal toxicity in the db/db mice. 3. IUS increased DPPH and ABTS radical scavenging activity and decreased ROS production in RAW 264.7 cells. 4. IUS significantly decreased IL-1β, IL-6, and TNF-α production in RAW 264.7 cells. 5. IUS increased HDL cholesterol and significantly decreased total cholesterol and triglyceride in db/db mice. 6. IUS significantly decreased the atherogenic index and cardiac risk factor. 7. In contrast with the control group, fat infiltration in the liver and aorta decreased in IUS treated mice. The cell nucleus was located in the central area in H&E staining of liver. And endomembranes also were more thinner than the control group in H&E staining of aorta.Conclusions: These results suggest that IUS might be effective in the prevention and treatment of dyslipidemia.

        가미자음강화탕(加味滋陰降火湯)이 SHR의 혈압에 미치는 영향

        Objective : This study was done to investigate the effect of KJT on hypertension. Methods : Spontaneous Hypertensive Rats were sensitized and challenged with Kamijaeumganghwatang(KJT) for 5 weeks. Experimental group was treated with $200mg/day/2m{\ell}$ of KJT orally and control group was treated with $2m{\ell}/day$ of normal saline instead. Results : 1. KJT significantly showed significant protection against cytotoxicity and toxicity in the liver and the kidney. 2. KJT significantly decreased the blood pressure in SHR. 3. KJT significantly decreased the levels of aldosterone in SHR. 4. KJT significantly decreased the levels of dopamine, norepinephrine and epinephrine in SHR. 5. KJT significantly decreased the levels of sodium and potassium in SHR, but it did not decrease the levels of chloride in SHR. Levels of calcium in SHR increased, but only insignificantly. 6. KJT significantly decreased the levels of IL-6 in SHR, and significantly increased the levels of IL-10 in SHR. 7. For histological effects, KJT dillated capillary vessels in the kidney, significantly decreased eosinophilic changes in heart cells, and significantly decreased endothelial damage in the aorta. Conclusion: These results suggest that KJT is useful in treatment of hypertension.

        가미청열도담탕(加味淸熱導痰湯)이 DOCA-salt로 유발된 고혈압 백서에 미치는 영향

        This study was done to investigate the effect of Gamichungyeoldodam-tang (GCDT) in DOCA-salt induced hypertensive rats. The results were obtained as follows : GCDT showed safety against cytotoxicity and toxicity in the liver and the kidney. GCDT showed an inhibitory effect on ACE. GCDT significantly decreased the heart rate and the blood pressure in DOCA-salt induced hypertensive rats. GCDT significantly decreased the levels of aldosterone in DOCA-salt induced hypertensive rats. GCDT significantly decreased the levels of dopamine, norepinephrine, and epinephrine in DOCA-salt induced hypertensive rats. GCDT significantly decreased the levels of chloride in DOCA-salt induced hypertensive rats. These results suggest that GCDT might be effective in treatment of hypertension.

        가미옥녀전(加味玉女煎)이 고혈압백서(高血壓白鼠)의 혈압(血壓) 유관인자(有關因子)변화에 미치는 영향(影響)

        Objective : This study was done to investigate the effect of Gamioknyeo-jeon (GOJ) on spontaneously hypertensive rats. Methods : Spontaneously hypertensive rats were sensitized and challenged with GOJ for 4 weeks. the experimental group was treated with 56.7mg/kg/day of GOJ orally while the control group was treated with 56.7mg/kg/day of normal saline instead. Results : 1. GOJ significantly showed safety against cytotoxicity and toxicity in the liver. 2. GOJ significantly decreased the heart rate and the blood pressure in Spontaneously hypertensive rats. 3. GOJ significantly decreased the levels of aldosterone in Spontaneously hypertensive rats. 4. GOJ significantly decreased the levels of dopamine and norepinephrine in Spontaneously hypertensive rats. 5. GOJ significantly decreased the levels of potassium and chloride in Spontaneously hypertensive rats. 6. GOJ significantly decreased the levels of uric acid, BUN and creatinine in Spontaneously hypertensive rats. Conclusion : These results suggest that GOJ might be useful in treatment of hypertension.

        계혈등(鷄血藤)이 Rat의 말초신경 재생에 미치는 효과

        Objective : Gyehyuldeung (GHD) has been widely used in oriental medicine for the treatments of cardiovascular and neurological disorders. Thus, its potential facilitatory activity on axonal regeneration was investigated in the rats. Methods: Sprague-Dawley rats were given crush injury at the sciatic nerve and the changes of axon growth after nerve injury on each nerve injury model were investigated with anti-NF-200 antibody, DiI, GAP-43 protein and Cdc2 protein Results : GHD-mediated enhancement of axonal regeneration after crush injury was measured in both qualitative and quantitative ways by immunofluorescence staining with anti-NF-200 antibody and retrograde tracing of fluorescence dye DiI. GAP-43 protein levels were elevated by GHD treatments in the distal injured sciatic nerve and DRG sensory neurons. The neurite outgrowth of DRG sensory neurons was facilitated by GHD treatment when co-cultured with Schwann cells and astrocytes prepared from injured sciatic nerves and injured spinal cord tissues, respectively. It was observed that Cdc2 protein was up-regulated in co-cultured Schwann cells or astrocytes and Cdc2 protein signals were co-localized to a certain extent with those of phospho-vimentin protein. Conclusions : These results suggest that GHD may play a facilitatory role in axonal regeneration by acting on the injured axons and adjacent non-neuronal cells. The current findings may be useful for the development of therapeutic targets through more specific explorations on molecular interactions between herbal components and endogenous factors.
