Henna has been used long time as a cosmetic dye for skin painting, hair coating / coloring and nails. And natural henna is relatively safe and allergic reaction are rare. In this study, ROS scavenging effect and cell viability of henna were investigated for involvement of oxidative damage and nephrotoxicity by enzyme kinetics, cell culture and staining. Powder of natural henna was prepared with 80% ethanol, and ethanol extract was separated to 5 different fractions. Ethanol extract was analyzed general constituent, and ethanol extract and all fractions were used for measurement of ROS (Reactive Oxygen Species, DPPH and hydroxy radical) scavenging effect and cell viability and for observation of morphological change on vero cell. In the result, henna was contained phenolics (+), glycosides (+), organic acid (++). Ethanol extract and 5 different fractions were not shown 100% DDPH scavenging effect. Hydroxy radical scavenging effect was not observed in hexane and chloroform fraction, but OH scavenging effect was appeared in ethylacetate, butanol and water fraction. About 5% of cell viability were shown in concentration of 450 ㎍/ ㎖, 300 ㎍/ ㎖of hexane and chloroform fraction. And depressed cell growth and differentiation, cytoplasm lysis, nucleus breaking were observed on relative low concentration hexane (450 ㎍/㎖) and chloroform fraction (300 ㎍/㎖) treated Vero cell. No criteria of cytoplasm lysis, nucleus breaking were shown in concentration of 600 ㎍/㎖ ethylacetate, butanol and water fraction. In conclusion, henna may be involved partly in redox cycling and induce to oxidative damage. And lowsone that is a component of henna may be have low nephrotoxicity.
세포독성과 형태학적 변화 관찰을 통한 5종 에센셜 오일의 안전성 검색

In medical- and esthetic area the aromatherapy has spread widely by using of inhalation, bath and topical massage with essential oil. Thereby the safety, danger of irritation and sensitive reaction and injury of essential oil must be considered for clients and therapists. Therefore, in this study the safety of 5 species essential oils (ginger, cypress, rosemary, lavender and black pepper) was proven by the measurement of MTT-assay and sirius red staining. Liver cell, brain cell and kidney cell were chosen for the cell viability assay and morphological change observations. In this result, ginger and rosemary have shown as 50% of liver cell viability in concentration of 0.0012㎕/㎖ and 0.0014㎕/㎖. Cell lysis and nucleus breaking were shown in concentration of 0.002% cypress and lavender treated liver cell, and 0.001% black pepper and cypress treated brain cell. Ginger, rosemary and lavender were appeared as very low cytotoxicity on brain cell (in concentration of 0.0006 ㎕/㎖(ginger oil), 0.0011㎕/㎖(rosemary) and 0.00054 ㎕/㎖(lavender)). And lavender, rosemary and ginger revealed very low toxicity on kidney cell, and cell lysis and nucleus breaking did not observed in concentration of 0.002% lavender and rosemary treated kidney cell. Cypress has shown a low cytotoxicity on liver, brain and kidney cell. In conclusion, 5 essential oils such as ginger, cypress, rosemary, lavender and black pepper may be use carefully in very low dosage for inhalation and the concentrated essential oil should not be directly touched and exposed. Absolute essential oils must be diluted with carrier oil for topical and systematic massage.

Urbanization from agricultural/ forest areas has been causing increased runoff and pollutant loads from it. Thus, numerous models have been developed to estimate NPS loading from urban area and Long Term Hydrologic Impact Analysis L THIA model has been used to evaluate effects of landuse changes on runoff and pollutant loads. However, the L THIA model could not consider rainfall intensity in runoff evaluation. There fore, the L THIA model, capable of simulating runoff using 10 minute rainfall data, was applied to the study areas for evaluation of estimated runoff and NPS. The estimated Nash Sutcliffe coefficient NSE values were over 0.6 for runoff, BOD, TN, and TP for most sites and watershed. The calibrated model was further extended to other counties for pollutant load potential evaluation. Pollutant load potential maps were developed and target areas were identified. As shown in this study, the L THIA 2012 can be used for evaluation runoff and pollutant loads with limited data sets and its estimation could be used in identifying pollutant load hot spot areas for implementation of site specific Best Management Practices.
