A rapid antimicrobial susceptibility test based on single-cell morphological analysis

<P>A rapid antibiotic susceptibility test (AST) is desperately needed in clinical settings for fast and appropriate antibiotic administration. Traditional ASTs, which rely on cell culture, are not suitable for urgent cases of bacterial infection and antibiotic resistance owing to their relatively long test times. We describe a novel AST called single-cell morphological analysis (SCMA) that can determine antimicrobial susceptibility by automatically analyzing and categorizing morphological changes in single bacterial cells under various antimicrobial conditions. The SCMA was tested with four Clinical and Laboratory Standards Institute standard bacterial strains and 189 clinical samples, including extended-spectrum β-lactamase–positive <I>Escherichia coli and Klebsiella pneumoniae</I>, imipenem-resistant <I>Pseudomonas aeruginosa</I>, methicillin-resistant <I>Staphylococcus aureus</I>, and vancomycin-resistant <I>Enterococci</I> from hospitals. The results were compared with the gold standard broth microdilution test. The SCMA results were obtained in less than 4 hours, with 91.5% categorical agreement and 6.51% minor, 2.56% major, and 1.49% very major discrepancies. Thus, SCMA provides rapid and accurate antimicrobial susceptibility data that satisfy the recommended performance of the U.S. Food and Drug Administration.</P>

<P>Tuberculosis (TB) is a major global health problem, and multi-drug-resistant TB (MDR-TB) and extensively drug-resistant TB (XDR-TB) are spreading throughout the world. However, conventional drug susceptibility test (DST) methods, which rely on the detection of the colony formation on a solid medium, require 1-2 months to the result. A rapid and accurate DST is necessary to identify patients with drug-resistant TB and treat them with appropriate drugs. Here, we used microscopic imaging of Mycobacterium tuberculosis (MTB) immobilized in an agarose matrix for a rapid DST. The agarose matrix, which was molded in a microfluidic chip, was inoculated with MTB, and TB drugs in liquid culture medium diffused throughout the agarose to reach the MTB immobilized in the agarose matrix. After the responses of MTB to drugs were tracked with an automated microscopic system, an image-processing program automatically determined the susceptibility and resistance of MTB to specific doses of TB drugs. The automatic DST system was able to assess the drug susceptibility of various drug-resistant clinical TB strains within 9 days with an accuracy comparable to that of conventional method. Our rapid DST method based on microscopic time-lapse imaging greatly reduces the time required for a DST and can be used to rapidly and accurately treat TB patients.</P>

<P>Sepsis is one of the major causes of death in the US, necessitating rapid treatment with proper antibiotics. Conventional systems for antibiotic susceptibility testing (AST) take far too long (16-24 h) for the timely treatment of sepsis. This is because they rely on measuring optical density, which relates to bacterial growth, to determine the minimal inhibitory concentrations (MICs) of relevant antibiotics. Thus, there is a desperate need for more improved and rapid AST (RAST) systems. The RAST system can also reduce the growing number of clinical problems that are associated with antibiotic resistance caused by methicillin-resistant Staphylococcus aureus, vancomycin-resistant Staphylococcus aureus, and vancomycin-resistant enterococci. In this study, we demonstrate a microfluidic agarose channel (MAC) system that reduces the AST assay time for determining MICs by single bacterial time lapse imaging. The MAC system immobilizes bacteria by using agarose in a microfluidic culture chamber so that single cell growth can be tracked by microscopy. Time lapse images of single bacterial cells under different antibiotic culture conditions were analyzed by image processing to determine MICs. Three standard bacteria from the Clinical and Laboratory Standard Institute (CLSI) were tested with several kinds of antibiotics. MIC values that were well matched with those of the CLSI were obtained within only 3-4 h. We expect that the MAC system can offer rapid diagnosis of sepsis and thus, more efficient and proper medication in the clinical setting.</P>

본 연구는 조종사들이 브리핑 및 디브리핑시에 기동에 대하여 신속하게 활용할 수 있는 스케치 기반 애니메이션을 제작하여 사용할 수 있도록 제안한다. 기존의 연구에서는 항공기 가동을 위하여 복잡한 제작도구를 이용하여야 하며 이러한 것은 실제 다양한 공중상황에 대한 여러 가지 상황을 묘사하기에 어려움이 있다. 또한 항공기 기동은 상승, 강하, 수평비행등 3차원 환경이지만 실제 브리핑 경우에는 2차원으로 밖에는 표현할 수 없는 한계를 가지고 있어 조종사들의 인지에도 제한이 된다. 또한, 실제 항공기 기동할 수 없는 범위로 제작되는 경우 오히려 잘못된 기동을 묘사하게 된다. 따라서, 실제 항공기 기동과 유사하게 하기 위하여 항공기 추력을 적용한 3차원 애니메이션 제작 기법을 제안하다.
합성가스를 연료로 사용하는 고체산화물연료전지-가스터빈 하이브리드 시스템의 탈설계점 성능 특성

가스화기를 장착한 고체산화물연료전지와 가스터빈의 하이브리드 시스템의 사전 연구로서 합성가스에서 수소와 일산화탄소의 구성이 하이브리드 시스템의 성능특성에 미치는 영향을 조사하였다. 이때 다른 구성요소와 다른 발열량을 가진 연료의 특성이 가스터빈의 탈설계점 운전을 유발하여 결과적으로 전체 하이브리드 시스템의 다른 성능 특성을 야기할 것으로 예상된다. 합성가스의 구성요소가 고체산화물연료전지의 성능에 영향을 준다는 것과 일산화탄소를 사용하는 하이브리드 시스템의 성능이 수소를 사용하는 것보다 나쁘다는 것이 발견되었다. 부분부하 성능에서는 수소를 사용하는 경우 부분부하 운전시에 성능 저하가 일산화탄소의 경우보다 더 현저하였다. As a preliminary study on a SOFC-GT hybrid system integrated a with coal-gasification system, the influence of the concentrations of H₂ and CO in syngas on the performance characteristics of the hybrid system is investigated. It is expected that the differences in the heating values of fuels with different compositions trigger the offdesign operation of the gas turbine and result in different performance characteristics of the overall hybrid system. Syngas compositions are found to affect the SOFC performance. Performance of hybrid system with carbon monoxide is poorer than the case with hydrogen. In the case of part-load performance with syngas, performance degradation at part-load operating conditions with hydrogen is more dominant than the case with carbon monoxide.
LINEAR SPLINE을 이용한 스케치 기반 비행경로 시각화

본 연구는 스케치 기반으로 조종사들이 다양한 비행경로를 입력시 Linear Spline을 적용하여 브리핑 및 디브리핑시에 실시간으로 비행경로를 표시할 수 있도록 제안한다. 기존의 연구에서는 항공기 기동을 위하여 복잡한 제작도구를 이용하여야 하며 이러한 것은 실제 다양한 공중상황에 대한 여러 가지 상황을 묘사하기에 준비에 상당한 시간이 소요되는 단점이 있다. 따라서 기존 자료와 유사한 형태로 구현할 수 있고 브리핑 및 디브리핑시에 실시간으로 결과가 되는 비행경로를 시각화 할 수 있도록 하여 복잡한 환경에서의 기동을 보다 정확하게 인지하고 다양한 상황하에서의 최적의 결과를 도출할 수 있다.
