      • Well-designed Te/SnS<sub>2</sub>/Ag artificial nanoleaves for enabling and enhancing visible-light driven overall splitting of pure water

        <P><B>Abstract</B></P> <P>To produce hydrogen and oxygen from photocatalytic overall splitting of pure water provides a promising green route to directly convert solar energy to clean fuel. However, the design and fabrication of high-efficiency photocatalyst is challenging. Here we present that by connecting different nanostructures together in a rational fashion, components that cannot individually split water into H<SUB>2</SUB> and O<SUB>2</SUB> can work together as efficient photocatalyst with high solar-to-hydrogen (STH) energy conversion efficiency and avoid the use of any sacrificial reagent. Specifically, Te/SnS<SUB>2</SUB>/Ag artificial nanoleaves (ANLs) consist of ultrathin SnS<SUB>2</SUB> nanoplates grown on Te nanowires and decorated with numerous Ag nanoparticles. The appropriate band structure of Te/SnS<SUB>2</SUB> p-n junctions and the surface plasmon resonance of Ag nanoparticles synergistically enhance the quantum yield and separation efficiency of electron-hole pairs. As a result, Te/SnS<SUB>2</SUB>/Ag ANLs enable visible-light driven overall water-splitting without any sacrificial reagent and exhibit high H<SUB>2</SUB> and O<SUB>2</SUB> production rates of 332.4 and 166.2μmolh<SUP>−1</SUP>, respectively. Well-preserved structure after long-term measurement indicates its high stability.       • Fatty Acid Synthesis Pathway Genetic Variants and Clinical Outcome of Non-Small Cell Lung Cancer Patients after Surgery

        Over-expression of de novo lipogenesis (DNL) genes is associated with the prognosis of various types of cancers. However, the effects of single nucleotide polymorphisms (SNPs) in these genes on recurrence and survival of non-small cell lung cancer (NSCLC) patients after surgery are still unknown. In this study, a total of 500 NSCLC patients who underwent surgery treatment were included. Eight SNPs in 3 genes (ACACA, FASN and ACLY) of the DNL pathway were examined using the Sequenom iPLEX genotyping system. Multivariate Cox proportional hazards regression and Kaplan-Meier curves were used to analyze the association of SNPs with patient survival and tumour recurrence. We found that two SNPs in the FASN gene were significantly associated with the recurrence of NSCLC. SNP rs4246444 had a significant association with lung cancer recurrence under additive model (hazard ratio [HR], 0.82; 95% confidence interval [95%CI], 0.67-1.00; p=0.05). Under the dominant model, rs4485435 exhibited a significant association with recurrence (HR, 0.75; 95%CI, 0.56-1.01; p=0.05). Additionally, SNP rs9912300 in ACLY gene was significantly associated with overall survival in lung cancer patients (HR, 1.41; 95%CI, 1.02-1.94, p=0.04) under the dominant model. Further cumulative effect analysis showed moderate dose-dependent effects of unfavorable SNPs on both survival and recurrence. Our data suggest that the SNPs in DNL genes may serve as independent prognostic markers for NSCLC patients after surgery.

        Two new ursane-type triterpenoid saponins from Elsholtzia bodinieri

        Two new ursane-type triterpenoid saponins,bodiniosides M (1) and N (2), along with three knownsaponins, oblonganosides I (3), pseudobuxussaponin B (4)and bodinioside A (5), were isolated from the aerial partsof Elsholtzia bodinieri. The structures of compounds 1 and2 were characterized by spectroscopic data as well as acidhydrolysis and GC analysis as 3-O-b-D-xylopyranosyl-19ahydroxy-23-acetoxy-urs-12(13)-en-28-oic acid 28-O-a-Lrhamnopyranosyl-(1-2)-b-D-glucopyranoside and 3-O-b-Dglucopyranosyl-2a,19a-dihydroxy-urs-12(13)-en-28,20blactone. Compounds 1 and 5 exhibited potent anti-HCVactivities in vitro with a selective index of 6.53 and 4.41,respectively.

      • Effect of Grape Procyanidins on Tumor Angiogenesis in Liver Cancer Xenograft Models

        Background: In recent years a wide variety of flavonoids or polyphenolic substances have been reported to possess substantial anti-carcinogenic and antimutagenic activities. Grape proanthocyanidins (GPC) are considered as good examples for which there is evidence of potential roles as anti-carcinogenic agents. Methods: A xenograft model was established using H22 cells subcutaneously injected into mice and used to assess different concentrations of grape proanthocyanidins (GPC) and Endostar. Treatments were maintained for 10 days, then levels of vascular endothelial growth factor (VEGF) and microvessel density (MVD) were examined by immunohistochemistry, while VEGF mRNA was determined by real-time PCR in tumor tissue. Results: The expression of MVD and VEGF decreased gradually as the concentration of GPC increased.There was a significant positive correlation between MVD and VEGF. Conclusions: These results suggest that GPC restrains the growth of tumor, possibly by inhibiting tumour angiogenesis.

        HIV-negative plasmablastic lymphoma: report of 8 cases and a comprehensive review of 394 published cases

        BackgroundHuman immunodeficiency virus (HIV)-negative plasmablastic lymphoma (PBL) is a rare entity of diffuse large B-cell lymphoma (DLBCL). The clinicopathological features of and optimal treatment for HIV-negative PBL remain largely unknown.MethodsTo gain insight into this distinct lymphoma, we summarized the clinicopathologic charac-teristics of 8 unpublished HIV-negative PBLs and performed a comprehensive review of 394 published cases.ResultsOf the 8 unpublished PBLs, the median patient age was 53.0 years. Four patients pre-sented with stage IV disease. All 8 patients showed a plasma cell-like immunophenotype. Of the six patients who received anthracycline-based chemotherapy, including two who received bortezomib, three patients achieved a continuous complete response, two pa-tients died due to disease progression, and one patient was lost to follow-up. The other two patients achieved continuous complete response after receiving chemotherapy com-bined with radiotherapy and surgery. Of the 402 patients, the majority were male, with a mean age of 58.0 years. EBV infection was detected in 55.7% of the patients. The median survival times of the patients who received CHOP or CHOP-like regimens and intensive regimens were not reached and 23.0 months, respectively, and the intensive regimen did not improve the survival outcome (P=0.981). Multivariate analysis showed that EBER remained the only independent factor affecting overall survival (OS).ConclusionHIV-negative PBL is a distinct entity with a predilection for elderly and immunosup-pressed individuals. Intensive chemotherapy had no apparent survival benefits over the CHOP regimen in terms of OS; the prognosis of this disease is poor with current chemo-therapy methods, and treatment remains a challenge.

      • Knockdown of HMGN5 Expression by RNA Interference Induces Cell Cycle Arrest in Human Lung Cancer Cells

        HMGN5 is a typical member of the HMGN (high mobility group nucleosome-binding protein) family which may function as a nucleosomal binding and transcriptional activating protein. Overexpression of HMGN5 has been observed in several human tumors but its role in tumorigenesis has not been fully clarified. To investigate its significance for human lung cancer progression, we successfully constructed a shRNA expression lentiviral vector in which sense and antisense sequences targeting the human HMGN5 were linked with a 9-nucleotide loop. Inhibitory effects of siRNA on endogenous HMGN5 gene expression and protein synthesis were demonstrated via real-time RT-PCR and western blotting. We found HMGN5 silencing to significantly inhibit A549 and H1299 cell proliferation assessed by MTT, BrdU incorporation and colony formation assays. Furthermore, flow cytometry analysis showed that specific knockdown of HMGN5 slowed down the cell cycle at the G0/G1 phase and decreased the populations of A549 and H1299 cells at the S and G2/M phases. Taken together, these results suggest that HMGN5 is directly involved in regulation cell proliferation in A549 and H1299 cells by influencing signaling pathways involved in cell cycle progression. Thus, our finding suggests that targeting HMGN5 may be an effective strategy for human lung cancer treatment.

      • Dynamic Power System Zone Division Scheme using Sensitivity Analysis

        A dynamic zone division scheme is presented in this paper based on sensitivity analysis of power system. The concept of this scheme is to divide a large power system into small potential areas, which enclose strong correlated buses and have weak connections with each other. These areas can be considered as different individual zones in system analysis. In N-1 contingency analysis, the computation burden is largely reduced by operating in the level of potential areas, and the system cross-section distribution is determined dynamically. Analysis result can be used as an aid of power system operation and control. In this paper, IEEE-118 bus system is chosen as testing system, and this scheme is proved to be feasible and effective for large scale power system, it will be used to build the functional module of next generation EMS in Guangdong power grid and will serve as a function in power grid operation in the future.

        Cloning and Sequence Analysis of Glyceraldehyde-3-Phosphate Dehydrogenase Gene in Yak

        In order to study the biological function of gapdh gene in yak, and prove whether the gapdh gene was a useful intra-reference gene that can be given an important role in molecular biology research of yak, the cDNA sequence encoding glyceraldehyde-3-phosphate dehydrogenase from yak was cloned by the RT-PCR method using gene specific PCR primers. The sequence results indicated that the cloned cDNA fragment (1,008 bp) contained a 1,002 bp open reading frame, encoding 333 amino acids (AAs) with a molecular mass of 35.753 kDa. The deduced amino acids sequence showed a high level of sequence identity to Bos Taurus (99.70%), Xenopus laevis (94.29%), Homo sapiens (97.01%), Mus musculus (97.90%) and Sus scrofa (98.20%). The expression of yak's gapdh gene in heart, spleen, kidney and brain tissues was also detected; the results showed that the gapdh gene was expressed in all these tissues. Further analysis of yak GAPDH amino acid sequence implied that it contained a complete glyceraldehyde-3-phosphate dehydrogenase active site (ASCTTNCL) which ranged from 148 to 155 amino acid residues. It also contained two conserved domains, a NAD binding domain in its N-terminal and a complete catalytic domain of sugar transport in its C-terminal. The phylogenetic analysis showed that yak and Bos taurus were the closest species. The prediction of secondary structures indicated that GAPDH of yak had a similar secondary structure to other isolated GAPDH. The results of this study suggested that the gapdh gene of yak was similar to other species and could be used as the intra-reference to analyze the expression of other genes in yak.
