      • 조혈모세포로 형질도입에 있어서 Vesicular Stomatitis G Envelope의 효과

        목적 : 조혈모세포에서 viral vector를 이용한 안정된 유전자 전달은 유전성 혈액질환의 유전자 치료의 실현에 우선되어야 한다. 그러나 실제로 조혈모세포의 생물학적 특성 등으로 인해 유전자 도입율이 현저히 낮아 임상적인 적용에는 제한이 있다. 저자들은 조혈모세포로 VSV-G 외막을 사용한 HIV vector의 형질도입 효과를 알아보고자 하였다. 방법 : VSV-G를 이용한 HIV-1 vector를 calcium phosphate coprecipitation 방법으로 293T 세포에 형질도입시켜 재조합 농축 바이러스를 얻었다. 분리한 CD34+세포에 재조합 농축 바이러스(10배 농축)를 넣은 다음 4㎍/㎖ polybrene(Sigma)를 첨가하였다. 각 well당 RPMI로 총 500㎕로 맞춘 다음 37℃, 5% CO_2 환경에서 배양하면서 24시간, 48시간 후에 형질도입의 효과를 유세포기와 면역형광 현미경으로 분석하였다. 결과 : HIV 재조합 바이러스가 형질도입된 CD34+ 세포는 YFP의 발현으로 초록색의 형광을 나타내었다. 유세포 분석상 CD34+ 세포로 형질도입율은 배양 24시간 후에는 4.7±2.4%였고, 48시간 후에는 5.7±2.7%였다. VSVG를 사용하지 않은 경우에서는 0.23±0.33%로 저조하였다. 결론 : VSV-G 외막에 의한 HIV vector를 사용하여 CD34+ 세포로 형질도입이 효과적임을 증명하였다. 이런 결과는 조혈모세포를 이용한 유전자 치료의 확대에 기여할 수 있을 것이다. Objectives : Stable gene transfer to human pluripotent hematopoietic stem cells is an attractive strategy for the curative treatment of many genetic hematologic disorders. In clinical tral, the level of gene transfer to this cell population have generally been low. In this study we have evaluated the efficiency of gene transfer to human umbilical cord blood(UCB) CD34+cells using vesicular stomatitis virus glycoprotein G(VSV-G) pseudotyped HIV-1 vector. Method : High titers of replication-defective VSV-G pseudotyped HIV-1 based vector encoding the enhanced yellow fluorescent protein were produced by transient transfection. Human CD34+cells purified from UCB were incubated with pseudotyped HIV supernatants for 24-48 hours. The transduction efficiency were measured by marker gene expression under the microscopy and flow cytometry. Results : Transduction rates into CD34+ were low at 0 and 24 hours, reflecting 4.7±2.4% at 24 hours, they were increased to 5.7±2.7% at 48 hours. Conclusion : We demonstrate efficient transduction of purified human UCB CD34+ by HIV vectors pseudotyped with VSV-G. The results extend the lentiviral vector to clinical gene therapy using human pluripotent hematopoietic stem cells.

        본 연구의 목적은 남성들의 의류 중에서 첫째, 정장용 맞춤복, 시스템오더, 기성복에 대한 구매 행동을 의복관심도, 위험지각, 의복만족도 요인을 측정하고 둘째, 의복관심도 수준에 따른 위험지각과 의복 만족도 차이를 알아보고자 한다. 본 연구의 조사방법은 20대-50대 남성을 대상으로 자기기입식 설문지를 사용하여 실시하였다. 수집한 자료는 SPSS Package Program을 사용하여 분석하였으며, 자료분석은 빈도와 백분율, 평균 및 표준편차를 이용하여 분석한 후 요인분석, ANOVA, Duncan's test를 이용하였다. 본 연구의 요약 및 결과는 다음과 같다. 첫째, 의복관심도 요인분석으로는 유행성 차원 요인, 즐거움 차원 요인, 신분상징성 차원 요인으로 분석되었고, 위험지각 요인으로는 사회적 위험 요인, 유행 · 심리적 위험 요인, 의복관리 위험 요인, 편의성 위험 요인, 경제성 위험 요인으로 분석되었으며, 의복만족도 요인으로는 사회적 만족도 요인, 유행 · 심리적 만족 요인, 품질 만족 요인, 경제적 만족 요인으로 구분되었다. 둘째, 의복관심도가 높은 집단이 기성복과 맞춤복 집단에서는 모든 위험지각 요인이 높게 나타났고, 시스템오더 집단에서는 위험지각 요인 중 유행 · 심리적 위험요인, 의복 관리위험 요인과 경제성 위험 요인에서 높게 나타났다. 셋째, 기성복 집단에서는 의복관심도 수준이 높은 집단이 모든 만족도 요인이 높았으며, 시스템 오더 집단과 맞춤복 집단에서는 유행/심리요인에서만 높게 나타났다. The purpose of the study was to investigate differences in risk recognition and clothing satisfaction of consumers of ready-made suits, system oder suits and custom made suits according to the level of their clothing interest. As an instrument for the survey, the questionnaire was developed by the researchers and distributed to male consumers aged from 20 to 50. To analyze the data, factor analysis, ANOVA, and Duncan's test were used. As the result, first, in all consumer groups, there were significant differences in risk recognition and satisfaction according to level of their clothing interests. Second, the ready-made suit consumers and the custom-made suit consumers who had higher clothing interest had higher recognition in all the factors of risk. But the system order suit consumers who had higher clothing interest had higher recognition in fashion and psychological risk, maintenance risk and economical risk. Third, the ready-made suit consumers who had higher clothing interest were satisfied more in all the satisfaction factors. But he system order suit consumers who had higher clothing interest were more satisfied in fashion and psychological factor and social factor, and the custom-made suit consumers who had higher clothing interest were more satisfied only in fashion and psychological factor.

        20세기 패션 디자인의 조형성 표현방법 연구 : 오브제 사용기법을 중심으로 Focused on the Using Techniques of Object

        Objet, which showed up with the art of 20C, is now an important element giving a creative idea to fashion designers in modern times. The purpose of the study is to review how the objet technique was paid attention and recreated in the fashion, through the analysis of works, and the formative features of each technique for fashion, in order to identify the connection of arts and fashion, and the position of fashion as art. The techniques using object appeared in the 20th century fashion are as follows: First, papier-colle, which is adding printed materials onto the surface, is such a technique that adds cut-feeling materials to impose a new texture, or arrange again the cloth-cuts to create a different clothing from the existing one, which went to the extension of materials in the fashion, Second, collage of daily materials expresses directly and emotionally through direct presentation of the objets. Especially, collage of patch-work is reproduced into a new fabric depending on the objet used, giving a standing over the form. Third, ready-made which presents the material meaning only of the objet expands the range of objets which could be used in the fashion by introducing the daily materials having a meaning itself as a fashion. Forth, an attempt to approach to the objets of popular image by designed techniques come out in modern fashion as a graffiti look or a typography look, making the clothing itself an objet to transmit a message directly to the masses. Introduction of various objets and development of expression technique brought out the diversification of materials, and enrichment and extension of expression sphere, which resultingly spreaded the freedom of expression and progressed into the art sphere, making a direct motif to solidify its standing as a formative art.

      • 미세절편체에 의한 절화류의 기내증식

        Experiments were conducted to find out the optimum cultural conditions for micro-cross sectioned leaf segments of Gypsuphila paniculata L. Gerbera hybrida and Dianthus caryophyllus. Survival percentages of micro-cross sectioned segments were the highest in gypsophila, followed by carnation. Gerbera showed on survived segments. In gypsophila, segments thicker than 400㎛ were required to obtain over 75% survival. The percentage of adventitious bud formation was much higher with 600㎛ segment as compared with 400㎛. The regeneration of micro-cross sectioned segments of gypsophila was not sensitively affected by the auxins at varying concentrations. The elongation of adventitious shoot was promoted by kinetin compared with BA. In segments of 600㎛ thickness, adventitious shoot formation was promoted by BA compared with kinetin Adventitious shoot formation or shoot growth was not affected by KNO_3 concentrations with the segments of 400㎛ thickness. But adventitious shoot elongation was promoted significantly with segments in 600㎛ thickness. KH_2PO_4 (170 mg/1, standard concentration of MS medium) was effective for adventitious shoot formation. MgSO_4 also promoted adventitious shoot formation myo-Inositol tended to be suppressed regeneration of gypsophila.

      • 小兒 驚風의 鍼灸治療에 對한 文獻的 考察

        Infantile Convulsion, one of common emergency symptoms in pediatrics, arises from sudden derangement of the central nerve system, and can cause a sudden loss of consciousness and spasm. It falls into three categories; Acute Infantile cnvulsion, Chronic Infantile convulsion and Chronic Spleen convulsion. According to research, approximately 6~7% of all babies undergo spasm more than once. Since the treatment must be done immediately, acupuncture & moxibustion treatment can be one of the most important treatments in this particular case. Therefore, the focus of this study is on how acupuncuture & moxibustion can be utilized in the treatment of Infantile Convulsion, and the literary findings are as follows: 1. The meridian points used on acute infantile convulsion are Sugu(GV26), T´aech´ung(Liv3), Hapkok(LI4). 2. The meridians used on acute infantile convulsion are Governor Vessel(GV), Bladder Meridian(BL), Stomach Meridian(ST). 3. The meridian points used on accompanied symptoms with acute infantile convulsion are Haenggan(Liv2), Yangnungch´on(Liv3) on spasm, Paek´oe(GV14) on opisthotonus, Kokchi(LI11), Taech´u(GV14) on fever, Nogung(P8), Yongch´on(K1) on fainting spell, Chok-samri(S36) on body weakness. 4. The meridian points used on chronic infanitle convulsion are Shinguol(CV8), Ch´onchj´u(S25), T´aech´ung(Liv3), Kwanwon(CV4), Ch´ukt´aek(L5). 5. The meridians used on chronic infantile convulsion are Conception Vessel(CV), Governor Vessel(GV), Stomach Meridian(ST). 6. The meridian points used on accompanied symptoms with chronic infantile convulsion are Ch´onchj´u(S25), Kolli(CV11) on diarrhea, Taenung(P7), Shinmun(H7) on fainting spell, Kansu(B18), T´aech´ung(Liv3) on spasm. 7. The meridian points and meridians are Paek´oe(GV14), Sangsung(GV23), Sugu(GV26) of Governor Vessel(GV) and Chonjung(CV16), Shinguol(CV16) of Conception Vessel(CV) and Taedon(Liv1), Changmun(Liv13).

        The characteristics of invertase enzyme immobilized in Ca Alginate were investigated. and several effects on the hydrolysis of sucrose were verified in the fluidized bed reactor. In a batch experiments, optimum values of pH and temperature for immobilized invertase were pH 4.5 and 25℃ respectively. and sucrose concentration showed substrate inhibition over 0.20mol/L. In a fluidized experiments. the amount of invert sugar was increased with the increase of sucrose concentration and the decrease of volumetric flow rate. In this system, kinetic model was satisfied as follow. PS?-Km In(I-P)=?=? where. R(operating parameter)=0.3601 mol/h.
