        Debrisoquine이 개의 신장기능에 미치는 영향

        This study was attempted to investigate the action of debrisoquine, a sympathetic blocking agent presently employed in treating hypertension, on renal function and to elucidate the mechanism of its action. Debrisoquine, given intravenously, elicited increased urine flow, osmolar and free water clearances, along with marked increases in excretion of both sodium and potassium. Glomerular filtration rate also increased, but renal plasma flow tended to decrease, so that the filtration fraction tended to increase. Rates of reabsorption of sodium and potassium in renal tubules were also significantly diminished. The diuresis induced by debrisoquine was completely blocked by treatment with phentolamine and reserpine, and also markedly inhibited by acute renal denervation. Debrisoquine, when injected directly into a renal artery, produced antidiuretic effect and a reduction in urinary excretion of sodium and potassium, along with diminished renal plasma flow and increased filtration fraction. The above observations indicate that debrisoquine, when given intravenously, induces diuresis in the dog as a result of both diminished tubular reabsorption of electrolytes and of renal hemodynamic changes, which seem to be related to its inhibitory action of catecholamine-release from the sympathetic nerve endings.

      • Influence of Intraventricular Taurine on the Cardiovascular System of the Rabbit

        Taurine은 유황 함유 아미노산으로서 뇌내에서 억제작용을 갖는 신경전달물질 역할을 하는것으로 알려져 있다. Taurine을 가토의 측뇌실내에 투여하여 혈압 및 심박에 미치는 영향을 검토하고 그 작용 기전을 규명코자 본 연구를 시행하여 얻은 연구결과는 다음과 같다. Taurine $(0.15{\sim}1.5\;mg)$은 urethane 마취 가토의 측뇌실내로 주사하였을때 혈압강하작용 및 심박감소작용이 현저하고 지속적인 용량반응 곡선을 나타내었다. 이와 동시에 상당한 호흡억제 작용도 관찰되었다. 그러나 동일량의 taurine을 정맥내에 투여시 혈압 및 심박에 아무런 영향을 미치지 못하였다. Taurine의 혈압 하강작용은 chlorisondamine, clonidine, strychnine, bicuculline등으로 전처치시 유의하게 억제되었으나 atropine, 양측 미주신경절단, propranolol 및 metoclopramide등의 전처치로 영향을 받지 않았다. 더우기 taurine은 norepinephrine의 승압 반응에도 영향을 미치지 못하였다. Taurine의 심박감소 작용은 chlorisondamine, propranolol, clonidine, strychnine 및 bicuculline등으로 전처리시 현저히 차단되었으나 atropine, 양측 미주신경절단, metoclopramide 등의 전처리로 영향을 받지 않았다. 이상의 연구한 결과로 보아, taurine은 가토의 측뇌실내 투여시 지속적이며 현저한 혈압하강 및 심박감소를 일으키며 이러한 작용은 뇌내의 catecholamine 뉴론과 관련있는 taurine (glycine) 및 gamma-aminobutyric acid (GABA) 수용체를 통해서 나타나는 것으로 사료된다. The purpose of the present study is an attempt to investigate the effect of intraventricular taurine, which is a naturally occuring amino acid containing sulfur and has inhibitory action in brain, on heart rate and blood pressure in the urethane anesthetized rabbits and also to elucidate the mechanism of its cardiovascular actions. Taurine $(0.15{\sim}1.5\;mg)$ injected into the lateral ventricle of anesthetized normontensive rabbits produced a dose-related fall in arterial blood pressure and heart rate, which were marked and long-lasting along with considerable respiratory depression. However, the intravenous administration of taurine at the same dose with intraventricular injection did not induce any changes in blood pressure as well as heart rate. Depressor responses induced by taurine were inhibited significantly by pretreatment with chlorisondamine, clonidine, strychnine and bicuculline but not by atropine, vagotomy, propranolol and metoclopramide. Moreover, taurine did not affect the pressor responses of norepinephrine. Taurine-induced bradycardic effects were blocked clearly by pretreatment with chlorisondamine, propranolol, clonidine, strychnine and bicuculline, while they were not influenced by atropine, vagotomy and metoclopramide. These experimental results suggest that intraventricular taurine causes long-lasting hypotensive and bradycardic actions, and that these cardiovascular effects may be exerted through taurinergic (glycinergic) and GABAergic receptors which are associated with catecholaminergic neurons in brain.

      • Influence of Histaminergic Receptor Activation on Catecholamine Secretion in The Perfused Rat Adrenal Gland

        흰쥐 관류부신에서 histamine의 catecholamine (CA) 분비작용의 특성과 기전을 규명코자 연구한 결과는 다음과 같다. Histamine $(37.5{\sim}150\;{\mu}g)$을 부신정맥내에 주사 하였을 때 현저한 용량 의존성의 CA 분비작용을 나타내었다. 그러나 histamine $(150\;{\mu}g)$을 120분 간격으로 반복 투여시 제 3차 투여시부터는 CA 분비효과가 뚜렷이 감소하였다. 즉, histamine의 반복투여로 인한 반응급강현상을 관찰할 수 있었다. Histamine의 CA 분비작용은 chlorisondamine, diphenhydrarmine, ranitidine, $Ca^{++}-free$ Krebs 용액의 관류, nicardipine 및 TMB-8 등의 전처치로 유의하게 억제 되었으나 pirenzepine의 전처치에 의해서는 별다른 영향을 받지 않았다. 더우기 histamine $(6.8{\times}10^{-5}M)$으로 30분간 관류시킨 후에 ACh $(50{\mu}g)$의 CA 분비작용이 상당히 억제됨을 나타내었다. 이상과 같은 연구 결과로 보아 histamine은 흰쥐 적출관류 부신에서 현저한 CA 분비작용을 나타내었으며 칼슘 의존성이었다. 이러한 CA 분비작용은 $H_1-$ 및 $H_2-$ 양수용체의 활성화를 통해서 일어나며 또한 부신의 nicotine 수용체와도 관련성이 있는 것으로 사료된다. The present study was conducted to examine the characteristics of histamine on catecholamine secretion in the isolated perfused rat adrenal gland and to clarify the mechanism of its secretory action. Histamine (37.5 to 150 ug) injected into an adrenal vein evoked a dose-dependent significant secretory response of catecholamines (CA) from the rat adrenal gland. However, upon the repeated injection of histamine (150 ug) at 120 min intervals, CA secretion was rapidly decreased after third injection of histamine. Tachyphylaxis to releasing effects of CA evoked by histamine was observed by the repeated administration. The histamine-induced CA secretion was markedly inhibited by the pretreatment with chlorisondamine, diphenhydramine, ranitidine, $Ca^{++}-free$ Krebs solution, nicardipine and TMB-8 while was not affected by pirenzepine. Moreover, the CA secretion evoked by ACh was considerably reduced by the prior perfusion of histamine $(6.8{\times}10^{-5} M)$ for 30 min. These experimental data suggest that histamine causes secretion of CA in a calcium dependent manner from the perfused rat adrenal gland and that its secretory effect is mediated through activation of both $H_1-$ and $H_2-histaminergic$ receptors located in adrenal medulla, which may be associated with stimulation of cholinergic nicotinic receptors.

      • Forskolin-Induced Potentiation of Catecholamine Secretion Evoked By Ach, DMPP, McN-A-343 and Excess $K^+$ From the Rat Adrenal Gland

        Adenylate cyclase 효소를 활성화시키는 약물인 Forskolin의 흰쥐 적출관류 부신으로부터 Ach, excess $K^+$, McN-A-343 및 caffein에 의한 catecholamines (CA) 분비작용에 대한 영향을 검색하고, 그 기전을 규명코자 연구를 시행하여 다음과 같은 연구결과를 얻었다. Forskolin (1.0 uM)은 흰쥐 부신적출정맥내로 1분동안 관류시킨 후 Ach(50 ug), excess $K^+$(56 mM), DMPP (100 uM) 및 caffeine (0.3 mM)에 의한 CA 분비작용을 현저히 증강시켰으나 McN-A-343에 의한 CA분비작용에는 영향을 미치지 않았다. Forskolin 자체는 CA분비작용을 일으키지 못하였다. 또한 세포의 calcium을 제거한 상태에서도 위 약물에 의한 CA분비작용에 대하여 유의한 증강작용을 나타내었다. 그러나 McN-A-343의 CA작용에는 영향이 없었으나 위의 약물의 CA분비작용을 유의하게 강화시켰다. Cyclic AMP를 증가시키는 약물로 알려져 있는 dibutyryl cyclic AMP (DBcAMP)는 300 uM농도를 1분간 관류시 Ach, excess $K^+$ 및 DMPP의 CA 분비작용을 뚜렷하게 증강시켰으나 McN-A-343 및 caffeine의 CA분비에는 별다른 영향이 없었다. DBcAMP 자체도 CA분비작용에는 영향을 미치지 못하였으나 또한 DBcAMP는 세포의 칼슘제거시에도 위의 약물에 의한 CA분비작용을 의의있게 증강시켰다. 그렇지만, McN-A-343의 CA분비작용은 증강시키지 못하였다. 이상의 연구결과로 보아 Forskolin는 adenylate cyclase를 활성화 시킴으로써 cyclic AMP 농도를 증가시켜 세포내로 칼슘유입을 증강시키며, 또한 세포내의 칼슘이동에도 관여함으로써 cholinergic nicotinic stimulation 및 depolarization에 의한 CA분비작용을 상승시키는 것으로 사료되어진다. The present study was an attempt to investigate the effect of forskolin on secretion of catecholamines (CA) evoked by Ach, excess $K^+$, DMPP, McN-A-343 and caffeine from the isolated perfused rat adrenal glands and to elucidate its mechanism of action. The perfusion with forskolin (1.0 uM) for 1 min into the adrenal vein enhanced markedly the secreation of CA evoked by Ach (50 ug), excess $K^+$ (56 mM) DMPP (100 uM) and by caffeine (0.3 mM) but did not that by McN-A-343. Forskolin alone did not potentiate the CA secretion. Moreover, forskolin augmented the CA release evoked by the above same stimulation even in the absence of extracellular calcium. The 1 min perfusion of 300 uM-dibutyryl cyclic AMP (DBcAMP), which is known to increase cyclic AMP levels, led to enhancement of Ca secretion evoked by Ach, excess $K^+$ and DMPP but did not that by McN-A-343 and caffeine. DBcAMP by itself also did not augment the CA secretion. In the calcium-free medium DBcAMP significantly enhanced the CA secretion by the same stimulation, except for the case of McN-A-343. These experimental results suggest that forskolin activates adenylate cyclase, resulting the elevation of cyclic AMP which may potentiate cholinergic nicotinic receptor-mediated and also depolarization-dependent CA secretion and that it may alter the intracellular calcium homeostasis in the rat adrenal glands.

      • The Mode of Action of Pentazocine on Catecholamine Secretion from the Perfused Rat Adrenal Medulla

        Pentazocine은 opioid 수용체에 대한 흥분작용과 길항작용을 겸유한 opioid계 약물로 알려져 있다. 본 연구에서 흰쥐 적출 관류부신으로 부터 pentazocine의 catecholamine(CA) 분비작용을 관찰하여 그 기전을 규명하고 또한 다른 opioid의 작용과 비교하여 얻어진 결과는 다음과 같다. Pentazocine$(30{\sim}300\;ug)$을 부신정맥내에 주사하였을때 현저한 용량의 존성의 CA 분비작용을 나타내었다. Pentazocine의 이러한 CA 분비작용은 chlorisondamine $(10^{-6}\;M)$, naloxone $(1.22{\times}10^{-7}\;M)$, morphine $(1.73{\times}10^{-5}\;M)$, enkephalin $(9.68{\times}10^{-6}\;M)$, nicardipine $(10^{-6}\;M)$ 및 TMB-8$(10^{-5}\;M)$등의 전처치로 뚜렷이 억제되었으나 pirenzepine (2과$10^{-6}\;M)$의 전처치에 의해서는 영향을 받지 않았다. $Ca^{++}$-free Krebs 용액으로 30분간 관류한 후에 pentazocine의 CA 분비작용은 현저한 감소를 나타냈었다. Pentazocine $(1.75{\times}10^{-4}\;M)$을 20분간 관류시킨 후에 ACh $(5.32{\times}10^{-3}\;M)$과 DMPP $(10^{-4}\;M)$에 의한 CA 분비작용이 의의 있게 감약되었다. 이상과 같은 연구결과를 종합하면, pentazocine은 횐쥐 적출 관류부신에 투여시 현저한 CA 분비작용을 일으키고 있는 칼슘의존성 exocytotic mechanism에 의한 것으로 생각되며, 이러한 pentazocine의 CA 분비작용은 부신 chromaffin cell에 있는 opioid 수용체의 활성화를 통하여 나타나며, 또한 부신의 nicotine 수용체의 흥분작용과도 관련성이 있는 것으로 사료된다. The present study was attempted to investigate whether pentazocine, which is known to possess both opioid agonistic and antagonistic properties, produces catecholamines (CA) secretion from the isolated perfused rat adrenal gland, and to establish the mechanism of its action, and also to compare its action with that of some opioids. Pentazocine (30 to 300 ug) injected into an adrenal vein caused a dose-dependent secretory response of CA from the rat adrenal medulla. The pentazocine-evoked secretion of CA was remarkably diminished by the preloading with chlorisondamine $(10^{-6}\;M)$, naloxone $(1.22{\times}10^{-7}\;M)$, morphine $(1.7{\times}10^{-5}\;M)$, met-enkephalin $(9.68{\times}10^{-6}\;M)$, nicardipine $(10^{-6}\;M)$ and TMB-8 $(10^{-5}\;M)$ while was not influenced by the pretreatment of pirenzepine $(2{\times}10^{-6}\;M)$. The perfusion of $Ca^{++}$-free Krebs solution for 30 min into the gland also led to the marked reduction in CA secretion evoked by pentazocine. Furthermore, the CA release evoked by ACh and/or DMPP was greatly inhibited by the pretreatment with pentazocine $(1.75{\times}10^{-4}\;M)$ for 20 min. From these experimental results, it is thought that pentazocine causes markedly the increased secretion of CA from the isolated perfused rat adrenal medulla by a calcium-dependent exocytotic mechanism. The secretory effect of pentazocine appears to be mediated through activation of opioid receptors located on adrenal chromaffin cells, which may be also associated with stimulation of cholinergic nicotinic receptors.

      • Metoclopramide가 휜쥐의 血壓에 미치는 影響

        Influences of metocopramide (MCP), which is a seletive dopaminergic antagonist, on blood pressure of the rat and its mechanism of action were investigated in the present study. MCP administered into a femoral vein of the rat caused markedly a dose-related fall in blood pressure followed by secondary transient pressor response. The depressor action evoked by MCP was not blocked by pretreatment of atropine or chlorisondamine, while the pressor action was inhibited significantly. Prazosin treatment reduced markedly both of depressor and pressor induced by MCP. MCP-evoked pressor action was not affected by clonidine, but the depressor response was attenuated significantly. Debrisoquin treatment made the inhibited response to MCP-induced pressor response, while did not affect the depressor action. Both of pressor and depressor responses evoked by MCP were not influenced by cyproheptadine. The hypertensive activity induced by nor epinephrine was weakened markedly by the infusion of MCP (1.50 mg/kg/30min.), but the pressor action evoked by dopamine not affected. These experimental data suggest that MCP produces biphasic responses (depressor and pressor) in blood pressure of the rat, and that the hypotensive activity is due to adrenergic alpha-receptors blockade, and that pressor activity is exerted through stimulation of cholinergic nicotinic receptors in autonmic ganglia.
