<P>The human immunodeficiency virus type 1 (HIV-1) Tat protein transduction domain (PTD) is responsible for highly efficient protein transduction across plasma membranes. In a previous study, we showed that Tat-Cu,Zn-superoxide dismutase (Tat-SOD) can be directly transduced into mammalian cells across the lipid membrane barrier. In this study, we fused the human SOD gene with a Tat PTD transduction vector at its N- and/or C-terminus. The fusion proteins (Tat-SOD, SOD-Tat, Tat-SOD-Tat) were purified from Escherichia coli and their ability to enter cells in vitro and in vivo compared by Western blotting and immunohistochemistry. The transduction efficiencies and biological activities of the SOD fusion protein with the Tat PTD at either terminus were equivalent and lower than the fusion protein with the Tat PTD at both termini. The availability of a more efficient SOD fusion protein provides a powerful vehicle for therapy in human diseases related to this anti-oxidant enzyme and to reactive oxygen species.</P>

<P>Carbon coils could be synthesized using C2H2/H2 as source gases and SF6 as an incorporated additive gas under thermal chemical vapor deposition system. Nickel catalyst layer deposition and then hydrogen plasma pretreatment were performed prior to the carbon coils deposition reaction. According to the different reaction processes, the injection time of SF6 gas flow was varied. The characteristics (formation density, morphology, and geometry) of the deposited carbon coils on the substrates were investigated according to the different reaction processes. Finally, the large-scale synthesis of carbon coils and their geometry control could be achieved merely by manipulating SF6 gas flow injection time. Three cases growth aspects were proposed according to SF6 gas flow injection time in association with the fluorine's characteristics for etching the materials or enhancing the nucleation sites.</P>

Substituted imidazolidin-2-ones deduced as potential inhibitors of IleRS by docking simulations were synthesized from an aziridine-2-carboxaldehyde. Reductive amination of an aziridine-2-carboxaldehyde with dipeptides for the substituents at N1 and followed by aziridine-ring expansion with triphosgene afforded 4-chloromethylimidazolidin-2-ones whose chloride were further manipulated towards phenylurea, pyrimidin-2-yl-urea or benzenesulfonamide at C4.

Ceruloplasmin (CP) is the major plasma antioxidant and copper transport protein. Monoclonal antibodies (mAbs) against human CP were produced and characterized. A total of five hybridoma cell lines were established (CP2, CP10, CP20, CP25, CP30). From the epitope mapping analysis, two subgroups of mAbs recognize different peptide fragments were identified. When the purified CP was incubated with the mAbs, the ferroxidase activity of CP was inhibited up to a maximum 57%. Immunoblotting with various tissue homogenates indicated that all the mAbs specifically recognize a single protein band of 130 kDa. They also appear to be extensively cross-reactive among different mammalian including human and avian sources. These results demonstrated that only one type of immunologically similar CP is present in all of the mammalian tissues including human. The CP mAbs could be of great benefit to design the diagnostic kit for CP-related diseases such as Wilson's disease.

Family meeting program(FMP) is to share information and discuss treatment between the patient, family members and health care professionals. In family centered society of Korea, when a patient is diagnosed as cancer, it is difficult to disclose the diagnosis directly to the patient. In this social setting, the primary purpose of our FMP focused on delivering the medical information to the patient himself or herself. The aim of this study is to improve the quality of FMP by analyzing the records of FMP. Patients who first admitted to oncology department for stage IV lung cancer before treatment and their family members were invited to FMPs. The house staff resident physician provided the current medical status of the patients, including diagnosis and stage of the cancer. The attending oncologist provided further treatment plan and prognosis. Data characterizing the meetings were collected by oncology specialized nurse using a data collection sheet. A total of 136 FMPs were conducted during an 18-month period. Prior to FMPs, family member and patients were aware of their diagnosis in 89.7% and 76.5% respectively. To further acquire information of their current medical status, patients actively participated in 102 of 136 meetings (75%). 13 patients refused the attention to the meeting. The family members of 18 patients concerned about the distress of the patients, therefore, they refused to invite the patients to the meetings. 3 patients were so poor to attend FMPs. Diagnosis, stage and primary goal of the treatment were informed in detail. However, the prognosis and life expectancy was discussed only in 93 meetings (68.4%). All patients and family members satisfied about the FMPs. 98 patients (95.1%) showed good comprehension during the meeting, and in 123 cases (90.4%), family members comprehended well. Although most of the patients participated the FMPs, a few patients did not join the meeting because their family members concerned about the distress of the patients. However, the patients participated the meeting actively wanted to know about their disease status, treatment plan, prognosis and life expectancy. The FMPs advanced the comprehension of the disease for the patients and their family members.

본 연구의 목적은 PS-2 (N141I) 알츠하이머 형질전환 모델 생쥐를 대상으로 트레드밀 운동이 뇌의 세포질과 미토콘드리아의 Aβ-42, cytochrome c, SOD-1, 2 and Sirt-3 단백질 발현에 미치는 효과를 알아보는데 있다. 우선 알츠하이머 형질전환 생쥐를 Non-Tg-sedentary (n=5), Non-Tg-treadmill exercise (n=5) 집단과 Tg-sedentary (n=5), Tg-treadmill exercise (n=5) 집단으로 구분하고 트레드밀 운동을 통한 신경보호 효과를 검증하기 위해 Tg와 Non-Tg집단에 12주간 트레드밀 운동을 수행한 후 인지능력을 살펴보고 뇌의 세포질과 미토콘드리아의 Aβ-42, cytochrome c, anti-oxidant enzymes (SOD-1, SOD-2)와 Sirt-3 단백질을 분석하였다. 먼저 트레드밀운동은 Tg 집단에서 인지능력의 개선을 나타냈으며 미토콘드리아의 Aβ-42와 세포질의 cytochrome c 단백질의 감소와 항산화 효소인 SOD-1, SOD-2를 유의하게 증가시켰다. 게다가 트레드밀 운동은 모든 집단에서 Sirt-3 단백질의 발현을 증가시켰다. 따라서 트레드밀 운동은 인지능력의 향상과 세포 내 스트레스를 유발하는 Aβ-42를 억제시켜 알츠하이머 질환을 개선시킬 수 있는 효과적인 방법이라고 생각된다. The purpose of this study was to investigate the effects of treadmill exercise on Aβ-42, cytochrome c, SOD-1, 2 and Sirt-3 protein expressions in brain cytosol and mitochondria in mutant (N141I) presenilin-2 transgenic mice with Alzheimer's disease (AD). The mice were divided into four groups (Non-Tg-sedentary, n=5; Non-Tg treadmill exercise, n=5; Tg-sedentary, n=5; Tg treadmill exercise, n=5). To evaluate the neuroprotective effect of treadmill exercise, Non-Tg and Tg mice were subjected to exercise training on a treadmill for 12 wk, after which their brain cytosol and mitochondria were evaluated to determine whether any changes in the cognitive performance, Aβ-42 protein, cytochrome c protein, anti-oxidant enzymes (SOD-1, SOD-2) and Sirt-3 protein had occurred. The results indicated that treadmill exercise resulted in amelioration in cognitive deficits of Tg mice. In addition, the expressions of mitochondrial Aβ-42 and cytosolic cytochrome c protein were decreased in the brains of Tg mice after treadmill exercise, whereas antioxidant enzymes, SOD-1 and SOD-2 were significantly increased in response to treadmill exercise. Furthermore, treadmill exercise significantly increased the expression of Sirt-3 protein in Non-Tg and Tg mice. Taken together, these results suggest that treadmill exercise is a simple behavioral intervention which can sufficiently improve cognitive performance and inhibit Aβ-induced oxidative stress in AD.
