Antifungal Activities Against Plasmodiophora brassicae Causing Club Root

Club root is one of the major diseases that occur in crucifers. It id caused by Plasmodiophora brassicae. In order to discover microbial biopesticides against P. brassicae, forty-eight Streptomyces isolated from soil were screened. Among these, three strains showed excellent pesticidal activities. We report result on in vivo screening with fermentation broths of these strains and identification of the strain taxa.

Screening tests against fungus causing rice blast, Magnaporthe grisea, were performed in order to develop biopesticides. More than 400 actinomycetes collected at several sites near Hanla Mountain on Jeju Island, Korea were tested, and strain BG2-53 showed potent antifungal activity. The in vivo screening was performed with fermentation broth, and the strain taxon was identified.

Antifungal activities of 6-[(N-4-bromophenyl)amino]-7-cholor-5,8-quinolinedione (RCK7) were tested. The MIC values of RCK7 were determined for antifungal suceptibility, in vitro against Aspergillus niger, Cryptococcus neoformans and Trichophyton mentagrophyte by standard agar streak method. In vitro, RCK7 showed more potent antifungal activity than fluconazole and ketoconazole. Also, RCK7 was tested for in vivo antifungal activity in the treatment of systemic infection with Candida albicans in normal mice. The therapeutic potential of RCK7 had been assessed by evaluating their survival rate against systemic infections compared with that of ketoconazole. ED_50 of intraperitoneally administered RCK7 was 2.05±0.30 ㎎/㎏, but that of ketoconazole was 8.00±0.73 ㎎/㎏, respectively. When RCK7 was adminstered in travenously at the ED_50(2.05 ㎎/㎏), the colony of Candida albicans in the liver after 7 days and 14 days were reduced as likely as ketoconazole at the ED_50 (8.00 ㎎/㎏), and the better survival rates than ketoconazole's were achieved after 14 days. The results suggest that RCK7 may be a potent antifungal agent.

Antifungal Activity of Streptomyces sp. Against Puccinia recondite Causing Wheat Leaf Rust

To discover a potent strain against wheat leaf rust, soil samples collected from Ilgamho, Seoul, Korea were tested in vivo and a strain belonging to Streptomyces sp. was found to show good antifungal activity when fermented in a broth. The identification of the strain was carried out based on 16s rDNA analysis, and the active compound was separated from the fermented broth. Even though its structure was not determined completely, the authors report the results obtained so far indicate that the fermented broth of the strain showed activity against wheat leaf rust. Therefore, we propose that this may be a potential novel strain showing antifangal activity against Puccinia reconditu.

Phytochemicals have been considered as alternatives for synthetic fungicides because of their biodegradability and low toxicity. In this study, we found that the methanolic extract of Corydalis ternata suppressed the development of plant diseases caused by Puccinia triticina and Colletotrichum coccodes. As the antifungal substance, three isoquinoline alkaloids (dehydrocorydaline, stylopine, and corydaline) were isolated from C. ternata. These active compounds also exhibited in vivo antifungal activity against P. triticina and C. coccodes. Taken together, our results suggest that C. ternata and its active compounds can be used to control plant diseases.

배추 뿌리혹병에 대한 효율적인 살균 활성 검정법을 확립하기 위하여, 배추 품종, 기주 식물의 생육 시기, 접종원 농도, 토양 종류 등의 발병 조건에 따른 살균제 fluazinam, ethaboxam 및 cyazofamid의 배추 뿌리혹병에 대한 방제 효과를 온실에서 실험하였다. 배추의 생육 시기에 따른 살균제들의 방제효과는 파종 7일 및 14일 유묘에서는 21일 유묘보다 더 높은 방제효과를 보였다. 그리고 토양종류에 따른 살균제의 방제효과는 원예용 상토보다 혼합토양에서 더 높은 방제효과를 나타냈다. 무처리구의 뿌리혹병 발생은 배추가 커질수록 증가하였으며, 혼합토양보다 원예용 상토에서 병 발생이 더 많았다. 그러나 배추품종에 따른 살균제들의 방제효과 차이는 거의 없었다. 한편 P. brassicae 포자 농도가 100배 증가하여도 무처리구의 뿌리혹병 발생은 거의 차이가 없었으나, 살균제들의 배추 뿌리혹병 방제효과는 크게 감소하였다. 이상의 결과로부터 살균제 fluazinam, ethaboxam 및 cyazofamid는 토양 종류, 배추 생육시기 및 접종원 농도 등의 발병 조건에 따라 배추 뿌리혹병에 대하여 다양한 방제효과를 보였으며, 이들 중 포자 농도가 살균제의 방제효과에 가장 큰 영향을 미친다고 생각되었다. To develop the efficient screening methods for antifungal compound active to Chinese cabbage clubroot caused by Plasmodiophora brassicae, the control efficacy of three fungicides fluazinam, ethaboxam, and cyazofamid on the disease was tested under several conditions such as soil types, cultivars of Chinese cabbage,growth stages of the host, and inoculum concentrations. The in vivo antifungal activities of the fungicides on clubroot of two Chinese cabbage cultivars were hardly different. At 7- and 14-day-old seedlings, the fungicides were more effective to control of clubroot than at 21-day-old seedlings. In a commercial horticulture media soil (CNS), disease severity of untreated controls was higher and control activity of the fungicides was less than in a mixture of CNS and upland soil (1:1, v/v). Disease development of the seedlings inoculated with P. brassicae at 1.8×10^7 spores/pot to 1.1×10^9 spores/pot was almost same, but control efficacy of the fungicides was negatively correlated with inoculum dosages. To effectively select in vivo antifungal compound on Chinese cabbage clubroot, 14-day-old seedlings need to be inoculated with P. brassicae by drenching the spore suspension to give 1×10^8 spores/pot 1 day after chemical treatment. To develop clubroot,the inoculated plants are incubated in a growth chamber at 20℃ for 2 days, and then cultivated in a greenhouse (20±5℃) for four weeks.

The therapeutic potential of RCK11 had been assessed by evaluating their activities (survival rates) against systemic infections in normal mice. $ED_{50}$ of intraperitoneally administered RCK11 was $0.10\pm0.01 mg/kg$ but that of ketoconazole had $8.00\pm0.73 mg/kg$ respectively. When RCK11 was administered intravenously at the $ED_{50}$(0.10 mg/kg), the colony counts of Canoliola albicans in the liver after 7 days and 14 days were reduced as likely as ketoconazole at the $ED_{50}$ (8.00 mg/kg), and the better survival rates than ketoconazole were achieved after 14 days.
