NG108-15 세포에서 Angiotensin Ⅱ에 의한 Rac의 활성화 조절

Purpose : Differentiation of neuron ceils were induced by various stimuli such as nerve growth factor, basic fibroblast growth factor, reactive oxygen species and some hormones. This study was investigated on the activation of Rac, Rho GTPase and the neuronal differentiation by Angiotensin II (Ang Ⅱ) in NG108-15 cells. Materials and Methods : NG108-15 cells were incubated on 100 nM Ang Ⅱ for 48 hr to induce the neuronal differentiation. Differentiated cells with one or more neurite longer than the length of cell body were counted under inverted microscope. Activation of Rac was analyzed by PBD (p21 binding domain)-pulldown assay Results : Treatment of Ang Ⅱ in NG108-15 cells induced about 2-fold increase of differentiated cells compared with control cells. Also Rac was activated by 100 nM Ang II for 5-10 min. However, Rho A wasn't activated by Ang II in the same condition. Conclusion : These results suggest that Ang Ⅱ induced neurite outgrowth and Rac activation in NG108-15 cells. 연구 목적 : 신경세포의 분화는 다양한 자극에 의해 유도된다. 본 연구는 신경세포 분화 모델인 NG108-15 세포에서 Angiotensin Ⅱ(Ang Ⅱ)에 의해 분화가 유도되는가를 확인하고 그 분화 유도과정에 Rho GTPase인 Rac의 활성화가 일어나는지를 관찰하고자 하였다. 재료 및 방법 : NG108-15 세포는 100nM AngⅡ를 48 시간 처리하여 분화를 유도하였다. 분화 양상은 세포에 비해 2배 이상의 길이로 신경돌기가 확장된 세포만을 측정하였다. Rac의 활성화는 Rac-GTP에만 결합하는 PAK(p21-activated kinase)의 p21 binding domain(PBD)을 이용한 GST-PBD pulldown 방법을 실시하였다. 결과 : AngⅡ는 NG-108-15세포의 신경돌기확장을 처리하지 않은 세포에 비해 2배 증가시켰다. 또한 Ang Ⅱ 처리 후 5분과 10분에서 Rac의 활성화 되는 것을 관찰할 수 있었다. 반면에 이 시간대에 Rho A는 전혀 활성화 되지 않았다. 이는 신경세포의 분화에 AngⅡ에 의한 Rac의 활성화가 관여할 가능성을 시사하는 결과이다. 결론 : 이상의 결과는, NG-108-15 세포에서 AngⅡ는 Rac 활성화를 유도하고, 이는 신경돌기의 확장에 직접적인 영향을 줄 수 있을 것이다.
