        Optimization of silver nanoparticle synthesis by chemical reduction and evaluation of its antimicrobial and toxic activity

        Background: Chemical reduction has become an accessible and useful alternative to obtain silver nanoparticles (AgNPs). However, its toxicity capacity depends on multiple variables that generate differences in the ability to inhibit the growth of microorganisms. Thus, optimazing parameters for the synthesis of AgNPs can increase its antimicrobial capacity by improving its physical-chemical properties. Methods: In this study a Face Centered Central Composite Design (FCCCD) was carried out with four parameters: AgNO3 concentration, sodium citrate (TSC) concentration, NaBH4 concentration and the pH of the reaction with the objective of inhibit the growth of microorganisms. The response variables were the average size of AgNPs, the peak with the greatest intensity in the size distribution, the polydispersity of the nanoparticle size and the yield of the process. AgNPs obtained from the optimization were characterized physically and chemically. The antimicrobial activity of optimized AgNPs was evaluated against Staphylococcus aureus, Escherichia coli, Escherichia coli AmpC resistant, and Candida albicans and compared with AgNPs before optimization. In addition, the cytotoxicity of the optimized AgNPs was evaluated by the colorimetric assay MTT (3- (4,5- Dimethylthiazol- 2- yl)- 2, 5 - Diphenyltetrazolium Bromide). Results: It was found that the four factors studied were significant for the response variables, and a significant model (p <0.05) was obtained for each variable. The optimal conditions were 8 for pH and 0.01 M, 0.0 6M, 0.01 M for the concentration of TSC, AgNO3, and NaBH4, respectively. Optimized AgNPs spherical and hemispherical were obtained, and 67.66% of it had a diameter less than 10.30 nm. A minimum bactericidal concentration (MBC) and minimum fungicidal Concentration (MFC) of optimized AgNPs was found against Staphylococcus aureus, Escherichia coli, Escherichia coli AmpC resistant, and Candida albicans at 19.89, 9.94, 9.94, 2.08 μg/mL, respectively. Furthermore, the lethal concentration 50 (LC50) of optimized AgNPs was found on 19.11 μg/mL and 19.60 μg/mL to Vero and NiH3T3 cells, respectively. Conclusions: It was found that the factors studied were significant for the variable responses and the optimization process used was effective to improve the antimicrobial activity of the AgNPs.

        Solar polarimetry in the K I <i>D</i><sub>2</sub> line : A novel possibility for a stratospheric balloon

        <P>Of the two solar lines, K I<I>D</I>1 and <I>D</I>2, almost all attention so far has been devoted to the <I>D</I>1 line, as <I>D</I>2 is severely affected by an O2 atmospheric band. This, however, makes the latter appealing for balloon and space observations from above (most of) the Earth’s atmosphere. We estimate the residual effect of the O2 band on the K I<I>D</I>2 line at altitudes typical for stratospheric balloons. Our aim is to study the feasibility of observing the 770 nm window. Specifically, this paper serves as a preparation for the third flight of the Sunrise balloon-borne observatory. The results indicate that the absorption by O2 is still present, albeit much weaker, at the expected balloon altitude. We applied the obtained O2 transmittance to K I<I>D</I>2 synthetic polarimetric spectra and found that in the absence of line-of-sight motions, the residual O2 has a negligible effect on the K I<I>D</I>2 line. On the other hand, for Doppler-shifted K I<I>D</I>2 data, the residual O2 might alter the shape of the Stokes profiles. However, the residual O2 absorption is sufficiently weak at stratospheric levels that it can be divided out if appropriate measurements are made, something that is impossible at ground level. Therefore, for the first time with Sunrise III, we will be able to perform polarimetric observations of the K I<I>D</I>2 line and, consequently, we will have improved access to the thermodynamics and magnetic properties of the upper photosphere from observations of the K I lines.</P>

        Synthesis and characterization of a silver nanoparticle-containing polymer composite with antimicrobial abilities for application in prosthetic and orthotic devices

        Background: The presence of skin problems in patients using external lower limb prosthesis is recurrent. This has generated the need to develop interfaces for prosthesis with the ability to control microbial growth. Silver nanoparticles (AgNPs) have been implemented in the development of biomaterials because of their high antimicrobial activity. This article discusses the development of an AgNP-containing polymer composite with antimicrobial activity for developing prosthetic liners. Methods: AgNPs were synthesized using a photochemical method and certain physicochemical properties were characterized. Furthermore, the antimicrobial activity of AgNPs against Staphylococcus aureus ATCC 25923 and methicillin-resistant Staphylococcus aureus (MRSA), was assessed on the basis of their minimum inhibitory concentrations (MICs). AgNPs were incorporated into a silicon elastomer to assess certain physicomechanical properties, antimicrobial activity and cytotoxic effect of the material. Results: Themaximum antimicrobial activity of the material against Staphylococcus aureus ATCC 25923 andMRSA was 41.58% ±2.97% at AgNP concentration of 32.98 μg/mL and 14.85% ±5.94% at AgNP concentration of 16.49 μg/mL, respectively. Additionally, the material exhibited tensile yield strength, rupture tensile strength, and tensile modulus of elasticity of 0.70 - 1.10 MPa, 0.71–1.06 MPa, and 0.20 - 0.30 MPa, respectively. The mechanical characteristics of the material were within the acceptable range for use in external lower limb prosthetic and orthotic interfaces. Conclusions: It was possible to incorporate the AgNPs in a silicone elastomer, finding that the composite developed presented antimicrobial activity against Staphylococcus aureus ATCC 25923 and MRSA when compared to non-AgNP material samples.

        Determination of Orbital Elements and Ephemerides using the Geocentric Laplace’s Method

        This paper presents a methodology for Initial Orbit Determination (IOD) based on a modification of the Laplace’s geocentric method. The orbital elements for Near-Earth asteroids (1864) Daedalus, 2003 GW, 2019 JA8, a Hungaria-type asteroid (4690) Strasbourg, and the asteroids of the Main Belt (1738) Oosterhoff, (2717) Tellervo, (1568) Aisleen and (2235) Vittore were calculated. Input data observations from the Minor Planet Center MPC database and Astronomical Observatory of the Technological University of Pereira (OAUTP; MPC code W63) were used. These observations cover observation arcs of less than 22 days. The orbital errors, in terms of shape and orientation for the estimated orbits of the asteroids, were calculated. The shape error was less than 53 × 10–3 AU, except for the asteroid 2019 JA8. On the other hand, errors in orientation were less than 0.1 rad, except for (4690) Strasbourg. Additionally, we estimated ephemerides for all bodies for up to two months. When compared with actual ephemerides, the errors found allowed us to conclude that these bodies can be recovered in a field of vision of 95’ × 72’ (OAUTP field). This shows that Laplace’s method, though simple, may still be useful in the IOD study, especially for observatories that initiate programs of minor bodies observation.

        Accuracy of the 2008 Simplified Criteria for the Diagnosis of Autoimmune Hepatitis in Children

        Purpose: Classical criteria for diagnosis of autoimmune hepatitis (AIH) are intended as research tool and are difficult to apply at patient’s bedside. We aimed to study the accuracy of simplified criteria and the concordance with the expert diagnosis based on the original criteria.Methods: A cohort of children under study for liver disorder was selected through consecutive sampling to obtain the prevalence of AIH within the group of differential diagnoses. AIH was defined, based on classical criteria, through committee review of medical reports. Validity indicators of the simplified criteria were obtained in an intention to diag-nose approach. Optimal cut-off and the area under the receiver operating characteristic (ROC) curve were calculated. Results: Out of 212 cases reviewed, 47.2% were AIH. For the optimal cut-off (6 points), the simplified criteria showed a sensitivity of 72.0% and a specificity of 96.4%, with a 94.7% positive and a 79.4% negative predictive value. The area under the ROC curve was 94.3%. There was a good agreement in the pre-treatment concordance between the classical and the simplified criteria (kappa index, 0.775). Conclusion: Simplified criteria provide a moderate sensitivity for the diagnosis of AIH, but may help in indicating treatment in cases under suspicion with 6 or more points.

        Chylomicron Retention Disease: a Description of a New Mutation in a Very Rare Disease

        Chylomicron retention disease, also known as Anderson’s disease, is a rare hereditary hypocholesterolemic dis-order, recessive inherited, characterized by nonspecific symptoms as abdominal distension, steatorrhea, and vomit-ing associated with failure to thrive. We describe a patient with failure to thrive, chronic diarrhea and steatorrhea who the diagnosis of chylomicron retention disease was established after several months of disease progression. The genetic study confirmed a homozygosity mutation in SAR1B gene, identifying a mutation never previous de-scribed [c.83_84delTG(p.Leu28Argfs*7)]. With this case report the authors aim to highlight for this very rare cause of failure to thrive and for the importance of an attempting diagnosis, in order to start adequate management with low fat diet supplemented with fat-soluble vitamins, reverting the state of malnutrition and avoiding possible irrever-sible and desvantating complications.

        SLAMF7 is critical for phagocytosis of haematopoietic tumour cells via Mac-1 integrin

        <P>Cancer cells elude anti-tumour immunity through multiple mechanisms, including upregulated expression of ligands for inhibitory immune checkpoint receptors(1,2). Phagocytosis by macrophages plays a critical role in cancer control(3-6). Therapeutic blockade of signal regulatory protein (SIRP)-alpha, an inhibitory receptor on macrophages, or of its ligand CD47 expressed on tumour cells, improves tumour cell elimination in vitro and in vivo(7-10), suggesting that blockade of the SIRP alpha-CD47 checkpoint could be useful in treating human cancer(11-14). However, the prophagocytic receptor(s) responsible for tumour cell phagocytosis is(are) largely unknown. Here we find that macrophages are much more efficient at phagocytosis of haematopoietic tumour cells, compared with non-haematopoietic tumour cells, in response to SIRP alpha-CD47 blockade. Using a mouse lacking the signalling lymphocytic activation molecule (SLAM) family of homotypic haematopoietic cell-specific receptors, we determined that phagocytosis of haematopoietic tumour cells during SIRP alpha-CD47 blockade was strictly dependent on SLAM family receptors in vitro and in vivo. In both mouse and human cells, this function required a single SLAM family member, SLAMF7 (also known as CRACC, CS1, CD319), expressed on macrophages and tumour cell targets. In contrast to most SLAM receptor functions(15-17), SLAMF7-mediated phagocytosis was independent of signalling lymphocyte activation molecule-associated protein (SAP) adaptors. Instead, it depended on the ability of SLAMF7 to interact with integrin Mac-1 (refs 18-20) and utilize signals involving immunoreceptor tyrosine-based activation motifs(21,22). These findings elucidate the mechanism by which macrophages engulf and destroy haematopoietic tumour cells. They also reveal a novel SAP adaptor-independent function for a SLAM receptor. Lastly, they suggest that patients with tumours expressing SLAMF7 are more likely to respond to SIRP alpha-CD47 blockade therapy.</P>
