        Metastasis associated genomic aberrations in stage II rectal cancer

        Genomic aberrations of rectal carcinoma, especially DNA copy number changes associated with metastasis were largely unclear. We aim to identify the metastasis associated biomarkers in stage II rectal cancer. Formalin-fixed, paraffin-embedded primary tumor tissues of stage II rectal carcinoma were analyzed by array-based comparative genomic hybridization, and genomic aberrations were identified by Genomic Workbench and SAM software. Copy number changes and mRNA expressions were validated by Real-time PCR in an independent rectal cancer samples. The results showed that the most frequent gains in stage II rectal cancer were at 1q21.2-q23.1, 3p21.31, 11q12.2-q23.3, 12q24.11-q24.31, 12q13.11-q14.1 and losses in 18q11.2-q23, 17q21.33-q22, 13q31.1-q31.3, 21q21.1-q21.3, 8p23.3-p23.1 and 4q22.1-q23. Twenty-two amplifications and five homozygous deletions were also identified. We further found that S100A1 (1q21.3-q23.1), MCM7 (7q22.1) and JUND (19p13.11) were amplified and overexpressed in stage II rectal cancer. Interestingly, the genomic aberrations affected 14 signaling pathways including VEGF signaling pathway and fatty acid metabolism. Most importantly, loss of 13q31.1-q34 and gain of 1q44 were associated with distant metastasis. Our results indicated that these metastasis associated genomic changes may be useful to reveal the pathogenesis of rectal cancer metastasis and identify candidate biomarkers.

        Active Fault Tolerant Control Scheme for Satellite Attitude Systems: Multiple Actuator Faults Case

        In this paper, an active fault tolerant control (FTC) design approach is proposed for the satellite attitude systems with exogenous disturbance and multiple actuator faults. Firstly, the nonlinear attitude system model of rigid satellite with multiple actuator faults is given. Next, an actuator fault diagnosis scheme, including a fault detection module and a fault estimation module, is given so as to detect the time of unknown actuator faults occurred and obtain their estimation values. Then, a terminal sliding mode-based fault tolerant attitude controller is designed using backstepping control technique, which guarantees that the closed-loop attitude systems of rigid satellite are asymptotically stable in the presence of multiple actuator faults. Numerical simulations illustrate the good performance of active FTC proposed in this study.

        Single-nucleus RNA sequencing reveals cell type-specific transcriptome alterations of Down syndrome hippocampus using the Dp16 mouse model

        Background Down syndrome (DS), the most frequently occurring human chromosomal disorder, is caused by trisomy 21. The exact molecular effects of trisomy on certain cell populations in the brain remain poorly understood. Objective The purpose of this study was to investigate the effects of trisomy on the transcriptomes of various types of neurons and nonneuronal cells in the hippocampus. Methods A total of 8993 nuclei from the WT and 6445 nuclei from the Dp16 hippocampus were analyzed by single-nucleus RNA sequencing (snRNA-seq). Cell clustering was achieved by the Seurat program. Results Hippocampal cells were grouped into multiple neuronal and nonneuronal populations. Only a limited number of trisomic genes were upregulated (q < 0.001) over 1.25-fold in a specific type of hippocampal cell. Specifically, deregulation of genes associated with synaptic signaling and organization was observed in multiple cell populations, including excitatory neurons, oligodendrocytes, and microglia. This observation suggests the potential importance of synapse deficits in DS. Interestingly, GO annotation of the upregulated genes suggested potential activation of the immune system by hippocampal excitatory neurons. Fewer trisomic genes were altered in nonneuronal cells than in neurons. Notably, microglial transcriptome analysis revealed significantly (q < 0.001) increased expression of C1qb and C1qc, which suggested potential involvement of complement-mediated synapse loss mediated by microglia in DS. Conclusion The trisomy-related hippocampal deficits should be driven by a small amount, not all, of the trisomic genes in a specific type of cell. Our work may help to narrow down both the molecular and cellular targets for future gene therapies in DS.

      • Expression of Ki67 in Papillary Thyroid Microcarcinoma and its Clinical Significance

        Purpose: To investigate the expression of Ki67 protein in papillary thyroid microcarcinoma(PTMC), and to analyze its clinical significance. Materials and Methods: Ki67 protein expression was evaluated in the tissues of 108 human PTMC and 50 other benign papillary hyperplasia of thyroid specimens using immunohistochemistry. Results: The expression intensity of Ki67 in PTMC and benign papillary hyperplasia of thyroid specimens were $1.45{\pm}1.83%$ and $0.46{\pm}0.46%$.The positive expression rates were 46.3% and 14%. There were significant differences between these two groups (p<0.01). There was no significant variation of the expression intensity and positive expression rates of Ki67 in PTMC with gender, age, position of the tumor and the level of TSH pre-operation (p>0.05), but these parameters varied with tumor size, invasion by membrane and cervical lymph node metastasis (p<0.05 or p<0.01). Conclusions: The expression of Ki67 in PTMC was related to tumor size, invasion by membrane and cervical lymph node metastasis, and could be the important indicator for judging clinical progress and estimating prognosis.

        Abietane Diterpenoids from Perovskia atriplicifolia and Their Anti-HBV Activities

        Bioassay-guided phytochemical investigation on the 90% EtOH extract of Perovskia atriplicifolia resulted in the isolation of eight abietane diterpenoids, including three new ones (1–3). Based on spectroscopic methods involving 1D and 2D NMR spectroscopy techniques, mass spectrometry, and optical rotation, the structures of the new compounds (1–3) were unambiguously characterized. Compounds 1–2 and 4–8 were evaluated for their anti-HBV (hepatitis B virus) activity in HepG 2.2.15 cell line. Results suggested rosmadial (8) had the most anti-HBV potency, suppressing the secretion of HBsAg and HBeAg, with IC50 values of 0.09 and 0.34 mM, respectively.

        Experimental study on the seismic performance of concrete filled steel tubular laced columns

        Concrete filled steel tubular (CFST) laced columns have been widely used in high rise buildings in China. Compared to solid-web columns, this type of columns has a larger cross-section with less weight. In this paper, four concrete filled steel tubular laced columns consisting of 4 main steel-concrete tubes were tested under cyclic loading. Hysteresis and failure mechanisms were studied based on the results from the lateral cyclic loading tests. The influence of each design parameter on restoring forces was investigated, including axial compression ratio, slenderness ratio, and the size of lacing tubes. The test results show that all specimens fail in compression-bending-shear and/or compression-bending mode. Overall, the hysteresis curves appear in a full bow shape, indicating that the laced columns have a good seismic performance. The bearing capacity of the columns decreases with the increasing slenderness ratio, while increases with an increasing axial compression ratio. For the columns with a smaller axial compression ratio (< 0.3), their ductility is increased. Furthermore, with the increasing slenderness ratio, the yield displacement increases, the bending failure characteristic is more obvious, and the hysteretic loops become stouter. The results obtained from the numerical analyses were compared with the experimental results. It was found that the numerical analysis results agree well with the experimental results.

        Distortional buckling calculation method of steel-concrete composite box beam in negative moment area

        'Distortional buckling&' is one of the predominant buckling types that may occur in a steel-concrete composite box beam (SCCBB) under a negative moment. The key factors, which affect the buckling modes, are the torsional and lateral restraints of the bottom plate of a SCCBB. Therefore, this article investigates the equivalent lateral and torsional restraint rigidity of the bottom plate of a SCCBB under a negative moment; the results of which show a linear coupling relationship between the applied forces and the lateral and/or torsional restraint stiffness, which are not depended on the cross-sectional properties of a SCCBB completely. The mathematical formulas for calculating the lateral and torsional restraint rigidity of the bottom plate can be used to estimate: (1) the critical distortional buckling stress of SCCBBs under a negative moment; and (2) the critical distortional moment of SCCBBs. This article develops an improved calculation method for SCCBBs on an elastic foundation, which takes into account the coupling effect between the applied forces and the lateral and/or torsional restraint rigidity of the bottom plate. This article analyzes the accuracy of the following calculation methods by using 24 examples of SCCBBs: (1) the conventional energy method; (2) the improved calculation method, as it has been derived in this article; and (3) the ANSYS finite element method. The results verify that the improved calculation method, as it has been proved in this article, is more accurate and reliable than that of the current energy method, which has been noted in the references.

        Flexural natural vibration characteristics of composite beam considering shear deformation and interface slip

        Based on Hamilton's principle, the flexural vibration differential equations and boundary conditions of the steel-concrete composite beam (SCCB) with comprehensive consideration of the influences of the shear deformation, interface slip and longitudinal inertia of motion were derived. The analytical natural frequencies of flexural vibration were compared with available results previously observed by the experiments, the results calculated by the FE model and the other similar beam theories available in the open literatures. The comparison results showed that, the calculation results of the analytical and Timoshenko models had a good agreement with the results of the experimental test and FE model. Finally, the influences of shear deformation and interface slip on the flexural natural frequencies of the SCCB were discussed. The shear deformation effect increases with the increase of the mode orders of flexural natural vibration, and the flexural natural frequencies of the higher mode orders ignoring the influence of shear deformations effect would be overestimated. The interface slip effect decrease with the increase of the mode orders of flexural natural vibration, and the influence of the interface slip effect on flexural natural frequencies of the low mode orders is significant. The influence of the degree of shear connection on shear deformation effect is insignificant, and the low order modes of flexural natural vibration are mainly composed of the rotational displacement of cross sections.

        Distortional buckling of I-steel concrete composite beams in negative moment area

        The predominant type of buckling that I-steel concrete composite beams experience in the negative moment area is distortional buckling. The key factors that affect distortional buckling are the torsional and lateral restraints by the bottom flange. This study thoroughly investigates the equivalent lateral and torsional restraint stiffnesses of the bottom flange of an I-steel concrete composite beam under negative moments. The results show a coupling effect between the applied forces and the lateral and torsional restraint stiffnesses of the bottom flange. A formula is proposed to calculate the critical buckling stress of the I-steel concrete composite beams under negative moments by considering the lateral and torsional restraint stiffnesses of the bottom flange. The proposed method is shown to better predict the critical bending moment of the I-steel composite beams. This article introduces an improved method to calculate the elastic foundation beams, which takes into account the lateral and torsional restraint stiffnesses of the bottom flange and considers the coupling effect between them. The results show a close match in results from the calculation method proposed in this paper and the ANSYS finite element method, which validates the proposed calculation method. The proposed calculation method provides a theoretical basis for further research on distortional buckling and the ultimate resistance of I-steel concrete composite beams under a variable axial force.

        Earth Pressure and Internal Forces of Tunnel Lining in Jet Grouting Reinforced Mud Stratum

        In this study, field measurements of the earth pressures and the main reinforcement strains were carried out at three shield tunnel linings in mud stratum reinforced by vertical jet-grouted piles during tunnel construction and post-construction. The measured strains were used to back-calculate the linings' internal forces. The measured earth pressures and the back-calculated internal forces were compared to corresponding design values. The results indicate that the earth pressure and the internal forces increased rapidly, before exhibiting a fluctuating decrease and finally a long-term slow change, after lining assembly. The vertical jet-grouted piles were beneficial to mobilise the soil arching effect and led to the real earth pressure on the lining significantly smaller than the full overburden pressure, even with a relatively shallow buried depth. The vertical jet-grouted piles intensified the influence of backfill grouting on the earth pressure and caused the lining segments near the backfill grouting holes to continuously bear more radial pressure in the long-term. The resultant internal forces during lining assembly were large, and the Conventional Model failed to consider this element of the internal forces. The findings can provide a reference for the design and the construction of similar projects.
