        In this paper we will examine some previous studies of the PA/SN distinction and show different discourse expressions in Korean that function as SN in dialogal spoken discourse, highlighting 'kulenikka as a dialogal SN discourse marker. As C. Lee (2005) argued, PA and SN in Korean are expressed, respectively, by the connective' -ciman' and '-anird'. H. Lee (2005b) added here '-nuntey' and '-ko'. We also observed the different forms '-na', '-to', '-myenseto' for PA and '-yo' for SN. However, we found that H Lee's (2005b) SN marker '-ko' also shows PA function when correlated with CT. This means that the PA-CT and SN-CF correlations can be a crucial guideline in distinguishing PA/SN (C. Lee 2005). It is generally believed that an SN discourse marker cannot be used in dialogal discourse while PA can. We found that these have interpersonal, social causes: To refute the other's argument in his/her presence is regarded as insolent and causes him/her to lose his/her face. For this reason many languages including Korean don't have single word dialogal SN discourse markers. Lastly, we evidently argued that 'kulenikka', originally a causal connective, is adjusted and extended and now functions as a dialogal SN discourse marker in spoken dialogue.

      • Quantification of Fewer than Ten Copies of a DNA Biomarker without Amplification or Labeling

        <P>Polymerase chain reaction (PCR) is a highly sensitive diagnosis technique for detection of nucleic acids and for monitoring residual disease; however, PCR can be unreliable for samples containing very few target molecules. Here, we describe a quantification method, using force distance (FD) curve based atomic force microscopy (AFM) to detect a target DNA bound to small (1.4-1.9 mu m diameter) probe DNA spots, allowing mapping of entire spots to nanometer resolution. Using a synthetic BCR-ABL fusion gene sequence target, we examined samples containing between one and 10 target copies. A high degree of correlation (r(2) = 0.994) between numbers of target copies and detected probe clusters was observed, and the approach could detect the BCR-ABL biomarker when only a single copy was present, although multiple screens were required. Our results dearly demonstrate that FD curve-based imaging is suitable for quantitative analysis of fewer than 10 copies of DNA biomarkers without amplification, modification, or labeling.</P>

      • Age-dependent decrease of GAD65/67 mRNAs but normal densities of GABAergic interneurons in the brain regions of <i>Shank3</i>-overexpressing manic mouse model

        <P><B>Abstract</B></P> <P>Dysfunction of inhibitory GABAergic interneurons is considered a major pathophysiological feature of various neurodevelopmental and neuropsychiatric disorders. The variants of <I>SHANK3</I> gene, encoding a core scaffold protein of the excitatory postsynapse, have been associated with numerous brain disorders. It has been suggested that abnormalities of GABAergic interneurons could contribute to the <I>SHANK3</I>-related disorders, but the limitation of these studies is that they used mainly <I>Shank3</I> knock-out mice. Notably, <I>Shank3</I>-overexpressing transgenic mice, modeling human hyperkinetic disorders, also show reduced inhibitory synaptic transmission, abnormal electroencephalography, and spontaneous seizures. However, it has not been investigated whether these phenotypes of <I>Shank3</I> transgenic mice are associated with GABAergic interneuron dysfunction, or solely due to the cell-autonomous postsynaptic changes of principal neurons. To address this issue, we investigated the densities of parvalbumin- and somatostatin-positive interneurons, and the mRNA and protein levels of GAD65/67 GABA-synthesizing enzymes in the medial prefrontal cortex, striatum, and hippocampus of adult <I>Shank3</I> transgenic mice. We found no significant difference in the measurements performed on wild-type versus <I>Shank3</I> transgenic mice, except for the decreased GAD65 or GAD67 mRNAs in these brain regions. Interestingly, only GAD65 mRNA was decreased in the hippocampus, but not mPFC and striatum, of juvenile <I>Shank3</I> transgenic mice which, unlike the adult mice, did not show behavioral hyperactivity. Together, our results suggest age-dependent decrease of GAD65/67 mRNAs but normal densities of certain GABAergic interneurons in the <I>Shank3</I> transgenic mice.</P> <P><B>Highlights</B></P> <P> <UL> <LI> The densities of PV<SUP>+</SUP> and SOM<SUP>+</SUP> interneurons are normal in the <I>Shank3</I> transgenic mice. </LI> <LI> The protein levels of GAD65 and GAD67 are normal in the <I>Shank3</I> transgenic mice. </LI> <LI> The mRNA levels of GAD65 and GAD67 are age-dependently decreased in the <I>Shank3</I> transgenic mice. </LI> </UL> </P>

        Diversity and Dynamics of Marine Arenicolous Fungi in Three Seasides of the Korean Peninsula

        Various arenicolous fungal species have been detected from the beach sand in the coastal area. However, little has been revealed regarding their distribution and dynamics. To investigate the overall diversity of marine arenicolous fungi (MAFs) in Korea and whether the composition of MAFs is affected by ocean currents, we isolated and analyzed the fungal community from the western, southern, and eastern seasides of the Korean Peninsula. In total, 603 strains were isolated and identified as 259 species based on appropriate molecular markers for each genus (ITS, BenA, CaM, tef1, and act). The composition of MAFs showed differences among the seasides. Our results indicate that many MAFs inhabit the beach sand on the Korean Peninsula, and the composition of MAFs is also affected by ocean currents flowing along each coast.

      • Intragenic CpG islands play important roles in bivalent chromatin assembly of developmental genes

        <P>CpG, 5'-C-phosphate-G-3', islands (CGIs) have long been known for their association with enhancers, silencers, and promoters, and for their epigenetic signatures. They are maintained in embryonic stem cells (ESCs) in a poised but inactive state via the formation of bivalent chromatin containing both active and repressive marks. CGIs also occur within coding sequences, where their functional role has remained obscure. Intragenic CGIs (iCGIs) are largely absent from housekeeping genes, but they are found in all genes associated with organ development and cell lineage control. In this paper, we investigated the epigenetic status of iCGIs and found that they too reside in bivalent chromatin in ESCs. Cell type-specific DNA methylation of iCGIs in differentiated cells was linked to the loss of both the H3K4me3 and H3K27me3 marks, and disruption of physical interaction with promoter regions, resulting in transcriptional activation of key regulators of differentiation such as PAXs, HOXs, and WNTs. The differential epigenetic modification of iCGIs appears to be mediated by cell type-specific transcription factors distinct from those bound by promoter, and these transcription factors may be involved in the hypermethylation of iCGIs upon cell differentiation. iCGIs thus play a key role in the cell type-specific regulation of transcription.</P>

        Late-Onset Eccrine Angiomatous Hamartoma Associated with a Ganglion Cyst on the Sole of the Foot

        Eccrine angiomatous hamartoma (EAH) is a benign,uncommon, combined vascular and eccrine malformation. Most cases of this disorder have been single or multiple nodules or plaques that appear red, yellow, blue, violaceous,or skin colored. EAH may be congenital or appear later in childhood; it rarely arises during puberty or adulthood. A 52-year-old female patient visited our department for tender subcutaneous cystic tumor on the right sole with a one month history. Histopathologic examination confirmed EAH. During excisional biopsy procedure, mucinous discharges were observed which were histopathologically diagnosed as ganglion. (Ann Dermatol 23(S2) S218∼S221, 2011)

      • Coupling scheme of multicomponent sectional equations and Mason equations via transition rate matrix of hygroscopic growth applied to international standard problem No. 44

        <P><B>Abstract</B></P> <P>Most fission products are released into the containment as aerosol particles during severe accidents. In the conventional codes, two sets of equations are used to analyze the aerosol behaviors in the containment. The first set of equations are multicomponent sectional equations, solutions of which are given with the mass concentrations in each discretized section. The multicomponent sectional equations require very large number of sections to remove the numerical diffusion when hygroscopic growth occurs with other aerosol dynamics simultaneously. For efficient calculations, Mason Equations are used to analyze the hygroscopic growth. The solutions of the equations, on the other hand, are given with the radius of an individual particle. Therefore, it is essential to couple the two equations in rigorous manner for efficient analyses on the aerosol behaviors when hygroscopic growth and other aerosol dynamics occur simultaneously. In this paper, we propose a new formulation for the coupling scheme of multicomponent sectional equations and Mason equations. In the formulation, the two equations are coupled via a transition rate matrix obtained by interpolation of the mass distribution within a section for each component in multicomponent sectional equations and the solutions of Mason equations. In order to validate the formulation, it is applied to the simplified GDE to compare with the analytic solutions. The formulation is also applied to international standard problem No. 44 for the validation with experimental data. Computation time for the formulation is also compared with those of the conventional method. With negligible computation burden as additional, the formulation would analyze the aerosol behaviors efficiently, accurately, and stably when hygroscopic growth occurs with other aerosol dynamics simultaneously.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Efficient, accurate, and stable coupling scheme for the aerosol dynamics in the containment. </LI> <LI> Validation with the analytic solutions of the simplified general dynamic equations. </LI> <LI> Analyses on international standard problem No. 44 to validate with the experimental data. </LI> <LI> Negligible computation burden required for the coupling scheme as additional. </LI> </UL> </P>
