고혈압은 다양한 유전적 요인과 환경적 요인들이 상호작용하여 발병하는 질환으로, 기존의 연구에서 dopamine D3 receptor(DRD3)와 고혈압과의 관련성에 관한 보고들이 있었다. 이에,본 연구에서는 DRD3유전자에 존재하는 Bal I 제한절편길이 다형성이 한국인 집단에서 고혈압과 어떠한 관련성이 있는 지를조사하였다. 환자-대조군 연구를 수행한 결과 이 유전자에 존재하는 다형성은 한국인 집단에서 고혈압과유의한 관련성을 나타내지 않았다. 그러나, 이 다형성을 구성하는 대립 유전자의 빈도를 여러 민족집단의결과들과 비교했을 때. 흑인 집단과 유의한 차이를 나타내었다. 따라서, 이러한 결과는 DRD3 유전자에 존재하는 다형성이 고혈압과의 관련성을 나타내는 지를 정확하게 이해하기 위해서는 혹인 집단을 비롯한다른 민족집단들을 대상으로 하여 광범위한 연구를 수행할 필요가 있을 것으로 생각된다.
First Report of Tobacco mild green mosaic virus Infecting Pepper in Korea

A rod-shaped virus was isolated from pepper showing mild mosic during the winter growing seasons of 2001 and 2002 in Korea. Based on its biological reactions, serological relationships, reverse transcription-poly-merase chain reaction (RT-PCR) using specific primers, and nucleotide sequence analysis of coat protein (CP) gene, the isolated virus was identified as Tobacco mild green mosaic virus (TMGMV) and designated as Korean pepper isolate (TMGMV-KP). Crude sap from infected tissue was mechanically transmitted to various indicator plants, which produced characteristic symptoms of tobamovirus infection. However, no symptom was observed in Gomphorena globosa. In RT-PCR assays with specific primers toy respective detection of TMGMV, Tobacco mosaic virus (TMV), Pepper mild mottle virue (PMMoV), and Tomato mosaic virus (ToMV), a single strong band of about 500 bp in length was produced from the sample used only with TMGMV primers. The amplified DNA was cloned and the nucleotide sequence was determined. Sequence comparisons with the CP gene of other tobamoviruses indicated that TMGMV-KP shared 99.3% identity with TMGMV Japanese isolate and only 59.1, 58.6, and 58.1% identity with TMV, PMMoV and ToMV, respectively. This is the first report of TMGMV in Korea.
Simultaneous Detection of Three Tobamoviruses in Cucurbits by Rapid Immunofilter Paper Assay

A multi-rapid immunofilter paper assay (multi-RIPA) system was prepared for simultaneous diagnosis of three Tobamoviruses, Cucumber green mottle mosaic virus (CGMMV), Kyuri green mottle mosaic virus (KGMMV), and Zucchini green mottle mosaic virus (ZGMMV) in cucurbitaceous crops. Each of these viruses was specifically detected by the multi-RIPA from cucumber, watermelon, zucchini, and bottle gourd inoculated with the three Tobamoviruses singly or in combination. The three viruses could infect cucumber, watermelon, and bottle gourd ; however, CGMMV could not infect zucchini as the latex-coated CGMMV antibody showed a negative reaction in the multi-RIPA of the virus-infected plant extract. When the minimum detection level of multi-RIPA was compared with that of double antibody sandwich-enzyme-linked immunosorbent assay (DAS-ELISA) using CGMMV, the latter was 10 times more sensitive than the former. The detection limit of the multi-RIPA for the purified CGMMV was 50 ng/ml. In a survey of the threeviruses in cucurbits growing in commercial fields in 1999 and 2000, CGMMV was detected in watermelon and cucumber, and ZGMMV was detected only in zucchini growing in plastic houses at the suburbs of Chonju, Korea. However, KGMMV was not found in the commercially growing cucurbit crops in our study, The results suggest that the multi-RIPA can be a simple, rapid, specific and convenient tool to detect simultaneously the Tobamoviruses.
한국어판 사회복지 가치 지향 척도(Professional Opinion Scale)의 신뢰도와 타당도 평가

본 연구는 국내 연구와 실천현장에서 사회복지 실천가들의 사회복지 가치 지향 정도를 가시적으로 측정해 낼 수 있는 적정한 척도가 부재하다는 한계점을 인식하고, 국외에서 개발된 대표적인 사회복지가치 척도인 Professional Opinion Scale(POS)을 우리말로 번안한 후, 신뢰도, 타당도를 평가하여 국내 사회복지 상황에 유용한 척도를 개발하고자 하였다. 확인적 요인분석 결과 한국어판 POS는 영어판 POS와 마찬가지로 4가지 차원(기본 권리에 대한 존중, 자기결정에 대한 지지, 사회적 책임감에 대한 의식, 개인 자유에 대한 책무)으로 구성되었음을 확인하였다. 한국어판 사회복지 가치 지향 척도의 신뢰도는 양호하였으며, 옹호척도와 정적상관관계를 보여, 척도의 수렴타당도가 입증되었다. 본 연구를 통해 사회복지 실천가들의 가치 지향과 관련한 논의가 활성화되고, 실천현장의 서비스와 사회복지가치 지향과의 관련성을 보다 용이하게 파악할 수 있길 기대한다. Although social work values are stressed among social work educators as well as practitioners, empirical studies on values have been very scarce. The objective of this study is to validate a Korean Version of the Professional Opinion Scale(POS). The Korean version was validated with a sample of 325 social worker working in various types of social work agencies. A series of confirmatory factor analysis suggested that 8 items be removed, resulting in 32 items with 4 factors. The Korean version has the same factor structure as the original version of the POS reported by its developer. The Korean version of the POS are found to be a reliable and valid instrument for measuring social work values. However, validation with more representative samples is needed to improve the quality of the Korean version of the POS.

Hypertension is characterized by multiple genetic and environmental factors. To establish the genetic basis of hypertension in Koreans, we investigated the genetic variations of dopamine D3 receptor (DRD3) gene in the Korean patients with hypertension and normotensive controls. There were no significant differences in the genotype and allele frequencies of Bal I RFLP in the DRD3 gene between two groups, respectively (P > 0.05). Our finding shows lack of association between a genetic marker of DRD3 gene and hypertension, suggesting that the genetic variation of DRD3 gene does not playa major role in the determination of hypertension in Korean population. Further studies in other ethnic groups will be required.
Three New Non-reducing Polyketide Synthase Genes from the Lichen-Forming Fungus Usnea longissima

Usnea longissima has a long history of use as a traditional medicine. Several bioactive compounds, primarily belongingto the polyketide family, have been isolated from U. longissima. However, the genes for the biosynthesis of these compounds areyet to be identified. In the present study, three different types of non-reducing polyketide synthases(UlPKS2, UlPKS4, andUlPKS6) were identified from a cultured lichen-forming fungus of U. longissima. Phylogenetic analysis of product templatedomains showed that UlPKS2 and UlPKS4 belong to group IV, which includes the non-reducing polyketide synthases with anmethyltransferase(MeT) domain that are involved in methylorcinol-based compound synthesis; UlPKS6 was found to belong togroup I, which includes the non-reducing polyketide synthases that synthesize single aromatic ring polyketides, such as orsellinicacid. Reverse transcriptase-PCR analysis demonstrated that UlPKS2 and UlPKS4 were upregulated by sucrose; UlPKS6 wasdownregulated by asparagine, glycine, and alanine.

Grapevine leafroll-associated 3 virus (GLRaV-3) and Grapevine fanleaf virus (GFLV) are important viral diseases of grapevine in the world. In this study, the most reliable woody indicator plants were selected for virus indexing. Two grapevines, LN33 (Couderc 1613x vitis berlandieri) and Vitis riparia Gloire, were selected for CLRaV-3 and CFLV graft indexing, respectively. The specific characteristics of Closterovirus isolated from grapevines cultivated in Korea were identified. filamentous virus-like particles only existed in the phloem parenchyma cell. In particular, the vesiculation of mitochondria was observed. This mitochondrial vesicu-lation was considered to be one of the most reliable cytopathic features of Closterovirus. During observation of GFLV-infected Chenopodium quinoa sections, virus-like particles arranged consistently were found forming several layers in cytoplasm. Moreover, virus-like particles in tubules were observed and were associated with plasmodesmata in cytoplasm. This is the first report on cytopathological characteristics of Closterovirus and Nepovirus identified from grapevines in Korea.
