        최근 배달 서비스에서는 배달비 인상에 대한 불만 해소를 위해 노선이 비슷한 이용객의 주문을 함께 배송하는 대신, 배달비를 줄이는 ‘알뜰배달’을 도입했다. 하지만 예정 배달 시간보다 더 지연되는 상황에서 사용자에게 즉각적인 안내와 사과 메시지가 없는 것이 문제로 제기되었다. 이를 개선하기 위하여, 안내 팝업의 UX 라이팅 측면과 이모지 유무에 따른 사용자 경험의 차이를 비교하고자 했다. 실험 결과, 배달 지연 상황에서 간접화행과 이모지를 함께 활용한 경우가 가장 긍정적으로 평가되었으며, 위 유형과 다른 유형들의 차이는 유의미한 것을 확인했다.

        Conservation of intronic minisatellite polymorphisms in the SCK1/SHC2 gene of Hominidae

        The neuronally expressed Shc adaptor homolog SCK1/SHC2 gene contains an unusually high number of minisatellites. In humans, twelve different minisatellite sequences are located in introns of SCK1/SHC2 and ten of them are highly polymorphic. Here we used primers developed for humans to screen ten intronic loci of SCK1/SHC2 in chimpanzee and gorilla, and undertook a comprehensive analysis of the genomic sequence to address the evolutionary events driving these variable repeats. All ten loci amplified in chimpanzee and gorilla contained hypervariable and low-variability minisatellites. The human polymorphic locus TR1 was monomorphic in chimpanzee and gorilla, but we detected polymorphic alleles in these apes for the human monomorphic TR7 locus. When we examined the repeat size among these hominoids, there was no consistent variation by length from humans to great apes. In spite of the inconsistent evolutionary dynamics in repeat length variation, exon 16 was highly conserved between humans and great apes. These results suggest that non-coding intronic minisatellites do not show a consistent evolutionary paradigm but evolved with different patterns among each minisatellite locus. These findings provide important insight for minisatellite conservation during hominoid evolution.

      • Rapid quantitative assessment of Salmonella contamination using real-time PCR assay

        Introduction: Salmonella infection poses a zoonotic threat in one health, and expeditious quantification of Salmonella cells in food and products may provide a useful toolkit for public health and animal industry. In this work, we utilized quantitative real time-PCR to estimate the number of Salmonella cells in a given sample, and attempted to validate the relationship between Cq value and DNA concentration with the number of existing bacterial cells. Materials and Methods: We used Salmonella Typhimurium ATCC 14028 to set a standard curve. After incubating overnight in BPW, cells were serially diluted in 1 ml saline. All diluents were plated on XLD agar plates, and subjected to genomic DNA extraction. DNA concentration for the sample from each diluent was measured using nano-drop. Quantitative real time-PCR was performed using DNA samples from all diluents and primers targeting invA gene. We then compared the Cq values with genomic DNA concentration and the cell number obtained by direct plating. Results: The Cq values increased reciprocally to the gDNA concentration and the number of ATCC 14028 cells in the diluents. Conclusions: Our results suggest reciprocal relationship between Salmonella gDNA concentration and the Cq value. Further investigations may include quantitative real time-PCR with diluted DNA samples.

        Development of Conversational Skills from Late Children to Adolescent

        배경 및 목적: 본 연구는 청소년을 대상으로 연령에 따른 대화 주제운용능력의 발달 차이를 살펴보았다. 방법: 연구대상은 초등 고학년생, 중학생, 고등학생 각 15명씩 총 45명이다. 대상자에게 한림 대화·화용 평가프로토콜에 따라 대화 자료를 수집하고 대화차례 수, 대화차례당 발화 빈도, 전체 주제 수, 주제당 대화차례 빈도, 주제 개시 비율, 주제 유지 비율 주제 변경 비율을 측정하였다. 결과: 대화차례 주고받기 수는 집단 간 유의한 차이가 있었으나 대화차례당 발화빈도는 유의한 차이가 없었다. 전체 주제 수, 주제당 대화차례 빈도는 집단 간 유의한 차이가 있었고, 주제 개시 비율, 주제 유지 비율, 주제 변경 비율 모두 집단 간 유의한 차이가 나타났다. 논의 및 결론: 청소년은 연령이 증가할수록 대화차례 수가 많아지고 대화 안에서 다루어지는 주제 수가 점차 감소하며 한 주제 안에서 더 많은 대화차례를 주고받는 것으로 나타났다. 연령이 증가할수록 주제를 변경하지 않고 한 주제를 일관되게 유지하며 대화를 이어나갔다. 이러한 결과는 청소년의 대화 발달을 이해하는 데 도움을 준다. Objectives: This study aimed to examine developmental differences in conversational skills from childhood to adolescents. Methods: A total of 45 students were participated in the study. Fifteen students were in the higher grade of elementary school, 15 were in middle school and 15 were high school students. Conversation samples were collected by the examiner through the Hallym Conversation & Pragmatic Assessment Protocol. The number of turn-taking, frequency of utterances per turn, the total number of topics, frequency of turns per topic, rates of initiation, maintenance, and change were measured. Results: The results revealed a significant difference in the total number of topics, frequency of turns per topic the rates of initiation, maintenance, and change excluding utterance per turn. Conclusion: The results of the study show that conversational skills develop continuously during late childhood and adolescents. These help us further understanding of conversational skills in adolescent periods.

        Quantitative assessment of the food-borne Salmonella contamination using a quantitative real-time polymerase chain reaction

        Salmonella is one of the most important bacterial pathogens responsible for many zoonotic food-related infectious diseases. Quantitative detection of the foodborne Salmonella contamination in various food sources is therefore critical for preventing the related disease outbreaks. In this study, we developed and evaluated a reliable real-time polymerase chain reaction (RT-PCR) assay to detect the Salmonella contamination quantitatively. The experimental results showed that our invA gene-specific quantitative RT-PCR (qRT-PCR) assay provides a strong correlation between the Cq values and the direct plate counts of Salmonella species in the artificially formulated samples. Further study may be necessary to identify more accurate correlation and equation that can apply to Salmonella spp.

        Probiotics-Mediated Bioconversion and Periodontitis

        Novel bioactive metabolites have been developed through a bioconversion of dairy products or other foods using probiotics isolated from dairy products or other fermented foods. These probiotics-mediated bioconversion (PMB) metabolites show antioxidant, anti-inflammatory, antimicrobial, epithelial barrier, and anticancer activities. In addition, the effect of PMB metabolites in periodontitis is recently reported in several studies. Periodontitis is a chronic inflammatory disease caused by infections, and the tooth support tissue is destroyed. Common treatments for periodontitis include scaling and root planning with systemic antibiotics. However, the overuse of antibiotics has led to the emergence of drug-resistant microorganisms and disturbs the beneficial bacteria, including lactobacilli in the oral cavity. For this reason, PMB metabolites, such as fermented milk, have been suggested as substitutes for antibiotics to reduce periodontitis. This paper reviews the recent studies on the correlation between periodontitis and PMB metabolites and classifies the efficacy of major PMB metabolites for periodontitis. The review suggests that PMB is effective for periodontitis, and further studies are needed to confirm the therapeutic effect of PMB metabolites on periodontitis.

        프로바이오틱스에 의한 알레르기 질환 제어 및 안전성

        Allergic diseases, including atopic dermatitis, asthma, allergic rhinitis, and food allergies, could be caused by dysbiosis that results in an immune system imbalance. The incidence of allergic diseases has been increasing and they are now one of the most common diseases throughout the world. Recently, probiotics have been suggested as an alternative intervention for the prevention and treatment of allergic diseases. Probiotics are endogenous microflora with functional effects within hosts. They have various clinical and immunological capacities and have recently been considered as a supplement for the treatment and prevention of allergic diseases. Probiotic bacteria modulate immune cells such as Th1, Th2, and regulatory T cells that are correlated with protection against atopic dermatitis, however, safety concerns for the use of probiotics have been raised. Therefore, further research is needed to clarify the efficacy and safety of probiotics in the treatment of allergic diseases.

        Antimicrobial Effects of EcoCal<sup>®</sup> and GF Bactostop<sup>®</sup> Formulated in Emulsified Sausages against Lactic Acid Bacteria

        본 연구는 EcoCal<sup>®</sup> (산화 칼슘) 및 GF Bactostop<sup>®</sup> (유기산염 믹스)을 소시지에 사용하였을 때 항균 효과를 확인하였다. 소시지 제조 시, 대조군과 0.1% EcoCal<sup>®</sup> (0.1ECO), 0.1% EcoCal<sup>®</sup>+0.5% GF Bactostop<sup>®</sup> (0.1ECO + 0.5GF), 0.2% EcoCal<sup>®</sup> (0.2ECO) 및 0.2% EcoCal<sup>®</sup>+0.5% GF GF Bactostop<sup>®</sup> (0.2ECO+0.5GF) 등 총 5개 군을 첨가하여 소시지를 제조하였다. 제조가 완료된 소시지를 진공포장하고 10주 동안 10℃에서 저장하며 7-10일 마다 시료를 균질화 하고 tryptic soy agar 및 Lactobacilli MRS agar에 도포하여 호기성 일반세균 및 젖산균을 각각 확인하였다. 그 결과, 0.1ECO+0.5GF 첨가한 소시지가 가장 효과적인 항균 효과를 나타냈으며, 0.1ECO 첨가 소시지가 두번째로 효과적인 항균 효과를 나타냈다(P<0.05). 일반세균수는 대조군, 0.2ECO 및 0.2ECO+0.5GF 첨가 소시지에서는 저장 후 42일까지 점차 증가했지만(P<0.05), 0.1ECO 및 0.1ECO+0.5GF 첨가 소시지에서는 저장 후 49일까지 일반세균의 생장이 억제되었다(P<0.05). 젖산균 수는 대조군, 0.2ECO 및 0.2ECO+0.5GF 첨가 소시지에서 약 49일까지 증가하였으나(P<0.05), 0.1ECO 및 0.1ECO+0.5GF 첨가 소시지에서 젖산균 생장이 억제되었다. 본 결과는 소시지 제형에 0.1% EcoCal<sup>®</sup>+0.5% GF Bactostop<sup>®</sup>을 첨가하면 젖산균의 생장을 억제하여 소시지의 보존 기한을 연장시키는데 유용할 것으로 판단된다. In this study we evaluated the antimicrobial effects of EcoCal<sup>®</sup> (calcium oxide) and GF Bactostop<sup>®</sup> (organic acids mix) in sausages during storage at 10℃. The sausages were formulated with 0.1% EcoCal<sup>®</sup> (0.1ECO), 0.1% EcoCal<sup>®</sup>+0.5% GF Bactostop<sup>®</sup> (0.1ECO+0.5GF), 0.2% EcoCal<sup>®</sup> (0.2ECO), and 0.2% EcoCal<sup>®</sup>+0.5% GF Bactostop<sup>®</sup> (0.2ECO+0.5GF). Total aerobic and lactic acid bacteria in the sausages were enumerated on tryptic soy agar and Lactobacilli MRS agar, respectively, during storage at 10℃ for 10 weeks. The 0.1ECO+0.5GF showed the most effective antimicrobial effects on the sausages, and 0.1ECO showed the second most effective antimicrobial effect. Total aerobic bacterial cell counts gradually increased in the control, 0.2ECO, and 0.2ECO+0.5GF groups, but cell growth was generally inhibited in 0.1ECO by approximately day 42 (P<0.05) and 0.1ECO+0.5GF by approximately day 49 (P<0.05). Lactic acid bacterial cell counts gradually increased in the control, 0.2ECO, and 0.2ECO+0.5GF groups, but the lactic acid bacteria growth was inhibited in 0.1ECO by approximately day 49 (P<0.05) and in 0.1ECO+0.5GF by approximately day 64. These results suggest that using 0.1% EcoCal<sup>®</sup>+0.5% GF Bactostop<sup>®</sup> in sausage formulation is useful for inhibiting lactic acid bacteria growth, thereby extending the shelf-life of the sausage product.
