<P>Protoplasts have been a useful unicellular system for various molecular biological analyses based on transient expression and single cell analysis using fluorescence-activated cell sorting (FACS), widely used as a powerful method in functional genomics. Despite the versatility of these methods, some limits based on low fluorescence intensity of a flow cytometric analysis (FCA) using protoplasts have been reported. In this study, the chloroplast targeting of fluorescent proteins (FPs) led to an eight-fold increase in fluorescence intensity and a 4.5-fold increase of transfection ratio from 14.7% to 65.7% as compared with their targeting into the cytoplasm. Moreover, the plot data of FCA shows that 83.3% of the K-sGFP population is under the threshold level, regarded as a non-transgenic population with background signals, while 65.7% of the K-sGFP population is spread on overall intervals. To investigate the reason underlying this finding, mRNA/protein levels and transfection efficiency were analyzed, and results suggest that mRNA/protein levels and transfection ratio are not much different between K-sGFP and KR-sGFP. From those results, we hypothesized that the difference of fluorescence intensity is not only derived from cellular events such as molecular level or transfection efficiency. Taken together, we suggest that the translocation of FPs into chloroplasts contributes to the improvement of fluorescence intensity in FCA and, apparently, plays an important role in minimizing the loss of the transfected population. Our study could be usefully applicable for highly sensitive FACS and FCA-investigations of green tissue.</P>
2009 개정 6학년 영어 3종 교과서의 프로젝트 활동 분석

The purpose of this study was to analyze project activities of three 6th grade English textbooks for the 2009 revised national curriculum. The activities were analyzed in terms of the following five criteria: language skills, student grouping patterns, learning materials, elements of multiple intelligence and creativity. The results showed that most of the activities integrated four language skills properly, although there were some activities that didn’t integrate all four skills. All three textbooks frequently used cooperative and collaborative activities, and thus group and pair activities accounted for the highest rate among other patterns of organizations. With regard to learning materials, writing supplies such as a pencil and paper were used for project activities most frequently in all three textbooks. The elements of verbal-linguistic and interpersonal intelligence most frequently appeared, whereas the elements of logical-mathematical intelligence and naturalistic intelligence were insufficient in all three textbooks. Among all elements of creativity, linguistic creativity and nonverbal creativity elements appeared most frequently in two textbooks, accounting for the highest rate, while such thinking skills as problem solving, critical thinking, and metaphorical thinking were rarely fostered. Based on the results, some suggestions for the project activities in English textbooks for 6th graders were included.
지방은행의 소비자-브랜드 관계형성에 있어서 소비자경험과 소비자만족의 영향력 분석

본 연구는 전북지역의 대표은행인 JB은행을 대상으로 은행이용에 대한 소비자 경험과 만족이 소비자-브랜드 관계에 연계되는 가를 파악하였다. 분석결과 이용경험 중 브랜드인지경험은 소비자만족에 유의한 영향을 미치지 않는 것으로 나타난 반면, 거래행동경험과 관계경험은 모두 소비자만족에 유의하게 긍정적인 영향을 미치는 것으로 나타났다. 다음으로 소비자만족은 소비자-브랜드관계에서 상호의존유형의 관계에만 유의하게 영향을 미치는 것으로 나타났고 자아연관이나 친밀유형의 관계에는 유의한 영향을 미치지 않는 것으로 나타났다. 더 나아가서 이용경험의 경우 브랜드 인지경험은 소비자-브랜드관계 중 자아연관유형에만 유의하게 부정적인 영향을 미치고, 상호유대나 친밀에는 유의한 영향을 미치지 않는 것으로 나타났으며, 거래행동경험은 소비자-브랜드 관계에 유의한 영향을 미치지 않는 것으로 나타났다. 반면, 관계경험은 자아연관, 상호유대, 친밀유형의 소비자-브랜드 관계에 모두 유의하게 긍정적인 영향을 미치는 것으로 나타났다. 즉, 소비자의 이용경험은 소비자만족에 유의한 영향을 미치고 더 나아가서 부분적이지만 소비자-브랜드 관계에 영향을 미치는 것으로 나타났다. 또한 브랜드 인지경험이나 관계경험의 경우 소비자만족을 매개하지 않고 직접적으로 소비자-브랜드 관계에 영향을 주는 것으로 나타났다. 따라서 경쟁력 있는 지방은행이 되기 위해서는 소비자들에게 거래경험이나 관계 관련 경험을 얻을 수 있도록 많은 체험기회를 제공할 수 있는 다양한 관계마케팅을 방안들을 모색할 필요성이 있다. The purpose of this study was to explain the relationship between consumer experience, consumer satisfaction and consumer-brand relationship for the local banking service in Chonbuk province. For this, consumer experience(brand recognition, transactional behavior, relationship), consumer satisfaction and consumer-brand relationship(self-connection, interdependence, intimacy) was tested as a structural model. This study found some results as follows: 1) Among the types of consumer experience, transactional behavior experience and relationship experience were found to significantly influence to the consumer satisfaction. Furthermore, consumer satisfaction was also found to significantly influence to only one type of consumer-brand relationship(interdependencey). Furthermore, brand recognition experience was found to have some negative effect to the consumer-brand relationship(intimacy) and relationship experience was found to have some positive effect to the consumer-brand relationship(self-connection, interdependence, intimacy). These results might imply that the banking service in the local area should concern for consumer experience and consumer satisfaction to build the long-term relationship with consumers to make a profit. In addition, it might imply that the local consumers might be more attached to the local area and thus perceive more close relationship as a partner. Therefore, the local banking service should develop some strategy to maintain and strengthen the relationship.
코치와 부모의 자율성 지지가 리듬체조 2부 경기 참가자의 기본 심리적 욕구와 내적동기에 미치는 영향

This study was aimed to verify the effect of autonomy supports of parents and coaches on the basic psychological needs and intrinsic motivation of the tier 2 rhythmic gymnastics competition participants, and define mediate effect of the basic psychological needs on the relationship between autonomy supports and intrinsic motivation, so the results can suggest proper direction for improvement, renewal of the tier 2 rhythmic gymnastics competition and promote young athletes. The 280 Tier 2 Rhythmic Gymnastics competition participants, who were not registered as an official athlete to the Korea gymnastics association between the periods from August, 2018 to October, 2018, came in on the study. Frequency Analysis, confirmatory factor analysis, correlation analysis, structure equation modeling were used for data analysis with SPSS 21.0 version program and AMOS 21.0 version program, and the results are shown below. The overall results showed the autonomy support from parents and coaches which is recognized by the tier 2 rhythmic gymnastics participants have positive effect on basic psychological needs and intrinsic motivation, and the autonomy support appears to improve intrinsic motivation through basic psychological needs.
