      • The Analysis and Optimization of Personalized Customization Model of Crowdsourcing Based on GSPN

        The development and customization of new products in crowdsourcing is an important crowdsourcing theory application issue. But as a new business model, there are usually low efficiency, poor effect in the process of enterprises, and in practical application and the combination of businesses. This paper proposes the generalized stochastic Petri net (GSPN) theory in the modeling and performance analysis in crowdsourcing business model, and assesses effectively about the overall model’s time performance and operation efficiency, which has been verified and optimized by 24% in an example. This paper starts from crowdsourcing contest business model to optimize the process and the frame of crowdsourcing through quantitative analysis results, so as to provide a reference for the decision-making party of the business model.

        Identification of a New Bacillus licheniformis Strain Producing a Bacteriocin-Like Substance

        The emergence of antibiotic resistance has spurred a great number of studies for development of new antimicrobials in the past decade. The purpose of this study was to screen environmental samples for Bacillus strains producing potent antimicrobial agents. A new strain, which showed strong antimicrobial activity against Staphylococcus aureus and Salmonella enterica ser. Pullorum, was isolated from soil and designated as B116. This new isolate was identified as Bacillus licheniformis by morphological, biochemical and genetic analyses. The production of bacteriocin-like substance (BLS) started at early exponential phase and achieved highest level at early stationary phase. The BLS was precipitated by ammonium sulfate and its molecular mass was determined as ~4 kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Culture supernatant of the new isolate exhibited antimicrobial activity against both Gram-positive and Gram-negative bacteria, including Bacillus cereus, Staphylococcus aureus, Listeria monocytogenes, Micrococcus luteus, Escherichia coli, and Salmonella spp. The BLS was resistant to heat, acid and alkaline treatment. Activity of the BLS was totally lost after digestion by pronase and partially lost after digestion by papain and lipase. The new isolate and relevant BLS are potentially useful in food and feed applications.

        Rapid Detection of Food-borne Listeria monocytogenes by Real-time Quantitative Loop-mediated Isothermal Amplification

        The purpose of this study was to develop a real-time quantitative loop-mediated isothermal amplication (LAMP) method for the rapid, sensitive, and convenient detection of Listeria monocytogenes in food. The LAMP method could amplify the hlyA gene of L. monocytogenes successfully at 63oC with a loopamp real-time turbidimeter. The detection limits of the LAMP for hlyA gene were 6colony forming units (CFU)/tube. A standard curve was generated for L. monocytogenes LAMP by plotting the graph based different log CFU values of L. monocytogenes and time of positivity through real-time monitoring of the amplication. Then, the LAMP method was employed to test 94 retail food samples effectively. Sensitivity in detection of L. monocytogenes by the LAMP was higher than that of PCR and none of the conventional methodpositive samples was missed by the LAMP method.

        Effect of High-Energy Electro-Pulses on the Compression Deformation Behavior of Ti-6Al-4V Alloy

        Compression tests of Ti–6Al–4V alloy were conducted using an electron universal testing machine with discharge voltages that range from 50 V to 70 V and strain rate of 0.01 s −1 . Isothermal compression at equivalent temperatures was also performed on a universal testing machine with furnace, with the same strain rate. Results show that the deformability of Ti–6Al–4V alloy can be significantly improved by high-energy electro-pulses, and the deformation resistance and yield stress were also decreased. Moreover, the effect of high-energy electropulses on compression mechanical properties of Ti–6Al–4V alloy increased with increasing discharge voltage. However, elongation and deformation resistance of compressions with high-energy electro-pulses were higher than isothermal compressions at equivalent temperatures. This result proved that the deformability improvement of Ti–6Al–4V alloy by high-energy electro-pulses results from the joint effects of temperature and electro-plasticity. Fracture modes in different experimental conditions were analyzed by observing the fracture morphologies of specimens by using a scanning electron microscope. Under high-energy electro-pulses, the micro morphology of compression fracture changes from elongated oval to equiaxed dimples, and the fracture modes transfer from intergranular to transgranular fracture.
