Twenty seafood samples, which are common edible species and commercially important items in Korea, were purchased at the local fisheries markets and were analyzed for polycyclic aromatic hydrocarbons (PAHs) using gas chromatography coupled to mass spectrometer detector (GC/MSD). The levels of sixteen PAHs in seafood from Korean coasts were 161 to 2,243 pg/g wet weight. The highest concentration was found at saury (Coloabis saira) and the lowest level was found at jacopever (Sebastes schlegeli). The concentrations of potentially carcinogeic PAHs of six species were in the range of 9 to 123 pg/g wet weight. The residues of PAHs in fishes from Korean coasts were slightly low of relatively moderate to other countries. There was no correlation between PAH residues and lipid contents in seafood samples. The predominant contributors in fish samples were lower-molecular-weight two and three ring aromatic PAHs such as naphthalene, acenaphthene, flourene and phenanthrene. Filter-feeding organisms like shirimp, crab and topshell were dominated by three- and four- ring aromatic PAHs. The PAH profiles in marine sediments, bivalves, fishes shrimp, crab and topshell according to exposure pathway were compared through factor analysis. The PAH profiles were clearly classified by the difference of species or environmental matrices. This result suggests that most of PAHs within the same sample behave identically in marine environment.

Background: Nutritional status is an important factor which can affect the prognosis of various tumors. The prognostic nutritional index (PNI) which is calculated based on the serum albumin concentration and peripheral blood lymphocyte count, was already reported a useful predictor for postoperative outcome in resectable pancreatic cancer patients. The aim of this study was to validate the prognostic value of PNI in all stage of pancreatic cancer patients. Methods: We retrospectively reviewed 499 patients with pancreatic cancer who were diagnosed and treated in Severance Hospital from January 2006 to December 2011, including all stage of pancreatic cancer. The value of PNI was calculated as 10 × serum albumin (g/dL) + 0·005 × total lymphocyte count (per mm3) at initial diagnosis. Demographic, clinical and laboratory data including performance status (ECOG), tumor marker (CEA and CA19-9), white blood cells, hemoglobin, platelet counts, total protein, albumin and PNI at initial diagnosis were analyzed. Results: The median age of the patients was 63 years (range 34- 88 years) and 289 (57.9%) were male. The study group comprised of localized disease (24.2 %), locally advanced disease (23.4 %) and metastatic disease (52.1 %). The median survival of all patients was 10 months. Univariate analysis indicated that initial lower PNI was signifi cantly associated with shorter survival in localized (p = 0.047), metastatic, stage (p< 0.001) and in the whole group (p<0.001), but not in locally advanced disease (p = 0.355). Multivariate analyses also showed the same fi ndings except in locally advanced disease (p = 0.047 for local disease, p = 0.003 for metastatic stage, and p=0.038 for all stages). Conclusions:The PNI could be a prognostic factor and useful predictor for assessing the nutritional condition of pancreatic cancer patients.

<P>A novel, Gram-stain positive, facultative anaerobic, non-motile and straight to curve rod shaped bacterium, strain LV19(T) was isolated from the larval gut of the rhinoceros beetle, Trypoxylus dichotomus, which was collected from Yeong-dong, Chuncheongbuk-do, South Korea. The colonies of the new isolate were convex, circular, cream white in color and 1-2 mm in diameter after 3 days incubation on Tryptic Soy Agar at 37 °C. Based on the 16S rRNA gene sequence similarity, the new isolate was most closely related to Erysipelothrix inopinata MF-EP02(T), E. rhusiopathiae ATCC 19414 (T) and E. tonsillarum T-305(T) (94.8, 93.8 and 93.7 % similarity, respectively). Strain LV19(T) grew optimally at 37 °C, at pH 8.0 and in the presence of 0.5 % (w/v) NaCl. Oxidase activity and catalase activity were negative. The major cellular fatty acids (>10 %) were C18:2 cis-9,12 (28.9 %), C18:1 cis-9 (22.3 %), C16:0 (22.2 %) and C18:0 (18.5 %). The cell-wall hydrolysates contained ribose as a major sugar. Major polar lipids were phosphatidylglycerol and three unidentified glycolipids. No quinone was detected. The G+C content of the genomic DNA was 36.3 mol%. The levels of DNA-DNA relatedness between strain LV19(T) and all the reference strains were less than 20 %. On the basis of polyphasic evidence from this study, the isolate is considered to represent a novel species of the genus Erysipelothrix, for which the name Erysipelothrix larvae sp. nov. is proposed; the type strain is LV19(T) (=KCTC 33523(T) = DSM 28480(T)).</P>

<P><B>Purpose</B></P><P>p21-activated kinases (PAKs) are involved in cytoskeletal reorganization, gene transcription, cell proliferation and survival, and oncogenic transformation. Therefore, we hypothesized that PAK expression levels could predict the sensitivity of pancreatic cancer cells to gemcitabine treatment, and PAKs could be therapeutic targets.</P><P><B>Materials and Methods</B></P><P>Cell viability inhibition by gemcitabine was evaluated in human pancreatic cancer cell lines (Capan-1, Capan-2, MIA PaCa-2, PANC-1, Aspc-1, SNU-213, and SNU-410). Protein expression and mRNA of molecules was detected by immunoblot analysis and reverse transcription polymerase chain reaction. To define the function of PAK4, PAK4 was controlled using PAK4 siRNA.</P><P><B>Results</B></P><P>Capan-2, PANC-1, and SNU-410 cells were resistant to gemcitabine treatment. Immunoblot analysis of signaling molecules reported to indicate gemcitabine sensitivity showed higher expression of PAK4 and lower expression of human equilibrative nucleoside transporter 1 (hENT1), a well-known predictive marker for gemcitabine activity, in the resistant cell lines. Knockdown of PAK4 using siRNA induced the upregulation of hENT1. In resistant cell lines (Capan-2, PANC-1, and SNU-410), knockdown of PAK4 by siRNA resulted in restoration of sensitivity to gemcitabine.</P><P><B>Conclusion</B></P><P>PAK4 could be a predictive marker of gemcitabine sensitivity and a potential therapeutic target to increase gemcitabine sensitivity in pancreatic cancer.</P>

Concentrations of polychlorinated dibenzo-p-dioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) were determined in marine sediments from Mokpo coastal water of Korea. Total and toxic equivalent (TEQ) concentrations of PCDD/Fs ranged from 19.4 to 175pg/g dry weight and ranged from 0.195 to 2.87 pg WHO-TEQ/g dry weight, respectively. These PCDD/F concentrations were below the safety sediment value (20 pg TEQ/g dry weight) of chronic toxicity. A highly significant correlation between the concentrations of PCDD/Fs and total organic carbon (TOC) in marine sediments was observed. Overall PCDD/F concentrations in the sediments were elevated in rivers and at inner locations close to harbors, indicating that these contaminants derived from local discharges of human activities. PCDD/F concentrations measured in our study were lower than those in several industrialized areas in Korea. Nonparametric multidimensional scaling (MDS) ordination showed that combustion processes from industrial complexes are the major source of PCDD/Fs in Mokpo coastal water.
