<P>Atherosclerosis was previously thought to be a disease that primarily involves lipid accumulation in the arterial wall. In this report, we investigated the effect of <I>Viola mandshurica</I> W. Becker (<I>V. mandshurica</I>) water extract on atherosclerosis in apolipoprotein E deficient (ApoE<TEX>$ ^{-\slash -}$</TEX>) mice. The administration of <I>V. mandshurica</I> to high-fat diet-fed mice reduced body weight, liver weight, and serum levels of lipids (total cholesterol, low-density lipoprotein-cholesterol, triglycerides), glucose, alanine transaminase, and aspartate transaminase. Histopathologic analyses of the aorta and liver revealed that <I>V. mandshurica</I> attenuated atherosclerotic lesions and reduced lipid accumulation, inflammatory responses and fatty acid synthesis. <I>V. mandshurica</I> also increased phosphorylation of adenosine monophosphate-activated protein kinase (AMPK), thereby reducing acetyl-CoA carboxylase (ACC) in liver tissue and inhibiting sterol regulatory element-binding protein 1c (SREBP-1c). <I>V. mandshurica</I> reduced protein expression levels of adhesion molecules (intercellular adhesion molecule-1, vascular cell adhesion molecule-1, and E-selectin) as well as ACC, fatty acid synthase, and SREBP-1c. In addition, quantitative analysis of <I>V. mandshurica</I> by high-performance liquid chromatography revealed the presence of esculetin and scopoletin. Esculetin and scopoletin reduced adhesion molecules in human aortic smooth muscle cells. Our results indicate that the anti-atherosclerotic effects of <I>V. mandshurica</I> may be associated with activation of the AMPK pathway. Therefore, AMPK-dependent phosphorylation of SREBP-1c by <I>V. mandshurica</I> may be an effective therapeutic strategy for combatting atherosclerosis and hepatic steatosis.</P>

BmCu/Zn SOD was isolated from early embryo of Bombyx mori using microarray analysis. The BmCu/Zn SOD gene was observed during the early embryonic stage with the strongest signal found at the unfertilizaion, fertilization and blastoderm stages. The BmCu/Zn SOD gene encodes a protein of 154 amino acids with a calculated Mr of 15 kDa. The deduced amino acid sequence of BmCu/Zn SOD indicated that the residues that form on the Cu/Zn binding site are conserved and that the sequence is a 60% identity to that of M. domestica. In a phylogenetic tree, Bm SOD was also close to Drosophila SODs rather than other insect SODs. The BmCu/Zn SOD gene exists as a single copy in the genome. Transcripts of BmCu/Zn SOD cDNA were identified by northern blot analysis. The expression of the BmCu/Zn SOD gene was observed weakly in most of larvae, pre-pupae, pupae and adult tissues. Also, the BmCu/Zn SOD gene was observed in early embryonic stage. Although the roles of SODs remains to be further elucidated, the high expression of BmCu/Zn SOD gene at before 24 h post fertilization suggests that this gene is of general importance during early embryogenesis in the Bombyx mod.
EST Analysis of Gene Expression in the Germ-Layer Formation Stage of the Bombyx mori

In order to explore the gene expression underlying early embyogenesis, expressed sequence tag (EST) analysis was conducted using a cDNA library made from the 24h embryo of a silkworm, Bombyx mori. A total of 8,928 clones were screened plasmid with cDNA insert and have partially sequenced the 5´ ends to generate 2,446 assembled sequences. The annotation of 2,446 high-quality ESTs by a BLAST search, revealed that 1,516 (62%) of the sequences represented known genes. Over 2,400 ESTs consisted of 1,568 single ESTs (singlets) and 878 redundant ESTs (contigs). The functional groups of those sequences with matches in the database (P<E-4) were constructed according to their putative molecular functions, biological process and cellular components. Seventy-five potential clones, associated with cell communication and development of the biological process, were selected in order to analyze their expression patterns by dot-blot assay. Most of them showed a predominant expression in the expected 24h embryo, the germ-layer formation stage. This study should provide information about the overall gene expression profiles of the Bombyx mori early embryo.

본 연구는 아동 보호 관련 전문가들을 대상으로 아동보호조치 시 아동청문권 이슈에 대한 델파이조사와 계층화분석법을 활용하여 아동보호조치 시 아동 청문권에 대한 타당성과 중요성을 파악하고 그 결과를 바탕으로 아동 청문권 실현을 위한 구체적인 개선방안과 아동복지적 함의를 제시하고자 하였다. 전문가 대상의 델파이조사는 1, 2차에 나누어 진행되었으며, 쌍대비교를 통해 도출한 우선순위 항목은 아동청문권의 보장 기준, 보호조치 시 아동 청문권 보장을 위해 필요한 요인, 보호조치 시 아동청문권 보장을 도울 수 있는 전문가, 아동 청문권 관련 문제가 발생하는 원인 요인, 보호조치의 각 단계 별 아동의견 청취의 중요도, 해외의 아동 청문권 보장제도, 보호조치 과정 중 의견상충 시 의견 중시, 보호조치 시 사법체계 내 개선사항을 포함하여 총 8개이다. 이를 바탕으로 아동을 권리의 주체로 바라보는 관점, 아동청문권에 관한 편견과 오해 해소, 궁극적인 아동보호의 시작점을 포함한 전반적인 아동청문권의 사회적 인식 증대, 아동보호 전문인력 및 예산 보충, 관련 법⋅지침⋅매뉴얼 신설, 아동청문 환경마련 등을 통한 아동 친화적 청문환경 조성, 분리 조치 이후 사례⋅사후관리 지속의 필요성이 주요 함의로 제시되었다. Using Delphi survey and Analytic Hierarchy Process (AHP) methods, this study explored how Korean child welfare experts viewed the child’s right to be heard in Korean child welfare services and what issues must be considered in relation to safeguarding the child’s right to be heard in Korean child welfare services, so that we can draw implications for child welfare services and policy. The Delphi survey was conducted in two phases, and priority items identified through pairwise comparisons included eight issues: 1) criteria for ensuring the child’s right to be heard, 2) factors necessary to guarantee the child’s right to be heard during child welfare services, 3) experts who can assist in ensuring the child’s right to be heard, 4) causal factors for issues related to the child’s right to be heard, 5) the importance of soliciting the child’s opinion at each stage of child welfare services, 6) applicability of international systems for ensuring the child’s right to be heard to Korean child welfare system, 7) the importance of respecting opinions in case of conflicting opinions, and 8) areas for improvement within the legal system during child welfare services. We asked Korean child welfare experts to set priorities among the eight items in conflictual situations or in child welfare adminstrative processes. Based on the findings, we drew child welfare implications such as enhancing a perspective that views the child as the subject of rights, ameliorating biases and misunderstandings on the child’s right to be heard, augmenting placement of specialized personnel and budgets for child protection, establishing relevant laws, guidelines, and manuals, and creating a child-friendly environment similar to legal system including ensuring ways to hear the child’s opinion regardless of the child’s age and maturity.
