        Assessment of acute and repeated pulmonary toxicities of oligo(2-(2-ethoxy)ethoxyethyl guanidium chloride in mice

        Oligo(2-(2-ethoxy)ethoxyethyl guanidinium chloride (PGH) and polyhexamethyleneguanidine phosphate (PHMG-P) are cationic biocides containing a guanidine group. Direct exposure of the lungs to PHMG-P is known to induce pulmonary inflammation and fibrotic changes. Few studies have assessed the pulmonary toxicity of PGH, another member of the guanidine family. In this study, we assessed the acute and repeated toxicity of PGH and PHMG-P to compare the pathological progression induced by both chemicals. PGH (1.5 mg/kg) or PHMG (0.6 mg/kg) was instilled intratracheally to mice once or three times every 4 days; subsequently, cytokine levels were quantified and a histopathological examination was performed. To verify the toxic mechanism of PGH, we quantified cell viability and cytokine production induced by PGH or PHMG-P in the presence or absence of anionic material in cells. Instillation of PGH and PHMG-P into the mouse lung increased cytokine production, immune cell infiltration, and pulmonary fibrotic changes. These pathological changes were exacerbated over time in the singleand the repeated-dose PHMG-P groups, but were resolved over time in the PGH groups. PGH or PHMG-P showed cytotoxic effects, IL-1β secretion, and ROS production in a dose-dependent manner in human cell lines. However, the co-treatment of anionic materials with PGH or PHMG-P significantly reduced these toxic responses, which confirmed that the cation of PGH disrupted the plasma membrane via ionic interaction, as observed for PHMG-P. In addition, we suggest the disruption of plasma membrane as a molecular initiating event of cationic chemicals-induced adverse outcomes when exposed directly to the lungs.

        Time course of polyhexamethyleneguanidine phosphate-induced lung inflammation and fibrosis in mice

        <P><B>Abstract</B></P> <P>Pulmonary fibrosis is a chronic progressive disease with unknown etiology and has poor prognosis. Polyhexamethyleneguanidine phosphate (PHMG-P) causes acute interstitial pneumonia and pulmonary fibrosis in humans when it exposed to the lung. In a previous study, when rats were exposed to PHMG-P through inhalation for 3 weeks, lung inflammation and fibrosis was observed even after 3 weeks of recovery. In this study, we aimed to determine the time course of PHMG-P-induced lung inflammation and fibrosis. We compared pathological action of PHMG-P with that of bleomycin (BLM) and investigated the mechanism underlying PHMG-P-induced lung inflammation and fibrosis. PHMG-P (0.9 mg/kg) or BLM (1.5 mg/kg) was intratracheally administered to mice. At weeks 1, 2, 4 and 10 after instillation, the levels of inflammatory and fibrotic markers and the expression of inflammasome proteins were measured. The inflammatory and fibrotic responses were upregulated until 10 and 4 weeks in the PHMG-P and BLM groups, respectively. Immune cell infiltration and considerable collagen deposition in the peribronchiolar and interstitial areas of the lungs, fibroblast proliferation, and hyperplasia of type II epithelial cells were observed. NALP3 inflammasome activation was detected in the PHMG-P group until 4 weeks, which is suspected to be the main reason for the persistent inflammatory response and exacerbation of fibrotic changes. Most importantly, the pathological changes in the PHMG-P group were similar to those observed in humidifier disinfectant-associated patients. A single exposure of PHMG-P led to persistent pulmonary inflammation and fibrosis for at least 10 weeks.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A single exposure of PHMG-P increases inflammatory response for 10 weeks. </LI> <LI> Collagen deposition and fibroblast proliferation were also observed until 10 weeks. </LI> <LI> Inflammasome activation is proposed to be one reason for the pathological action. </LI> </UL> </P>

      • Polyhexamethyleneguanidine phosphate induces cytotoxicity through disruption of membrane integrity

        <P><B>Abstract</B></P> <P>Polyhexamethyleneguanidine phosphate (PHMG-P) is a polymeric biocide with a guanidine group. It has multiple positive charges in physiological conditions due to nitrogen atom in the guanidine and this cationic property contributes antimicrobial effect by disrupting cell membranes. To determine whether the cationic nature of PHMG-P results in cytotoxicity in human cell lines, anionic compounds were treated with PHMG-P. The cytotoxic effect was evaluated with ROS production and HMGB1 release into media. To verify the protection effect of anion against PHMG-P-induced cell death <I>in vivo</I>, a zebrafish assay was adopted. In addition, membrane disruption by PHMG-P was evaluated using fluorescein diacetate and propidium iodine staining. As a result, anionic substances such as DNA and poly-<SMALL>L</SMALL>-glutamic acids, decreased PHMG-P induced cell death in a dose-dependent manner. While HMGB1 and ROS production increased with PHMG-P concentration, the addition of anionic compounds with PHMG-P reduced the ROS production and HMGB1 release. The mortality of the zebrafish increased with PHMG-P concentration and co-treatment of anionic compounds with PHMG-P decreased mortality in a dose-dependent manner. In addition, FDA and PI staining confirmed that PHMG-P disrupts plasma membrane. Taken together, a cationic property is considered to be one of the main causes of PHMG-P-induced mammalian cell toxicity.</P>

      • 계량서지학적 분석을 통한 두피 관리 분야의 연구 동향

        인간의 두피는 환경 변화에 따라 탈모를 비롯한 다양한 문제가 발생하기 쉬우므로 계속 적인 관리가 필요할 것이다. 최근에는 탈모에 관한 연구가 발표되고 있으며, 홈 케어의 일 종인 두피 관리에 대한 인식이 대중화 되고 있다. 이와 관련하여 두피 관리에 대한 필요성 과 관심은 증가되고 있지만 두피 관리에 대한 전반적인 연구 성과는 미비한 상태라고 할 수 있다. 두피 관리를 연구하는 학문의 체제와 연구 동향은 그 분야에 있는 연구자들의 사회적 관계와 커뮤니케이션 유형 및 변화를 분석하여 알 수 있으며, 또한 연구자들의 학술 문헌을 계량적으로 분석하여 알 수 있을 것이다. 이에 대해 본 연구는 두피 관리에 대한 국내 학위논문의 분류와 학술논문에 수록된 논문의 서명을 연도별로 구분하고 계량서지학 적으로 분석한다. 그 목적은 두피 관리에 대한 연구의 흐름이 어떤 분야와 융합하며, 어떤 분야와 연관이 있는지 분석하기 위함이다. 연구 결과 두피와 모발의 동시출현이 125건으로 가장 많은 나타났으며, 이는 모발 건강 은 두피가 많은 영향을 미치는 것으로 보인다. 그러나 두피마사지와 두피 관리는 동시출현이 한 건도 없었으며, 두피상태와 모발상태 또한 모발관리, 탈모 실태연구, 효과연구, 스트레스, 만족도, 두피마사지를 통한 분석 결과 동시출현이 한 건도 없었다. 이는 현재의 두피 상태나 모발 상태에 대한 연구보다 두피개선을 위한 연구 및 탈모나 모발 관리 방법에 더 많은 연구가 진행되었음을 알 수 있다. 결론적으로 본 연구에서 두피마사지와 두피관리는 동시출현이 한 건도 없었던 것과 두피와 두피관리, 모발과 두피, 모발과 모발관리, 두피와 탈모, 두피와 여성이 다소 많은 공 출현이 있었다. 따라서 두피 관리는 일반적으로 두피 마 사지가 포함되는 개념으로 볼 수 있으며, 두피는 모발에 많은 영향을 미치며 모발은 두피를 따로 생각할 수 없는, 두피의 건강이 모발의 건강으로 이루어진다는 것으로 해석할 수 있다. Human scalp in response to changes in the environment, since various problems are likely to occur, including hair loss, is necessary ongoing management. So in recent years, research on hair loss have been published, recognition of the scalp care which is a kind of home care has been popularized. Although increasing the need and interest of scalp care, the overall findings of the scalp care may be a state of deficiency. System and research trends in the study of the scalp care is possible to know to analyze and change social relationships and communication of the kind of researchers in the field. and it will be able to analyze to bibliometric researchers. In this study, to classify the papers for the scalp of care, bibliometrics to analyze based on the year the signing of papers that have been recorded in the academic papers. The purpose is to fuse flow studies scalp care and which areas, in order to analyze be associated with several areas, and presenting the direction of the scalp care industry from the analyzed word systematic and research trends. In results, the simultaneous appearance of scalp and hair is most common co-occurrence of 125. The health of the hair seems to be the scalp will give a lot of influence. However, management of the scalp massage and scalp, there was no simultaneous appearance. The results of analysis the state of the scalp and the state of the hair, also through. hair loss study, study of the effects, stress satisfaction analysis, there was no co-occurrence. This research on the current state of scalp conditions or hair, the research and the management method of hair removal or hair for the improvement of the scalp, it is possible to know that more study was conducted. In conclusion, in this study, management of the scalp massage and scalp, simultaneous appearance did not even scratch. Scalp and scalp care, hair and scalp, hair and hair management, scalp and hair loss, scalp and women there were some many appearance. Accordingly, the scalp care be seen concept in generally that includes the scalp massage, Scalp hair which had a lot of influence of the hair, can not be considered the scalp separately, it can be interpreted as health of the scalp are performed health of the hair.
