        Wound healing potential of antibacterial microneedles loaded with green tea extracts

        This study evaluates the utility of an antibacterial microneedle composed of green tea (GT) extract and hyaluronic acid (HA), for the efficient delivery of GT. These microneedles have the potential to be a patient-friendly method for the conventional sustained release of drugs. In this study, a fabrication method using a mold-based technique to produce GT/HA microneedles with a maximum area of ~50mm<SUP>2</SUP> with antibacterial properties was used to manufacture transdermal drug delivery systems. Fourier transform infrared (FTIR) spectrometry was carried out to observe the potential modifications in the microneedles, when incorporated with GT. The degradation rate of GT in GT/HA microneedles was controlled simply by adjusting the HA composition. The effects of different ratios of GT in the HA microneedles were determined by measuring the release properties. In HA microneedles loaded with 70% GT (GT70), a continuous higher release rate was sustained for 72h. The in vitro cytotoxicity assays demonstrated that GT/HA microneedles were not generally cytotoxic to Chinese hamster ovary cells (CHO-K1), human embryonic kidney cells (293T), and mouse muscle cells (C2C12), which were treated for 12 and 24h. Antimicrobial activity of the GT/HA microneedles was demonstrated by ~95% growth reduction of gram negative [Escherichia coli (E. coli), Pseudomonas putida (P. putida), and Salmonella typhimurium (S. typhimurium)] and gram positive bacteria [Staphylococcus aureus (S. Aureus) and Bacillus subtilis (B. subtilis)], with GT70. Furthermore, GT/HA microneedles reduced bacterial growth of infected wound sites in the skin and improved wound healing process of skin in rat model.

      • The Arabidopsis NAC transcription factor NTL4 participates in a positive feedback loop that induces programmed cell death under heat stress conditions

        Programmed cell death (PCD) is an integral component of plant development and adaptation under adverse environmental conditions. Reactive oxygen species (ROS) are one of the most important players that trigger PCD in plants, and ROS-generating machinery is activated in plant cells undergoing PCD. The membrane-bound NAC transcription factor NTL4 has recently been proven to facilitate ROS production in response to drought stress in Arabidopsis. In this work, we show that NTL4 participates in a positive feedback loop that bursts ROS accumulation to modulate PCD under heat stress conditions. Heat stress induces NTL4 gene transcription and NTL4 protein processing. The level of hydrogen peroxide (H<SUB>2</SUB>O<SUB>2</SUB>) was elevated in 35S:4ΔC transgenic plants that overexpress a transcriptionally active nuclear NTL4 form but significantly reduced in NTL4-deficient ntl4 mutants under heat stress conditions. In addition, heat stress-induced cell death was accelerated in the 35S:4ΔC transgenic plants but decreased in the ntl4 mutants. Notably, H<SUB>2</SUB>O<SUB>2</SUB> triggers NTL4 gene transcription and NTL4 protein processing under heat stress conditions. On the basis of these findings, we conclude that NTL4 modulates PCD through a ROS-mediated positive feedback control under heat stress conditions, possibly providing an adaptation strategy by which plants ensure their survival under extreme heat stress conditions.

        Immobilization of hemoglobin on functionalized multi-walled carbon nanotubes-poly-l-histidine-zinc oxide nanocomposites toward the detection of bromate and H<sub>2</sub>O<sub>2</sub>

        A novel biocompatible sensing strategy has been developed based on functionalized multi-walled carbon nanotube (f-MWCNT), poly-l-histine (P-l-His), and ZnO nanocomposite film for the immobilization of hemoglobin (Hb). The direct electron transfer properties and bioelectrocatalytic activity of the Hb in f-MWCNT-P-l-His-ZnO composite film is further investigated. The apparent heterogeneous electron transfer rate constant (k<SUB>s</SUB>) of Hb confined to f-MWCNT-P-l-His-ZnO nanocomposite is found to be 5.16s<SUP>-1</SUP> using Laviron's equation. Moreover, the surface coverage concentration (Γ) of the electroactive Hb in the f-MWCNT-P-l-His-ZnO film is estimated to be 1.88x10<SUP>-9</SUP>molcm<SUP>-2</SUP>. The fabricated electrochemical biosensor based on the immobilized Hb revealed a fast response time (<3s) with a wide linear range (4-18,000μM and 2-15,000μM) and detection limit (as low as 0.01μM and 0.30μM) for the electrocatalytic determination of a mediator-free H<SUB>2</SUB>O<SUB>2</SUB> and bromate under optimal experimental conditions. The ca. apparent Michaelis-Menten constant is 0.14mM, which indicates that the Hb has a high affinity to H<SUB>2</SUB>O<SUB>2</SUB>. The high sensitivity, good reproducibility, and long-term stability of the proposed nanocomposite film indicates that it can serve as an electrode for the development of an amperometric H<SUB>2</SUB>O<SUB>2</SUB> and bromate-based biosensor. The proposed third-generation biosensor was successfully applied to milk and urine samples for the detection of H<SUB>2</SUB>O<SUB>2</SUB> and bromate.

        The metastatic carcinoma of the brain comprises about 15 to 25 per cent of total brain tumors and most of them originate from the lung. Additional primary sites such as the breast, kidney, large intestine, prostate are rare and the stomach as primary site is very rare. We experienced a case of metastatic signet-ring cell carcinoma to the brain originated from the stomach, which was thought as subdural hematoma at the time of admission.

        Thickness-dependent solar power conversion efficiencies of catalytic graphene oxide films in dye-sensitized solar cells

        The catalytic effects of graphene oxide (GO) films were investigated for dye-sensitized solar cell (DSSC). The GO film consisted of a networked structure of two phases of GO with intercalated H<SUB>2</SUB>O and exfoliated GO. A maximum solar power conversion efficiency (η) of 2.30% was obtained for the ultrathin GO film. This η is higher than that of other reported types of DSSCs (0.84%) and organic solar cells (1.1%) with graphene electrodes. The catalytic effect of the GO film is attributed to the good ion permeability along the interlayer hydrophilic spaces; the electrons present in these spaces on the exfoliated GO were supplied to the iodide ions.

      • 多發性骨髓腫의 臨床的 觀察

        1977년 8월부터 1980년 7월까지 인제의대 부속 백 병원에서 13예의 자발성 골수종 환자를 관찰한 바 남녀의 비는 1.8 : 1이었으며 남자의 평균 연령은 51.4세, 여자의 평균 연령은 53.6세였다. 임상증상은 골동통이 77%에서 있었으며 류마치스성관절염, 퇴행성관절염, 신질환등으로 초진된 경우가 많았다. 혈액소견상 빈혈이 92%에서, 증가된 적혈구침강속도가 전체환자에서, 말초혈액소견상 형질세포화한 림파구가 31%에 있었다. 생화학검사는 총단백의 증가가 62%, 고칼슘혈증이 31%, 질소혈증이 69%, 고뇨산증이 89%, hypergammaglobulinemia가 38%에서 나타났다. 12예에서 전기영동을 시행한 결과 M-단백이 γ-region에 8예, β-region 3예, α2-region에 1예로 나타났다. 면역전기영동은 9예에서 시행결과 IgG 44%, IgA 22%, IgD 22%, light chain 11%였다. 방사선과적 검사는 85%에서 골용성병변이 있었으며 두개골 척추골 늑골 골반등의 순으로 있었다. 치료는 melphalan+prednisone이나 cyclophosphamide+prednisone을 사용하여 좋은 결과를 얻을 수 있었으며 melphalan 또는 cyclphosophamide 등의 alkylating agent와 prednisone 병용에 반응을 보이지 않은 환자에서는 제 2차적인 치료법으로 CCNU, Adriamycin 및 vincrstine을 이용한 병용요법을 시행하여 효과를 보았다. From August 1977 to July 1980, thirteen cases of multiple myeloma had been admitted to Paik Hospital, Inje Medical College. The ratio of male to female was 1.8 1 and the mean age was 51.4 years old in man and 53.6 years old in woman. The most common clinical findings were bone pain and tenderness. There were many cases which had been impressed as rheumatoid arthritis, degenerative arthritis, or renal problem at first medical examination. Hematologic findings were anemia in 92%, elevated E.S.R. in 100%, and plasmacytoid lymphocyte in 31%. Blood chemistry showed increased total protein in 62%, azotemia in 69%, hyperuricemia in 89%, hyperglobulinemia in 38%, and hypercalcemia in 31%. Serum protein electrophoresis in 12 cases revealed that M-spike was present in γ-region in 8 cases, in β-region in 3 cases and in α2-region in one case. Immunoelectrophoresis in 9 cases showed Ig G in 44%, Ig A in 22%, Ig D in 22% and light chain in 11%. Osteolytic lesions were noted in 85% and involed skull, spine and ribs in order. All 13 patients were treated with alkylating agent (melphalan or cyclophosphamide) and prednisone. Patients, who did not show response to the alkylating agent, were treated with CCNU, Adriamycin and vincristine in combination. Of 13 patients, six patients were lost to follow-up. One patient who had severe renal failure at the tinge of admission was dead during chemotherapy in conjuction with dialysis. Three patients survived 11, 13 and 32 moths each. The remaining three patients are still alive and well from 3+ months to 25+ months after initiation of chemotherapy.

        컴퓨터제어에 의한 자동초음파 탐상장치에 관한 연구

        Since the nuclear power plants in Korea have been operated in 1979, the nondestructive testing(NDT) of pressure vessels and/or piping welds plays an important role for maintaining the safety and integrity of the plants. Ultrasonic method is superior to the other NDT method in the viewpoint of the detectability of small flaw and accuracy to determine the locations, sizes, orientations, and shapes. As the service time of the nuclear power plants is increased, the radiation level from the components is getting higher. In order to get more quantitative and reliable results and secure the inspector from the exposure to high radiation level, automation of the ultrasonic equipments has been one of the important research and development(R & D) subject. In this research, it was attempted to visualize the shape of flaws presented inside the specimen using a Modified C-Scan technique. In order to develope Modified C-Scan technique, an automatic ultrasonic scanner and a module to control the scanner were designed and fabricated. IBM-PC/XT was interfaced to the module to control the scanner. Analog signals from the SONIC MARK II were digitized by Analog-Digital Converter(ADC 0800) for Modified C-Scan display. A computer program has been developed and has capability of automatic data acquisition and processing from the digital data, which consist of maximum amplitudes in each gate range and location. The data from Modified C-Scan results was compared with shape from artificial defects using the developed system. Focal length of focused transducer was measured. The automatic ultrasonic equipment developed through this study is essential for more accurate, reliable, and repeatable ultrasonic experiments. If the scanner are modified to meet to appropriate purpose, it can be applied to automation of ultrasonic examination of nuclear power plants and helpful to the research on ultrasonic characterization of the materials.
