        콩의 발아에 따른 카르니틴 함량변화

        콩나물 콩의 발아에 따른 크기별, 부위별 유리카르니틴, 단쇄 및 중쇄카르니틴, 총 카르니틴의 함량변화를 측정하였다. 콩나물 콩은 건조시료 그램당 약 136 nmol의 총 카르니틴을 함유하고 있었고, 발아가 진행됨에 따라 유리카르니틴, 단쇄 및 중쇄카르니틴, 총 카르니틴이 모두 증가하였다. 부위별 카르니틴 함량은 콩나물의 머리에 해당하는 자엽 부위에는 유리카르니틴 함량이 많고 뿌리의 분열조직 부위에 단쇄 및 중쇄카르니틴이 많았다. 이와같은 결과를 종합하여 볼 때 카르니틴 함량은 콩의 발아정도와 발아부위에 따라 상이함을 알 수 있었다. The changes of carnitine levels including nonesterified-carnitine (NEC), acid-soluble acylcarnitine (ASAC) and total carnitine (TC) were investigated in developing soybean sprouts. The concentrations of carnitines were determined in ungerminated and germinated soybean seeds, and in dissected axis segments and cotyledone of the germinated sprouts. Soybean seeds contain 136 nmol of TC per gram dry weight. The contents of NEC, ASAC, and TC were increased during the germination of soybean seeds. The concentrations of NEC and ASAC were highest in cotyledone and in meristematic tissues, respectively. These data indicate that developmental differences of carnitine levels do exist in plants, and that in developing soybean sprouts the levels of NEC and ASAC are highest in the cotyledone and in the youngest meristem, respectively.

        Acanthopanax senticosus Extract Prepared from Cultured Cells Improves Lipid Parameters in Rats Fed with a High Fat Diet

        Acanthopanax senticosus was grown by a novel, proprietary method, of culturing isolated cells in a bioreactor. An extract from the cells was evaluated for its effect on lipid metabolism in rats fed a high fat diet. Male Sprague-Dawley rats (n=6) were fed either an AIN-76 diet (control, NDCon), control diet plus Acanthopanax senticosus extract (ND+Ex), a modified AIN-76 diet supplemented with 20% beef tallow (high fat, HFCon), or a high fat diet plus Acanthopanax senticosus extract (HF+Ex), for 5weeks. Body weight gain was significantly higher in the HFCon group than the NDCon group. Feed consumption was significantly lower, but energy intake higher, in the groups fed high fat diets compared with the groups fed control diets. Serum HDL-cholesterol concentrations were significantly increased but serum LDL-cholesterol concentrations were decreased in the groups fed the Acanthopanax senticosus extract. Abdominal fat accumulation, serum leptin levels were significantly higher in the HFCon group than the other groups. Carnitine palmitoyltransferase-I (CPT-I) mRNA levels were increased in the groups fed Acanthopanx senticosus extract. These results suggest that supplementation of cell cultured Acanthopanax senticosus extract regulates CPT-I mRNA levels in liver, has an effect on the normalization of lipids in rats fed a high fat diet.

      • Effects of Regular Endurance Exercise or Acute-exercise and Rest on the Levels of Lipids, Carnitines and Carnitine Palmitoyltransferase-I in rats

        The effects of regular endurance exercise, or acute-exercise and rest on the levels of lipids, carnitines and carnitine palmitoyltransferase-I (CPT-I) were investigated in male Sprague-Dawley rats. The rats were exercise trained on a treadmill for 60 min per day for 60 days (long-term trained, LT), or non-trained for 59 days (NT) and exercised for 60 min on the 60th day. In NT rats, the levels of serum nonesterified carnitine (NEC), acidsoluble acylcarnitine (ASAC), and total carnitine (TONE) increased significantly during the post-exercise recovery period (PERP). In LT rats, ASAC, and TCNE, which increased right after the 60 min running session decreased to the levels of pre-exercise during the PERP. The levels of skeletal muscle ASAC in NT rats, which increased significantly by the acute-exercise, decreased to the pre-exercise levels during the PERP. However, the ASAC level in LT rats reached its peak at 4 h after running for 60 min. Liver triglyceride (TG) and total lipids (TL), which increased by the acute-exercise, decreased to the pre-exercise levels during the PERP in both NT and LT rats. CPT-I activity in NT rats increased significantly after 1 h of a 60-min exercise and slowly decreased to pre-exercise levels during the PERP. However, the CPT-I activity in LT rats, which increased significantly by the 60 min exercise, decreased slowly and reached its pre-exercise level within 8 h of the PERP. Northern blot analysis showed that the changes of CPT-I activities during the PERP coincided with changes in CPT-I mRNA levels. This study shows that both regular endurance exercise, and acute-exercise and rest, can influence differently the levels of carnitines, lipids and CPT-I in rats. The results suggest that regular endurance exercise, rather than the acute-exercise, can change effectively the distributions of carnitines, lipids and CPT-I in rats during exercise and rest.

        Carnitine and/or GABA Supplementation Increases Immune Function and Changes Lipid Profiles and Some Lipid Soluble Vitamins in Mice Chronically Administered Alcohol

        This study evaluated the effects of carnitine and/or GABA supplementation on immune function, lipid profiles and some vitamins in mice chronically administered alcohol. BALB/c mice were fed with either AIN-76 diet (N), control diet plus alcohol (4 g/㎏ bw, E), E plus 0.5 g/㎏ bw carnitine (EC), E plus 0.5 g/㎏ bw GABA (EG), or E plus 0.5 g/㎏ bw carnitine plus 0.5 g/㎏ bw GABA (ECG) for 6 weeks. Administrations of the carnitine and/or GABA prevented alcohol-induced increases in triglyceride concentrations in serum and liver. However, there was no difference among the supplemented groups. Serum vitamin E concentration was higher in mice supplemented with EC and EG, but not in mice given ECG. Phagocytic activity of peritoneal macrophages was increased in EG group compared with E group. The subpopulations of murine splenocyte’s TH cells were increased significantly in EC and ECG groups. These data suggest that immune function, lipid profiles and some immune-related lipid soluble vitamins were positively changed by supplementation of carnitine or GABA, but do not show any synergistic effect of mixed supplementation.

        Q 열에 의한 감염성 심내막염 2예

        Q fever is a zoonosis caused by Coxiella burnetii, Presenting as acute and chronic illness and it has been reported worldwide. Acute Q fever is usually asymptomatic or mild and self-limiting, but infective endocarditis is one of the most serious complications of chronic Q fever and can be fatal. Known risk factors for Q fever endocarditis are valvular heart disease, immunocompromised hosts, and pregnancy. There have been some reports on Q fever in Korea but there exists no report on Q fever endocarditis. We have experienced 2 cases of Q fever with underlying valvular heart disease; both Patients came to the hospital for evaluation of prolonged fever. Although Q fever and Q fever endocarditis are rare in Korea, Q fever endocarditis should be considered in the differential diagnosis of patient with infective endocarditis when causative microorganism cannot be identified.

        Gamma - Aminobutyric Acid and/or Carnitine Supplementation Alters Lipid and Some Immune Related Nutrient Levels in Mice

        This study investigated the effects of carnitine and/or γ-aminobutyric acid (GABA) supplementation on lipid profiles and some immune related nutrient in mice. Balb/c male mice were orally treated with either an AIN-76 diet (Con), a control diet plus carnitine (CS, 0.5 g/kg bw), a control diet plus GABA (GS, 0.5 g/kg bw) or a control diet plus carnitine plus GABA (CGS, 0.25 g/kg bw, respectively) for 6 weeks. There were no significant differences in feed consumption, energy intake, body weight gain or feed efficiency ratio among the groups during the experimental period. However, abdominal fat deposits were smaller in CS, GS and CGS groups compared with the Con group. Serum and liver triglycerides also were lower in CS, GS and CGS and serum total cholesterol was significantly lower in the CGS group compared with the Con group. Serum LDL cholesterol was lower in the CGS group and liver HDL cholesterol was significantly higher in the CS group compared with Con group. In serum, stearic acid and selecholeic acid were lower, but arachidic acid was higher in the GS group. Liver stearic acid was higher but oleic acid lower in CGS group compared with Con group. In carnitine supplemented groups, serum and liver nonesterified carntine (NEC), acidsoluble acylcarnitine (ASAC), total carnitine (TCNE) concentrations were higher in only the CS group, not CGS group. Serum vitamin A and E concentrations were not different among the groups. These results may suggest that carnitine and/or GABA supplementation improves lipid profiles in mice, but did not affect the immune-related nutrients that we measured under the experimental conditions of this study.
