In this paper, two statistical methods were applied to optimize medium components to improve the production of the milk-clotting enzyme by Bacillus amyloliquefaciens D4. First, wheat bran juice, skim milk powder, and Na2HPO4 were shown to have significant effects on D4 enzyme production using the Plackett?Burman experimental design. Subsequently, an optimal medium was obtained using the Box?Behnken method, which consisted of 3.31 g/l of skim milk powder, 5.0 g/l of sucrose, 0.1 g/l of FeSO4?7H2O, 0.1 g/l of MgSO4?7H2O, 0.1 g/l of MnSO4?2H2O, 0.1 g/l of ZnSO4?7H2O, 1.52 g/l of Na2HPO4, and 172.45 g/l of wheat bran juice. With this optimal medium, the milk-clotting enzyme production was remarkably enhanced. The milk-clotting enzyme activity reached 3,326.7 SU/ml after incubation of 48 h, which was 1.76-fold higher than that of the basic medium, showing that the Plackett?Burman design and Box?Behnken response surface method are effective to optimize medium components, and B. amyloliquefaciens D4 possessed a high rennet-producing capacity in the optimal medium.

Seventeen compounds derived from traditional Chinese medicines (TCMs) were tested for their antiviral activity against porcine reproductive and respiratory syndrome virus (PRRSV) in vitro. Visualization with the cytopathologic effect (CPE) assay and the 3-(4, 5-dimethyithiazol- 2-yl)-2,5-diphenyltetrazolium bromide test were used to determine the 50% cytotoxic concentration (CC50) and 50% effective concentration (EC50) in cultured Marc-145 cells. Among the tested compounds, chlorogenic acid and scutellarin showed potential anti-PRRSV activity. The EC50 values were 270.8 ± 14.6 μg/ml and 28.21 ± 26.0 μg/ml and the selectivity indexes were >5.54 and 35.5, respectively. The time-of-addition and virucidal assay indicated that the anti-PRRSV activity of the two compounds could be due to their inhibiting the early stage of virus replication and/or inactivating the virus directly. The inhibition of the virus attachment was not observed in the adsorption inhibition assay. The inhibition ratios of chlorogenic acid and scutellarin were, respectively, 90.8% and 61.1% at the maximum non-cytotoxic concentrations. The results have provided a basis for further exploration of their antiviral properties and mechanisms in vivo. We believe that the chlorogenic acid and scutellarin have a great potential to be developed as new anti-PRRSV drugs for clinical application.
Purification and Characterization of an Extracellular Protease from Bacillus pumilus CN8

The protease produced by a Bacillus pumilus CN8 strain was purified by DEAE-Cellulose-52 ion exchange. It has a molecular weight of approximately 96,920 Dalton. In the present study, this protease showed strong activity over a broad range of pH (6.5-9.5) and temperature from 40℃ to 60℃, and the protease performed the maximal activity at pH 7.3 at 42℃. The effect of metal ions on protease activity showed that K+ could slightly increase the protease activity, and other ions such as Zn²+, Fe²+, Na+, Ca²+, Mg²+ had no significant activation or inhibition to the protease (P > 0.05), and the more important is that Cu²+, Mn²+, Sn²+, Cd²+ had a strong inhibitory effect on the protease activity.
Over-expression of GmHAL3 modulates salt stresses tolerance in transgenic arabidopsis

The halotolerance protein HAL3, also known as SIS2, is a yeast protein that regulates the cell cycle and tolerance to salt stress through inhibition of the Ppz1 type 1 protein phosphatase. Although the roles of HAL3 have been demonstrated during the growth, development, and stress adaptation of Arabidopsis thaliana and Nicotiana tabacum, the function of HAL3 in other plant species, including soybean (Glycine max), has not been elucidated. In this study, GmHAL3a and GmHAL3b were isolated from Glycine max, and their roles were analyzed. GmHAL3a and GmHAL3b transcripts were detected in the roots, stems, leaves and seeds, with higher levels in the roots, and were induced by sodium chloride (NaCl), lithium chloride (LiCl), sorbitol, cold and ABA treatment. Overexpression of GmHAL3a or GmHAL3b in Arabidopsis accelerated the onset of flowering and resulted in more vigorous seed germination and increased tolerance to NaCl, LiCl, and sorbitol stress in seedlings, compared with wild type (WT) and empty vector control (VC) plants. Transgenic Arabidopsis plants accumulated proline and eliminated superoxide radical (O2 −) in response to the stress. In addition, transcription levels of the stress-related genes RD22 and P5CS1 were substantially higher in transgenic Arabidopsis than in WT and VC plants. Taken together, the data indicate that GmHAL functions as a positive regulator of the response to salt, lithium cations and sorbitol stress.

The pyrrolo[2,3-b]pyridine core structure, a bioisostere of quinolones, is found in several molecules that possess important biological activity. We describe here a new, concise, three-step synthesis of pyrrolo[2,3-b]pyridines starting from L-alanine. A series of 4,7-dihydro-4-oxo-1H-pyrrolo[2,3-b]pyridine-5-carboxylic acid derivatives, which have not been previously reported, were synthesized using this approach.
Methylenetetrahydrofolate Reductase C677T Polymorphism and Cervical Cancer Risk: a Meta-Analysis

Background: Methylenetetrahydrofolate reductase (MTHFR) is a key enzyme in the metabolism of folate, and the role of MTHFR C677T polymorphism in cervical carcinogenesis is still controversial. Method: We performed a meta-analysis of all relevant case-control studies that examined any association between the C677T polymorphism and cervical cancer risk. We estimated summary odds ratios (ORs) with their confidence intervals (CIs) to assess links. Results: Finally, 10 studies with a total of 2113 cervical cancer cases and 2804 controls were included. Results from this meta-analysis showed that significantly elevated cervical cancer risk was associated with the MTHFR T allele in the Asian population under conditions of two genetic comparison models (for TT vs. CC, OR = 1.37, 95%CI 1.00-1.87, P = 0.050; for TT vs. TC+CC: OR = 1.34, 95%CI 1.01-1.77, P = 0.039). However, there was no obvious association between the MTHFR C677T polymorphism and cervical cancer risk in the other populations. Conclusion: The MTHFR C677T polymorphism is associated with cervical cancer risk in Asians, while any possible link in the Caucasian population needs further studies.
Research on Aggregation and Propagation of Self-Similar Traffic in Satellite Network

It has already been confirmed that the traffic in high-speed terrestrial network presents self-similarity, but there is little research on self-similarity of traffic in satellite network. Considering time-varying network topology and link status, this paper analyzes the aggregation and propagation of self-similar traffic between nodes in satellite network. Furthermore, a sort of special network node called ground gateway is modeled, based on which the characteristics of the output traffic that the input traffic from terrestrial network passes gateway into satellite network are analyzed. Theoretically analyses demonstrate that after aggregation and propagation between satellite nodes, traffic is still self-similar, and the self-similarity of the output traffic generated by gateway from terrestrial network to satellite network is more often than not weakened.

Yunnan Baiyao is a famous Chinese medicine that has long been directly applied to wounds to reduce bleeding, pain, and swelling without causing infection. However, little is known about its ability to prevent infection. The present study aimed to assess in vitro the anti-virulence activity of an aqueous extract of Yunnan Baiyao (YBX) using Pseudomonas aeruginosa as a pathogenic model. We found that a sub-MIC (2.5 mg/ml) of YBX can efficiently interfere with the quorum-sensing (QS) signaling circuit. Real-time polymerase chain reaction analysis showed that a sub-MIC of YBX downregulated the transcriptions of lasR, lasI, rhlR, and rhlI,which resulted in global attenuation of QS-regulated virulence activities, such as biofilm formation, and secretion of LasA protease, LasB elastase and pyocyanin. Further, YBX reduced the motility of P. aeruginosa related to QS, and impaired the formation of biofilms. These results suggest that YBX may possess global inhibitory activity against the virulence of P. aeruginosa and that YBX may also exhibit antimicrobial activity in vivo. The present study suggests that Yunnan Baiyao represents a potential source for isolating novel, safe, and efficacious antimicrobial agents.

Multiple Node Flip Fast-SSC Decoding Algorithm for Polar Codes Based on Node Reliability

This paper presents a fast-simplified successive cancellation (SC) flipping (Fast-SSC-Flip) decoding algorithm for polar code. Firstly, by researching the probability distribution of the number of error bits in a node caused by channel noise in simplified-SC (SSC) decoder, a measurement criterion of node reliability is proposed. Under the guidance of the criterion, the most unreliable nodes are firstly located, then the unreliable bits are selected for flipping, so as to realize Fast-SSC-Flip decoding algorithm based on node reliability (NR-Fast-SSC-Flip). Secondly, we extended the proposed NR-Fast-SSC-Flip to multiple node (NR-Fast-SSC-Flip-ω) by considering dynamic update to measure node reliability, where ω is the order of flip-nodes set. The extended algorithm can correct the error bits in multiple nodes, and get good performance at medium and high signal-to-noise (SNR) region. Simulation results show that the proposed NR-Fast-SSC-Flip decoder can obtain 0.27dB and 0.17dB gains, respectively, compared with the traditional Fast-SSC-Flip [14] and the newly proposed two-bit-flipping Fast-SSC (Fast-SSC-2Flip-E2) [18] under the same conditions. Compared with the newly proposed partitioned Fast-SSC-Flip (PA-Fast-SSC-Flip) ( =4) [18], the proposed NR-Fast-SSC-Flip-ω (ω=2) decoder can obtain about 0.21dB gain, and the FER performance exceeds the cyclic-redundancy-check (CRC) aided SC-list (CRC-SCL) decoder (L=4).
