        A Zero Sequence Voltage Injection Method for Cascaded H-bridge D-STATCOM

        Load variations on a distribution line result in voltage fluctuations at the point of common coupling (PCC). In order to keep the magnitude of the PCC voltage constant at its rated value and obtain zero voltage regulation (ZVR), a D-STATCOM is installed for voltage correction. Moreover, the ZVR mode of a D-STATCOM can also be used to balance the source current during unbalanced loading. For medium voltage and high power applications, a D-STATCOM is realized by the cascaded H-bridge topology. In the ZVR mode, the D-STATCOM may draw unbalanced current and in this process is required to handle different phase powers leading to deviations in the cluster voltages. Zero sequence voltage needs to be injected for ZVR mode, which creates circulating power among the phases of the D-STATCOM. The computed zero sequence voltage and the individual DC capacitor balancing controller help the DC cluster voltage follow the reference voltage. The effectiveness of the control scheme is verified by modeling the system in MATLAB/SIMULINK. The obtained simulations are further validated by the experimental results using a dSPACE DS1106 and five-level D-STATCOM experimental set up.

        Effect of Non-Idealities on the Design and Performance of a DC-DC Buck Converter

        In this study, the performance of a direct current (DC)-DC buck converter is analyzed in the presence of non-idealities in passive components and semiconductor devices. The effect of these non-idealities on the various design issues of a DC-DC buck converter is studied. An improved expression for duty cycle is developed to compensate the losses that occur because of the non-idealities. The design equations for inductor and capacitor calculation are modified based on this improved expression. The effect of the variation in capacitor equivalent series resistance (ESR) on output voltage ripple (OVR) is analyzed in detail. It is observed that the value of required capacitance increases with ESR. However, beyond a maximum value of ESR (r<sub>c,max</sub>), the capacitor is unable to maintain OVR within a specified limit. The expression of r<sub>c,max</sub> is derived in terms of specified OVR and inductor current ripple. Finally, these theoretical studies are validated through MATLAB simulation and experimental results.

        Identification and validation of putative biomarkers by in silico analysis, mRNA expression and oxidative stress indicators for negative energy balance in buffaloes during transition period

        Objective: Transition period is considered from 3 weeks prepartum to 3 weeks postpartum, characterized with dramatic events (endocrine, metabolic, and physiological) leading to occurrence of production diseases (negative energy balance/ketosis, milk fever etc). The objectives of our study were to analyze the periodic concentration of serum beta-hydroxy butyric acid (BHBA), glucose and oxidative markers along with identification, and validation of the putative markers of negative energy balance in buffaloes using in-silico and quantitative real time-polymerase chain reaction (qRT-PCR) assay. Methods: Out of 20 potential markers of ketosis identified by in-silico analysis, two were selected and analyzed by qRT-PCR technique (upregulated; acetyl serotonin o-methyl transferase like and down regulated; guanylate cyclase activator 1B). Additional two sets of genes (carnitine palmotyl transferase A; upregulated and Insulin growth factor; downregulated) that have a role of hepatic fatty acid oxidation to maintain energy demands via gluconeogenesis were also validated. Extracted cDNA (complementary deoxyribonucleic acid) from the blood of the buffaloes were used for validation of selected genes via qRTPCR. Concentrations of BHBA, glucose and oxidative stress markers were identified with their respective optimized protocols. Results: The analysis of qRT-PCR gave similar trends as shown by in-silico analysis throughout the transition period. Significant changes (p<0.05) in the levels of BHBA, glucose and oxidative stress markers throughout this period were observed. This study provides validation from in-silico and qRT-PCR assays for potential markers to be used for earliest diagnosis of negative energy balance in buffaloes. Conclusion: Apart from conventional diagnostic methods, this study improves the understanding of putative biomarkers at the molecular level which helps to unfold their role in normal immune function, fat synthesis/metabolism and oxidative stress pathways. Therefore, provides an opportunity to discover more accurate and sensitive diagnostic aids.
