Despite advances in cancer therapy, the clinical outcome of patients with gastric cancer remains poor, largely due to tumor heterogeneity. Thus, finding a hidden vulnerability of clinically refractory subtypes of gastric cancer is crucial. Here, we report that chemoresistant gastric cancer cells rely heavily on endocytosis, facilitated by caveolin-1, for survival. caveolin-1 was highly upregulated in the most malignant stem-like/EMT/mesenchymal (SEM)-type gastric cancer cells, allowing caveolin-1-mediated endocytosis and utilization of extracellular proteins via lysosomal degradation. Downregulation of caveolin-1 alone was sufficient to induce cell death in SEM-type gastric cancer cells, emphasizing its importance as a survival mechanism. Consistently, chloroquine, a lysosomal inhibitor, successfully blocked caveolin-1-mediated endocytosis, leading to the marked suppression of tumor growth in chemorefractory gastric cancer cells in vitro, including patient-derived organoids, and in vivo. Together, our findings suggest that caveolin-1-mediated endocytosis is a key metabolic pathway for gastric cancer survival and a potential therapeutic target.
사용자 Context 파악을 통한 TV 시청 행태 연구

본 연구는 TV 의 새로운 역할과 기회영역을 찾기 위한 목적으로 시작되었다. 이를 위해 User Diary 방법론을 이용하여 가정 내 Context 를 분석하였다. User Diary 는 ‘TV 시청 Context’라는 특수성을 고려하여 기존의 문답식 실시간 Diary 가 아닌 회고의 형식으로 작성하였다. 이를 통해 분 단위의 자세한 Context 와 그에 따른 사용자의 감정 변화를 추적할 수 있었다. Diary 분석을 위해서 사용자의 문장을 기준에 맞게 유형화하여 개인별 Context Map 을 그린 후, 이를 중첩하여 가족들의 시청 Context 특성을 도출하였다. 이를 통해 세 가지 시간(평일 오전, 평일 저녁, 주말 낮)별 시청 특성과 세 가지 시청 행태(라디오형 시청, 개인화, 자녀 시청 통제 욕구)를 도출하였다. This research has been started from the purpose of exploring the new role and possibility of the TV. In order for this, through the ‘User Diary’ methodology this research analyzed the contexts of the living styles at home. Since we consider the contextualized traits of people’s TV watching patterns, this user diary was completed as the retrospective way which is different from the traditional user diary using a real time Q&A. Through this methodology, the change of the detailed context happening in minutes, and of the TV viewer’s emotions are able to be traced. For the analyses of the diary, we tried to find a pattern from the reviewer’s sentences and drew the context map according to such a pattern and finally layered them to find contextualized traits of TV watching in the family. Consequently, this study results in three kinds of time(weekday mornings, weekday evenings, weekend daytimes) and three types of TV watching style (watching TV like listening radio, individualization, desire of controlling kids’ TV watching).

LY6D is crucial for lipid accumulation and inflammation in nonalcoholic fatty liver disease

Nonalcoholic fatty liver disease (NAFLD) is a serious metabolic disorder characterized by excess fat accumulation in the liver. Over the past decade, NAFLD prevalence and incidence have risen globally. There are currently no effective licensed drugs for its treatment. Thus, further study is required to identify new targets for NAFLD prevention and treatment. In this study, we fed C57BL6/J mice one of three diets, a standard chow diet, high-sucrose diet, or high-fat diet, and then characterized them. The mice fed a high-sucrose diet had more severely compacted macrovesicular and microvesicular lipid droplets than those in the other groups. Mouse liver transcriptome analysis identified lymphocyte antigen 6 family member D (Ly6d) as a key regulator of hepatic steatosis and the inflammatory response. Data from the Genotype-Tissue Expression project database showed that individuals with high liver Ly6d expression had more severe NAFLD histology than those with low liver Ly6d expression. In AML12 mouse hepatocytes, Ly6d overexpression increased lipid accumulation, while Ly6d knockdown decreased lipid accumulation. Inhibition of Ly6d ameliorated hepatic steatosis in a diet-induced NAFLD mouse model. Western blot analysis showed that Ly6d phosphorylated and activated ATP citrate lyase, which is a key enzyme in de novo lipogenesis. In addition, RNA- and ATAC-sequencing analyses revealed that Ly6d drives NAFLD progression by causing genetic and epigenetic changes. In conclusion, Ly6d is responsible for the regulation of lipid metabolism, and inhibiting Ly6d can prevent diet-induced steatosis in the liver. These findings highlight Ly6d as a novel therapeutic target for NAFLD.
후자린산(Fusaric acid) 생성 Fusarium 종의 신속 검출 PCR

후자린산은 Fusarium이 생성하는 독소로서 다른 Fusarium 독소와 함께 독성을 증진시킬 수 있다. 후자린산 독소를 특이적으로 검출하기 위해 후자린산의 생합성유전자 중 전사인자인 FUB10을 증폭하는 프라이머를 제작하였다. Fub10-f와 Fub10-r 프라이머쌍으로 PCR을 수행했을 때, F. oxysporum, F. proliferatum, F. verticillioides, F. anthophilum, F. bulbicola, F. circinatum, F. fujikuroi, F. redolens, F. sacchari, F. subglutinans, F. thapsinum에서 약 550 bp의 단일밴드가 증폭되었으며 이들은 모두 후자린산을 생성하는 것으로 알려졌다. 반면 트라이코쎄신을 생성하는 종에서는 FUB10 특이 밴드가 증폭되지 않았다. 후자린산은 푸모니신을 생성하는 종에서 함께 생성될 수 있기 때문에 FUB10 프라이머와 푸모니신 생합성유전자인 FUM1 프라이머를 이용한 multiplex PCR을 수행하였다. 그 결과 푸모니신 생성종인 F. proliferatum과 F. verticillioides에서 밴드가 모두 증폭되었으며 이는 두 가지 독소를 생성할 수 있는 종에서 동시 검출이 가능함을 시사하였다. Fusaric acid is a mycotoxin produced by species of the fungus Fusarium and can act synergistically with other Fusarium toxins. In order to develop a specific detection method for fusaric acid-producing fungus, PCR primers were designed to amplify FUB10, a transcription factor gene in fusaric acid biosynthetic gene cluster. When PCR with Fub10-f and Fub10-r was performed, a single band (~550 bp) was amplified from F. oxysporum, F. proliferatum, F. verticillioides, F. anthophilum, F. bulbicola, F. circinatum, F. fujikuroi, F. redolens, F. sacchari, F. subglutinans, and F. thapsinum, all of which were known for fusaric acid production. Whereas the FUB10 specific band was not amplified from Fusarium species known to be trichothecene producer. Because production of fusaric acid can co-occur in species that also produce fumonisin mycotoxins, we developed a multiplex PCR assay using the FUB10 primers as well as primers for the fumonisin biosynthetic gene FUM1. The assay yielded amplicons from fumonisin producers such as F. proliferatum and F. verticillioides, allowing for the simultaneous detection of species with the genetic potential to produce both types of mycotoxins.
