Anti-tumor Activity of Paclitaxel Prodrug Conjugated with Polyethylene Glycol

The purpose of this study was to develop a method for increasing the solubility of paclitaxel in water to reduce its toxicity and make the drug more feasible for chemotherapy. A series of highly water soluble paclitaxel polyethylene glycol (PEG) esters were synthesized and evaluated for their anti-tumor activity and toxicity. The solubility of 7-polyethylene glycol paclitaxel carbonate 5 was 840 mg/mL and the acute toxicity ($LD_{50}$) was 286 mg/kg. Because of its reduced toxicity, compound 5 showed a dramatic reduction of tumor volume without any loss of animals in long-term treatment (daily consecutive injections for 15 days).

Antimutagenic and AntileukemicActivities of Aloes vera L.

We investigated that the extract of Aloe vera L. and its fractions exert antimutagenic activity against Salmonella typhimurium TA98 and TA100, and antileukemic effect against K562 human leukemia cell line. The aqueous ethanolic extract of A. vera L. was revealed to have antimutagenic effect on the AF-2 (2-(2-furyl)-3-(5-nitro-2-furyl)-acrylamide) in Salmonella mutation assay. Among the three fractions (fractions A, B and C) separated by silica gel chromatography, fraction C (50 ㎍/plate) exhibited the greatest antimutagenic effect on the AF-2 with inhibition rate of 84 and 90% in Salmonella typhimurium TA98 and TA100, respectively. The fraction C (500 ㎍/㎖) inhibited the growth of K562 human leukemia cell line by 93% in MTT assay. However, the components of A. vera L. did not exhibit cytotoxic effect against MDBK bovine normal kidney in MTT assay.

Exo-polysaccharide isolated from the culture of Grifola frondosa was modified by sodium periodate (NaIO₄) and sodium chlorite (NaClO₂) to delete polysaccharide part and phenolic compound, respectively, and was investigated what effect has each part of exo-polysaccharide against 2,2’-azobis(2-amidinopropane) dihydrochloride (AAPH)-induced oxidative stress in porcine kidney epithelial cells (LLC-PK1). Oxidative stress on LLC-PK1 cell was measured by cell viability, lipid peroxidation, superoxide dismutase (SOD), and glutathione peroxidase (GSH-px) activity. Exposure of LLC-PK1 cells to 1 mM AAPH for 24 hr resulted in significant decrease in cell viability, SOD, and GSH-px action, and significant increase in lipid peroxidation. The treatment of exo-polysaccharide and NaIO₄ modified sample protected LLC-PK1 cells from AAPHinduced cell damage such as cell viability, lipid peroxidation, SOD, and GSH-px activity in a dose dependant manner (10, 100, and 500 ㎍/㎖). However, the treatment of NaClO₂ modified sample did not affect for cell viability, lipid peroxidation, SOD, and GSH-px activity. The antioxidant activity of exo-polysaccharide was significantly decreased on AAPH-induced LLC-PK1 cell system when phenolic compound was deleted. The antioxidant activity was significantly correlated with the content of phenolic compound of exo-polysaccharide.

Ulmus davidiana, Formitella fraxinea, and Aloe vera extracts were detected to have inhibitory effects against topoisomerase I at treatment of Spg. Ulmus davidiana and Aloe vera extracts were found to show inhibitory effect similar to camptothecin, Formitella fraxinea extract was found to have weak activity. We also found the potential use of those extracts as a radiation sensitizer. Radiosensitizing effect at combination treatment was increased more than 2 times at single treatment of radiation, Ulmus davidiana or Formitella fraxinea extracts. Ulmus davidiana and Formitella fraxinea extracts were found to have significant radiosensitizing effect on test tumor cell line. But, Aloe vera extract was not detected to have activity as a radiosensitizer. Ulmus davidiana and Formitella fraxinea extracts are potent radiosensitizers on tumor cell lines and should be considered for further study of active compounds.

We investigated the effects of fucoidan and lutein against dextran sulfate sodium (DSS)-induced mice colitis. Evaluations were made of the body weight, histological index such as crypt injury and inflammation score, biochemical factor such as serum amyloid (SAA) and MPO level data. The combination of fucoidan and lutein reduced the score of crypt injury and inflammation and markedly showed more decrease of the SAA and MPO levels than 5-ASA group. In addition, each sample of fucoidan and lutein was reduced the level of IL-6 which is stimulated by a lipopolysaccharide (LPS) in HT-29 cell line in vitro. Therefore, fucoidan and lutein may be useful as a dietary substance for preventing inflammatory bowel disease in humans.
