        Validation of the Korean version of the Reflective Practice Questionnaire in clinical clerkship of Korean medical students

        Purpose: This study aims to verify whether the Reflective Practice Questionnaire (RPQ) developed by Priddis and Rogers is valid in the Korean context to identify the level of reflection of medical students in clinical practice. Methods: A total of 202 third- and fourth-year medical students from seven universities participated in the study. After receiving approval for use from the authors, a survey was conducted on the students through an adaptation process. The original scale consists of 10 factors with 40 items. The Self-efficacy in Clinical Performance Scale (SECP), Korean Self-reflection and Insight Scale (K-SRIS), and Reflection-in-Learning Scale (RinLS) were used to validate the scale. Exploratory factor, confirmatory factor, correlation, and reliability analyses were used for data analysis. Results: As a result of exploratory factor analysis, 10 subfactors were extracted (Kaiser-Meyer-Olkin=0.856, Bartlett’s test: χ2=5,044.337, degrees of freedom=780, p<0.001). Among the 40 items, one that showed a high overlapping load for other factors was excluded. As a result of confirmatory factor analysis, the 10-factor structure model was found suitable (χ2=1.980, comparative fit index=0.859, Tucker-Lewis index=0.841, root mean square error of approximation=0.070). As a result of the criterion validity test, most of the subfactors of the Korean version of the RPQ (K-RPQ) showed a positive correlation with K-SRIS, RinLS, and SECP. The reliability of 10 subfactors was satisfactory, ranging from 0.666 to 0.919. Conclusion: The K-RPQ was confirmed to be a reliable and valid tool to evaluate the level of reflection among Korean medical students in clinical clerkship. This scale can be used as a tool to provide feedback on each student’s level of reflection in clinical clerkship.

        소의 초기 배 발생단계별 Telomeric DNA 함량 및 Telomerase Activity 분석

        목 적: 소 수정란의 telomeric DNA 함량 및 텔로머레이스 활성도 (telomerase activity)를 분석하여 초기 배아형성과정에서 텔로미어의 변화 양상을 규명하고자 하였다. 연구방법: 소의 초기 배 발생단계별 체외수정란을 대상으로 양적형광보인법 (Q-FISH)을 이용한 telomeric DNA의 함량 분석과 Telomeric Repeat Amplification Protocol (TRAP)로서 텔로머레이스 활성도를 분석하였다. 결 과: 소의 초기 수정란의 telomeric DNA 함유율은 발생과정이 진행됨에 따라 유의적인 양적 증가를 나타내었다. 배반포배의 질 (quality)에 따른 텔로미어의 함량 차이는 나타나지 않았다. 소 수정란의 8 세포기, 상실배기 및 배반포기의 모든 초기 배 단계에서 텔로머레이스 활성도를 보였으며, 상실배에서 증가하여 배반포배에서 가장 강한 활성도를 나타내었다. 결 론: 소의 초기 배아형성과정이 진행되면서 텔로미어의 함량 및 텔로머레이스 활성도는 점진적으로 증가하고 이들 간에는 정 (positive)의 상관관계를 가진다. Telomeric DNA 함량의 증가를 유지하기 위하여 텔로머레이스의 활성화는 필수적인 것으로 보여진다. 배반포기 배아에서 높은 텔로미어 함량과 강한 텔로머레이스 활성도는 이 시기가 배발생 및 분화과정 중 가장 세포적 활성도가 높은 시점임을 의미하는 것으로 사료된다. Objective: This study was carried out to analyze the amount of telomeric DNA and telomerase activity in early bovine embryos. Methods: The amount of telomeric DNA in early bovine embryos at the 8 cell, morula and blastocyst stages was analyzed by Quantitative Fluorescence In Situ Hybridization (Q-FISH) technique using a bovine telomeric DNA probe. Telomerase activity was analyzed by Telomeric Repeat Amplification Protocol (TRAP assay). Results: The relative amount of telomeric DNA in early bovine embryos was gradually increased from 8 cell to blastocyst stage. It was not significantly associated with the grade of embryo quality. While telomerase activity was detected in the early bovine embryos at these stages, it significantly increased at morula stage and showed maximum activity at the blastocyst stage. Conclusion: The amount of telomeric DNA and the telomerase activity of bovine embryos increase during the progression of early embryogenesis, suggesting a positive correlation between telomeric DNA and telomerase activity. The telomerase activity seems to increase to maintain the levels of telomeric DNA through embryo development which are required for extensive cell division.

        <P>We present a specially designed materials chemistry that provides ultrathin adhesive layers with persistent tacky surfaces in solid, nonflowable forms for use in transfer printing and related approaches to materials and micro/nanostructure assembly.         THP-1 의 ROS 측정을 통한 당류의 p-toluenediamine sulfate PTD에 의한 피부 감작성 완화능 평가

        Allergic contact dermatitis (ACD) is undesired side effect caused by chemical allergens in the development of cosmetics, especially, hair coloring product. It has been reported that reactive oxygen species (ROS) produced by contact allergen plays a crucial role in dendritic cell maturation and contact hypersensitivity. In this study, we evaluated anti-allergic effect of various saccharides on hair dye precursor, p-toluenediamine sulfate (PTD) by use of ROS production in THP-1 cell. 80 μg/ml PTD showed significant increase in the level of ROS production in THP-1 cell. In addition, co-treatment with xylose or galactose showed dose-dependent ROS decrease at a range from 0.1 % to 1 %. Moreover, similar inhibition effect on PTD by xylose or galactose was also observed when the level of CD86 expression on THP-1 was evaluated. Base on these results, in vitro alternative method based on ROS production in THP-1 cells could be applied to screen the substances mitigating allergic contact dermatitis and xylose or galactose could be used in hair color product to reduce skin sensitization potential of aromatic amine hair dye precursors without any disturbing dyeing hair.

      • Fabrication and application of flexible, multimodal light-emitting devices for wireless optogenetics

        The rise of optogenetics provides unique opportunities to advance materials and biomedical engineering, as well as fundamental understanding in neuroscience. This protocol describes the fabrication of optoelectronic devices for studying intact neural systems. Unlike optogenetic approaches that rely on rigid fiber optics tethered to external light sources, these novel devices carry wirelessly powered microscale, inorganic light-emitting diodes (μ-ILEDs) and multimodal sensors inside the brain. We describe the technical procedures for construction of these devices, their corresponding radiofrequency power scavengers and their implementation in vivo for experimental application. In total, the timeline of the procedure, including device fabrication, implantation and preparation to begin in vivo experimentation, can be completed in ∼3–8 weeks. Implementation of these devices allows for chronic (tested for up to 6 months) wireless optogenetic manipulation of neural circuitry in animals navigating complex natural or home-cage environments, interacting socially, and experiencing other freely moving behaviors.

        <P><B>Abstract</B></P> <P>Conventional implantable electrocardiogram monitors are impenetrable to electromagnetic radiation, posing challenges in imaging (i.e. X-rays). In this study, we demonstrate a radiolucent implantable electrocardiogram (ECG) device using graphene sheets and Parylene substrate which allows bypassing of electromagnetic radiation through the implanted device. Evaluated on a canine model, the graphene electrodes detect signals comparable to those recorded using surface electrodes, presenting a valuable and novel modality for medium and long-term monitoring of cardiac rhythm. When compared with current technology, the new system has the distinct advantages of being flexible and radiolucent, thereby improving patient comfort, and avoiding device artifact and shadowing on clinical radiographic imaging.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

      • Immunologic and Tissue Biocompatibility of Flexible/Stretchable Electronics and Optoelectronics

        <P>Recent development of flexible/stretchable integrated electronic sensors and stimulation systems has the potential to establish an important paradigm for implantable electronic devices, where shapes and mechanical properties are matched to those of biological tissues and organs. Demonstrations of tissue and immune biocompatibility are fundamental requirements for application of such kinds of electronics for long-term use in the body. Here, a comprehensive set of experiments studies biocompatibility on four representative flexible/stretchable device platforms, selected on the basis of their versatility and relevance in clinical usage. The devices include flexible silicon field effect transistors (FETs) on polyimide and stretchable silicon FETs, InGaN light-emitting diodes (LEDs), and AlInGaPAs LEDs, each on low modulus silicone substrates. Direct cytotoxicity measured by exposure of a surrogate fibroblast line and leachable toxicity by minimum essential medium extraction testing reveal that all of these devices are non-cytotoxic. In vivo immunologic and tissue biocompatibility testing in mice indicate no local inflammation or systemic immunologic responses after four weeks of subcutaneous implantation. The results show that these new classes of flexible implantable devices are suitable for introduction into clinical studies as long-term implantable electronics.</P>
