        Specific Chicken Egg Yolk Antibody Improves the Protective Response against <i>Gallibacterium anatis</i> Infection

        <P><I>Gallibacterium anatis</I> is a pathogen associated with peritonitis and salpingitis in chickens and other avian species. Novel safety prevention strategies are urgently needed because of widespread multidrug resistance and antigenic diversity.</P><P><I>Gallibacterium anatis</I> is a pathogen associated with peritonitis and salpingitis in chickens and other avian species. Novel safety prevention strategies are urgently needed because of widespread multidrug resistance and antigenic diversity. The objective of this study was to produce a specific chicken egg yolk antibody and evaluate its protective response against a <I>G. anatis</I> infection model in 4-week-old chicks. Enzyme-linked immunosorbent assays showed that hens immunized with the recombinant N terminus of <I>Gallibacterium</I> toxin A (GtxA-N) had significantly increased antibody titers against GtxA-N in serum and egg yolk IgY. Western blotting showed that IgY antibody had specificity against GtxA-N in the egg yolks of immunized hens. The growth of <I>G. anatis</I> in brain heart infusion (BHI) broth and agar was significantly inhibited by the GtxA-N-specific IgY antibody. The protective effects of the specific IgY antibody were evaluated in <I>G. anatis</I>-infected chicks after intramuscular injection (10 mg/ml). The anti-GtxA-N antibody titers in the sera of <I>G. anatis</I>-challenged chicks following an injection of specific IgY antibody were significantly higher than those of the control and the nonspecific IgY groups, but lower lesion scores for the peritoneum, liver, and duodenum were found after specific IgY antibody treatment. The results from this study suggest that the GtxA-N-specific IgY antibody could potentially improve the protective response against <I>G. anatis</I> infection in chicks.</P>

        Transcriptome Analysis of Antrodia cinnamomea Mycelia from Different Wood Substrates

        Antrodia cinnamomea, an edible and medicinal fungus with significant economic value and application prospects, is rich in terpenoids, benzenoids, lignans, polysaccharides, and benzo- quinone, succinic and maleic derivatives. In this study, the transcriptome of A. cinnamomea cultured on the wood substrates of Cinnamomum glanduliferum (YZM), C. camphora (XZM), and C. kanehirae (NZM) was sequenced using the high-throughput sequencing technology Illumina HiSeq 2000, and the data were assembled by de novo strategy to obtain 78,729 Unigenes with an N50 of 4,463 bp. Compared with public databases, about 11,435, 6,947, and 5,994 Unigenes were annotated to the Non-Redundant (NR), Gene Ontology (GO), and Kyoto Encyclopedia of Genes and Genome (KEGG), respectively. The comprehensive analysis of the mycelium terpene biosynthesis-related genes in A. cinnamomea revealed that the expression of acetyl-CoA acetyltransferase (AACT), acyl-CoA dehydrogenase (MCAD), 3- hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA), mevalonate pyrophosphate decarboxylase (MVD), and isopentenyl diphosphate isomerase (IDI) was significantly higher on NZM com- pared to the other two wood substrates. Similarly, the expression of geranylgeranyltransfer- ase (GGT) was significantly higher on YZM compared to NZM and XZM, and the expression of farnesyl transferase (FTase) was significantly higher on XZM. Furthermore, the expressions of 2,3-oxidized squalene cyclase (OCS), squalene synthase (SQS), and squalene epoxidase (SE) were significantly higher on NZM. Overall, this study provides a potential approach to explore the molecular regulation mechanism of terpenoid biosynthesis in A. cinnamomea.

      • Innovative Approach of Non-Thermal Plasma Application for Improving the Growth Rate in Chickens

        <P>As an innovative technology in biological applications—non-thermal plasma technique—has recently been applied to living cells and tissues. However, it is unclear whether non-thermal plasma treatment can directly regulate the growth and development of livestock. In this study, we exposed four-day-incubated fertilized eggs to plasma at 11.7 kV for 2 min, which was found to be the optimal condition in respect of highest growth rate in chickens. Interestingly, plasma-treated male chickens conspicuously grew faster than females. Plasma treatment regulated the reactive oxygen species homeostasis by controlling the mitochondrial respiratory complex activity and up-regulating the antioxidant defense system. At the same time, growth metabolism was improved due to the increase of growth hormone and insulin-like growth factor 1 and their receptors expression, and the rise of thyroid hormones and adenosine triphosphate levels through the regulation of demethylation levels of growth and hormone biosynthesis-related genes in the skeletal muscles and thyroid glands. To our knowledge, this study was the first to evaluate the effects of a non-thermal plasma treatment on the growth rate of chickens. This safe strategy might be beneficial to the livestock industry.</P>

        Enhancement of transglutaminase production in Streptomyces mobaraensis DSM 40587 by non-nutritional stress conditions: Effects of heat shock, alcohols, and salt treatments

        Stress-mediated bioprocess is a strategy designed to enhance biological target productivity. In this study,an attempt was made to enhance transglutaminase (TGase) production by Streptomyces mobaraensis by using different stress conditions including heat shock, alcohols and salt stress. Results showed that the effects of stress on TGase production depended on the type applied. For heat shock, TGase production (1.32 U/ml) was recorded maximum in the culture treated at 48 h post inoculation in water bath at 60 oC for 1 min. For alcohols treatment, the maximum activity of TGase (1.77 and 1.75 U/ml) was obtained when 3% methanol was added into the medium at 0 or 24 h of fermentation. However, a 3.5-fold increased production of TGase (3.8 U/ml) was observed in the medium supplemented with 0.2mol/l MgCl2 compared with the basic medium at the beginning of fermentation. In conclusion, TGase production from S. mobaraensis was improved by heat shock, methanol and salt stress treatments, MgCl2 stress was the most effective.

      • Prognostic Significance of Circulating Tumor Cells in Small-Cell Lung Cancer Patients: a Meta-analysis

        Circulating tumor cells (CTCs) are believed to be particularly important and a reliable marker of malignancy. However, the prognostic significance of CTCs detected in patients with small cell lung cancer (SCLC) is still unclear. We therefore aimed to assess the prognostic relevance of CTCs using a meta-analysis. We searched PubMed for relevant studies and statistical analyses were conducted to calculate the hazard ratio (HR) and 95% confidence intervals (CIs) using fixed or random-effect models according to the heterogeneity of included studies. A total of 7 papers covering 440 SCLC patients were combined in the final analysis. The meta-analysis revealed that CTCs were significantly associated with shorter overall survival (HR=1.9; 95%CI: 1.19-3.04; Z=2.67; P<0.0001) and progression-free survival (HR=2.6; 95%CI: 1.9-3.54; Z=6.04; P<0.0001). The results thus suggest that the presence of CTCs indicates a poor prognosis in patients with SCLC. Further well-designed prospective studies are required to explore the clinical applications of CTCs in SCLC.

        The article seeks to observe the role of the European Union (EU) in the international rule of law campaign from the perspective of one of the special features of the EU-the ``dual identity`` of the EU on the world stage, i.e. i) the EU as a regional international organisation; ii) the EU as one of the international actors. By observing the EU under the international law theory, clarifying the ``dual identity`` of the EU, reasoning the potential role of the EU, the article concludes that the EU should be a normative power in the international rule of law campaign and it does have the potential to play a greater role because of its special ``dual identity``.

        Permeability-increasing effects of hydraulic flushing based on flow-solid coupling

        Shallow coal resources are increasingly depleted, the mining has entered the deep stage. Due to "High stress, high gas, strong adsorption and low permeability" of coal seam, the gas drainage has become more difficult and the probability of coal and gas outburst accident increases. Based on the flow solid coupling theory of coal seam gas, the coupling model about stress and gas seepage of coal seam was set up by solid module and Darcy module in Comsol Multiphysics. The gas extraction effects were researched after applying hydraulic technology to increase permeability. The results showed that the effective influence radius increases with the expanded borehole radius and drainage time, decreases with initial gas pressure. The relationship between the effective influence radius and various factors presents in the form: $y=a+{\frac{b}{\left(1+{(\frac{x}{x_0})^p}\right)}}$. The effective influence radius with multiple boreholes is obviously larger than that of the single hole. According to the actual coal seam and gas geological conditions, appropriate layout way was selected to achieve the best effect. The field application results are consistent with the simulation results. It is found that the horizontal stress plays a very important role in coal seam drainage effect. The stress distribution change around the drilling hole will lead to the changes in porosity of coal seam, further resulting in permeability evolution and finally gas pressure distribution varies.

      • Optimal design of EMI filters for PV system based on parasitic parameter and stability analysis

        Electromagnetic magnetic interference (EMI) issue has become one of the urgent problems to be solved in the photovoltaic (PV) inverter system. Therefore, it should not be ignored. In order to optimize the EMI filter design, high frequency characteristic of a PV system is studied in this paper. A conducted EMI model is proposed. The proposed model take into consider the influences of PV array parameters, DC side cable impedance, inverter parasitic capacitance and the unbalance of AC side impedance. Based upon that model, a passive EMI filter used for the DC side of a PV system is designed and fabricated. And the designing steps have been given thoroughly. To acquire the best performance of the filter, a stabilization analysis based on Routh criterion is also made. Finally, experimental results are presented to verify the efficient of the proposed EMI filter.
