Changes of Tumor Infiltrating Lymphocytes after Core Needle Biopsy and the Prognostic Implications in Early Stage Breast Cancer: A Retrospective Study

        Jiahui Huang,Xiaosong Chen,Xiaochun Fei,Ou Huang,Jiayi Wu,Li Zhu,Jianrong He,Weiguo Chen,Yafen Li,Kunwei Shen 대한암학회 2019 Cancer Research and Treatment Vol.51 No.4

Purpose The purpose of this study was to investigate the changes of tumor infiltrating lymphocytes (TILs) between core needle biopsy (CNB) and surgery removed sample (SRS) in early stage breast cancer patients and to identify the correlating factors and prognostic significance of TILs changes. Materials and Methods A retrospective study was carried out on 255 patients who received CNB and underwent surgical resection for invasive breast cancer. Stromal TILs levels of CNB and SRS were evaluated respectively. Tumors with  50% stromal TILs were defined as lymphocyte-predominant breast cancer (LPBC). Clinicopathological variables were analyzed to determine whether there were factors associated with TILs changes. Log-rank tests and Cox proportional hazards models were used to analyze the influences of TILs and TILs changes on survival. Results SRS-TILs (median, 10.0%) were significant higher than CNB-TILs (median, 5.0%; p < 0.001). Younger age (< 60 years, p=0.016) and long surgery time interval (STI,  4 days; p=0.003) were independent factors correlating with higher TILs changes. CNB-LPBC patients showed better breast cancer-free interval (BCFI, p=0.021) than CNB-non-LPBC (CNB-nLPBC) patients. Patients were categorized into four groups according to the LPBC change pattern from CNB to SRS: LPBCLPBC, LPBCnLPBC, nLPBCLPBC, and nLPBCnLPBC, with estimated 5-year BCFI 100%, 100%, 69.7%, and 86.0% (p=0.016). nLPBCLPBC pattern was an independent prognostic factor of worse BCFI (hazard ratio, 2.19; 95% confidence interval, 1.06 to 4.53; p=0.035) compared with other patterns. Conclusion TILs were significantly higher in SRS than in CNB. Higher TILs changes were associated with younger age and long STI. Changing from nLPBC to LPBC after CNB indicated a worse BCFI, which needs further validation.

Nanoclay, Intumescent Flame Retardants, and Their Combination with Chemical Modification for the Improvement of the Flame Retardant Properties of Polymer Nanocomposites

        Guobo Huang,Jianguo Yang,Xu Wang,Jianrong Gao 한국고분자학회 2013 Macromolecular Research Vol.21 No.1

The flame retardancy of nanoclay, intumescent flame retardants (IFRs), and their combination with chemical modification is investigated in poly(methyl methacrylate) (PMMA) nanocomposites. 2,4,8,10-Tetraoxa-3,9-diphosphaspiro[5.5]-undecane-3,9-dioxide-disubstitutio-acetamide-N,N,N-triethyl-ammonium bromide (PDEABMMT)and 2,4,8,10-tetraoxa-3,9-diphosphaspiro[5.5]-undecane-3,9-dioxide-disubstitutio-acetamide-N,N-dimethyl-N-hexadecyl-ammonium bromide (PDHAB-MMT) are prepared by ion exchange of sodium montmorillonite with phosphorus-nitrogen containing quarternary ammonium salts including PDEAB and PDHAB. The X-ray diffraction (XRD) and transmission electron microscopy (TEM) results show that PDHAB-MMT achieves tood dispersion in the PMMA matrix and the corresponding nanocomposites PMMA/PDHAB-MMT are formed. The incorporation of PDHAB-MMT into the PMMA matrix obviously improves the nanocomposite thermal properties. Cone calorimeter experiments indicate that the flame retardancy of nanoclay, IFRs, and their combination with chemical modification showed a 5%, 5%, and 9% reduction in the peak heat release rates of PMMA, respectively. The scanning electron microscopy and TEM images confirm that a compact and dense char is formed for PMMA/PDHAB-MMT nanocomposites after combustion.

A novel ceramic GEM used for neutron detection

        Zhou, Jianrong,Zhou, Xiaojuan,Zhou, Jianjin,Jiang, Xingfen,Yang, Jianqing,Zhu, Lin,Yang, Wenqin,Yang, Tao,Xu, Hong,Xia, Yuanguang,Yang, Gui-an,Xie, Yuguang,Huang, Chaoqiang,Hu, Bitao,Sun, Zhijia,Chen, Korean Nuclear Society 2020 Nuclear Engineering and Technology Vol.52 No.6

A novel ceramic Gas Electron Multiplier (GEM) has been developed to meet the demand of high counting rate for the neutron detection which is an alternative to 3He-based detector at China Spallation Neutron Source (CSNS). An experiment was performed to measure the neutron transmittance of ceramic-GEM and FR4-GEM at the small angle neutron scattering (SANS) instrument. The result showed the ceramic-GEM has higher transmittance and less self-scattering especially for cold neutrons. One single ceramic GEM could give a gain of 102-104 in the mixture gas of Ar and CO2 (90%:10%) and its energy resolution was about 27.7% by using 55Fe X ray of 5.9 keV. A prototype has been developed in order to investigate the performances of the ceramic GEM-based neutron detector. Several neutron beam tests, including detection efficiency, spatial resolution, two-dimensional imaging, and wavelength spectrum, were carried out at CSNS and China Mianyang Research Reactor (CMRR). The results show that the ceramic GEM-based neutron detector is a good candidate to measure the high intensity neutrons.

Inhibition of quorum sensing, biofilm, and spoilage potential in Shewanella baltica by green tea polyphenols

        Junli Zhu,Xuzheng Huang,Fang Zhang,Lifang Feng,Jianrong Li 한국미생물학회 2015 The journal of microbiology Vol.53 No.12

We investigated the quorum sensing (QS) system of Shewanella baltica and the anti-QS related activities of green tea polyphenols (TP) against spoilage bacteria in refrigerated large yellow croaker. Autoinducer-2 (AI-2) and the diketopiperazines (DKPs) cyclo-(L-Pro-L-Leu) and cyclo-(L-Pro-L-Phe) were detected in the culture extract of S. baltica XH2, however, no N-acylhomoserine lactones (AHLs) activity was observed. Green TP at sub-inhibitory concentrations interfered with AI-2 and DKPs activities of S. baltica without inhibiting cell growth and promoted degradation of AI-2. The green TP treatment inhibited biofilm development, exopolysaccharide production and swimming motility of S. baltica in a concentration- dependent manner. In addition, green TP decreased extracellular protease activities and trimethylamine production in S. baltica. A transcriptional analysis showed that green TP repressed the luxS and torA genes in S. baltica, which agreed with the observed reductions in QS activity and the spoilage phenotype. Epigallocatechin gallate (EGCG)-enriched in green TP significantly inhibited AI-2 activity of S.baltica. These findings strongly suggest that green TP could be developed as a new QS inhibitor for seafood preservation to enhance shelf life.

De novo assembly, characterization and annotation for the transcriptome of Sphaeroma terebrans and microsatellite marker discovery

        Chong Han,Qiang Li,Xiufeng Li,Zhipeng Zhang,Jianrong Huang 한국유전학회 2018 Genes & Genomics Vol.40 No.2

Sphaeroma terebrans, an economically and ecologically important marine wood-boring isopod, is mainly distributed in tropical and subtropical mangroves. Nevertheless, available genomic and transcriptomic information for this isopod is extremely deficient. Here, we first performed the assembly of S. terebrans transcriptome by Illumina sequencing. A total of 51,092 high-quality unigenes with an average length of 641 bp were obtained and 19,915 unigenes were successfully annotated in four public databases. Only 9932 out of 19915 unigenes were commonly annotated by all four databases. In addition, 9609 unigenes were categorized into 54 function categories of Gene Ontology (GO), and 14,512 unigenes were successfully grouped into 25 functional categories of the EuKaryotic Orthologous Groups (KOG) database. Moreover, 11,507 unigenes were assigned to 228 pathways of the Kyoto Encyclopedia of Genes and Genomes (KEGG). Out of 51,092 unigenes, a total of 4257 different microsatellites with motifs range from di- to hexanucleotide were identified from 3324 unigene sequences. Among 64 primer pairs selected for validation, 35 were successful in PCR amplification and 13 exhibited obvious repeat polymorphisms in the wild population of S. terebrans in Dongzhaigang (Hainan Island). The transcriptome dataset and the identified microsatellite markers (SSRs) will provide abundant information for researches on the discovery of new genes, metabolic mechanism and genetic diversity of S. terebrans.

Comparison of the Distribution Pattern of 21-Gene Recurrence Score between Mucinous Breast Cancer and Infiltrating Ductal Carcinoma in Chinese Population: A Retrospective Single-Center Study

        Jiayi Wu,Shuning Ding,Linling Yin,Xiaochun Fei,Caijin Lin,Lisa Andriani,Chihwan Goh,Jiahui Huang,Jin Hong,Weiqi Gao,Siji Zhu,Hui Wang,Ou Huang,Xiaosong Chen,Jianrong He,Yafen Li,Kunwei Shen,Weiguo Che 대한암학회 2020 Cancer Research and Treatment Vol.52 No.3

Purpose This retrospective study aimed to evaluate the distribution pattern and prognostic value of 21-gene recurrence score (RS) in Chinese patients with mucinous breast cancer (MC) and compared with infiltrating ductal carcinoma (IDC). Materials and Methods Patients diagnosed with MC or IDC from January 2010 to January 2017 were retrospectively recruited. Reverse transcriptase–polymerase chain reaction assay of 21 genes was conducted to calculate the RS. Univariate and multivariate analyses were performed to assess the association between RS and clinicopathological factors. Survival outcomes including disease-free survival (DFS) and overall survival (OS) were estimated by Kaplan-Meier method and compared by log-rank test. Results The MC cohort included 128 patients and the IDC cohort included 707 patients. The proportions of patients with a low (RS < 18), intermediate (18-30), or high risk (RS > 30) were 32.0%, 48.4%, and 19.5% in MC cohort, and 26.9%, 46.8% and 26.3% in IDC cohort. The distribution of RS varied significantly according to different Ki-67 index and molecular subtype in both cohorts. Moreover, the receipt of chemotherapy was associated with RS in both cohorts. Among patients with MC, tumor stage was related to the DFS (p=0.040). No significant differences in DFS and OS were found among MC patients in different RS risk groups (OS, p=0.695; DFS, p=0.926). Conclusion RS was significantly related to Ki-67 index and molecular subtypes in MC patients, which is similar in IDC patients. However, RS was not able to predict DFS and OS in patients with MC.
