Spry genes encode cytoplasmic membrane-associated proteins and that Spry4 (sprouty RTK signaling antagonist 4) proteins have been shown to negatively regulate a variety of receptor tyrosine kinase (RTK) signaling pathways by specific interference with the Ras/MAPK pathway. The MAPK signalling pathway has been intensively studied by cancer biologists because of its effects on regulation of cell proliferation and survival1,2, but this pathway is also important throughout mouse embryogenesis3. The pathway has been investigated in numerous embryonic processes. Here, we performed a broad-based basic phenotyping for Spry4 mutant mice. Spry4 KO mice showed abnormal phenotype in various areas, such as growth rate, grip strength, IPGTT and X-ray observations, indicating a condition characterized by growth retardation along with bone abnormalities. Further studies should be performed to understand the underlying mechanisms for this phenotype.
Rxfp2, an Insl3 receptor might be associated with the failure of spermatogenesis in Phf7 ko mice

PHD finger protein 7 (Phf7) is a member of the PHF family, which contain the PHD domain known as a histone methylation reader and RING domain known as a E3 ubiquitin ligase. Phf7 is expressed testis, specifically in germ cells and Leydig cells but not in Sertoli cells. The deficiency of Phf7 induces male infertility. And it was found that Phf7 acts as a histone H2A E3 ligase in histone-to-protamine exchange during spermatogenesis. Until now, most studies on Phf7 have been focused on spermatogenesis. Here, we tried to found any phenotypes other than male infertility in the Phf7 Ko mice. We found that there are growth-retardation and reduction of bone mineral density in Phf7 knockout (ko) mice. We also tried to found any mechanism that may associated with abnormal spermatogenesis in the Phf7 ko mice. It is known that Rxfp2 is expressed in the postmeiotic germ cells in testis and that Rxfp2 with Insl3, a hormone released from Leydig cells constitute a paracrine hormone-receptor system in testis. Therefore, we determined the expression level of Insl3 and Rxfp2 in testis of Phf7 ko mice. We found that the Insl3 mRNA level was higher in the ko mice, but the Rxfp2 mRNA expression level was reduced in the mutant mice then wild mice. In conclusion, we suggest that the failure of spermatogenesis observed in Phf7 ko mice might related with the reduced expression of Rxfp2 in the developing germ cells.
The sex and age dependant differences in ECG values in C57BL6/N mice

Eelectrocardiogram (ECG) is a surface recording of the electric current passing through the heart, revealing the electrical activity associated with each heartbeat, and allowing visualization of the electrical state of the heart. In a state of consciousness, normal rat heart rates range from 500 to 700 beats per minute (bpm). ECG reflects the cycle of the heart and consists of P, Q, R, S and T waves. The non-invasive ECG system has the advantage of acquiring information not only on heart rate, but also on heart rate variability, RR, QT and PR intervals, as well as acquiring cardiac abnormalities in conscious animals. In this study, we conducted experiments ECG analysis with 12 weeks and 56 weeks aged male and female C57BL/6N mice. We compared the ECG values between male and female mice and between 12 weeks and 56 weeks aged mice, comparing age and gender differences. There were sex differences in HR and RR in 12 weeks aged mice and in HR and QRS in 56 weeks aged mice. We also found age dependant differences in QRS, ST, HR and RR in male mice and in PR, ST and QTc in female mice. These result indicate that there is age and sex differences in ECG values.
