        USP18 alleviates neurotoxicity induced by sevoflurane via AKT and NF-κB pathways

        Background Sevoflurane has obvious side effects during anesthesia, which caused neuronal apoptosis and neuroinflammation. How to reduce Sevoflurane-induced neurotoxicity is an urgent problem to be solved. Objective To assess the role of USP18 in anesthetic Sevoflurane-induced neurotoxicity, detect its effects on neuroinflammation, oxidative stress, and apoptosis, and explore the related potential signaling pathway. Results USP18 alleviated Sevoflurane-induced learning and memory dysfunction and changed distribution of neurons. USP18 also reduced Sevoflurane-induced neuroinflammation. We further found that USP18 reduced the Sevoflurane-induced oxidative stress in the mice model. USP18 also attenuated Sevoflurane-induced neuronal apoptosis. Mechanically, USP18 reduced Sevoflurane-induced neurotoxicity via AKT and NF-κB pathway. Conclusion USP18 was involved in the pathology of Sevoflurane-induced neurotoxicity.

      • Croton Tiglium Extract Induces Apoptosis via Bax/Bcl-2 Pathways in Human Lung Cancer A549 Cells

        Objective: To investigate the impact of a Croton tiglium extract on cellular proliferation and apoptosis in a non-small cell lung cancer cell line (A549) in vitro. Methods: A Croton tiglium seed methanol extract was prepare and assessed for effects on A549 cells regarding cellular proliferation, apoptotic rates, and expression of apoptosis related genes and proteins using real-time PCR and immunofluorescence. Results: The tested Croton tiglium extract inhibited A549 cell proliferation in a dose- and time-dependent manner, with significant elevation of apoptotic indexes at various concentrations after 24 h. In addition, rates in both early and late stages were higher in treated than untreated groups, the $100{\mu}g/ml$ dose causing the highest levels of apoptosis. RT-PCR showed that A549 cells treated with $100{\mu}g/ml$ Croton tiglium extract for 24 h has markedly higher Bax mRNA expression levels and obviously lower Bcl-2 expression levels than controls, equivalent results being observed for proteins by immunofluorescence. However, the mRNA expression levels of Fas and caspase-8 were not significantly altered. Conclusion: A Croton tiglium extract can inhibit proliferation of A549 cells and promote apoptosis though Bax/Bcl-2 pathways.

        Determination of the energy contents and nutrient digestibility of corn, waxy corn and steam-flaked corn fed to growing pigs

        Objective: The research was conducted to determine the digestible (DE) and metabolizable energy (ME) contents as well as the apparent total tract digestibility (ATTD) of nutrients in corn, waxy corn and steam-flaked corn fed to growing pigs. Methods: Eighteen growing pigs with initial body weight of 15.42±1.41 kg were randomly allotted to three diets including a corn diet, a waxy corn diet and a steam-flaked corn diet in a completely randomized design. Each treatment contained six replicates. The experiment lasted for 12 days, which comprised 7-d adaptation to diets followed by a 5-d total collection of feces and urine. The energy contents and the nutrient digestibility in three ingredients were calculated using direct method. Results: Compared to normal corn, both the amylose and dietary fiber contents in waxy corn were numerically lower, but the starch gelatinization degree was numerically greater. Moreover, the DE and ME contents as well as the ATTD of neutral detergent fiber and acid detergent fiber (ADF) in waxy corn were significantly greater (p<0.05) than those in normal corn when fed to growing pigs. Furthermore, the steam-flaked corn had greater (p<0.05) DE and ME contents, and ATTD of ether extract and ADF compared to normal corn. Conclusion: Both variety and processing procedure have influence on chemical compositions, energy contents and nutrient digestibility of corn. The waxy corn and steam-flaked corn had greater degree of starch gelatinization and DE and ME contents compared to normal corn when fed to growing pigs.

        Electrochemical Exfoliation of Graphene Flake Embedded in SiNWs as Counter Electrode for Dye-Sensitized Solar Cells

        In this paper, a three-dimensional graphene wrapped silicon nanowire (SiNWs) architecture was synthesized by electrochemical exfoliation method and proposed to be the counter electrode for dye-sensitized solar cells (DSSCs). The results show that the few high-quality layers of graphene sheets have been obtained by electrochemical exfoliation method. The size of graphene is distributed from 550 nm to 650 nm, appropriately dispersed onto the SiNW supporter. The graphene/SiNW nanostructure still vertically aligned to the substrate finely. The channel between each wire is clear. The fabricated graphene/SiNWs electrode is a promising structure for future applications. The performance of the graphene/SiNWs as the counter electrode is found to be dependent on its dispersion in the whole backbone, with better dispersion offering more surface areas for the catalytic reduction reaction. Electrochemical tests reveal that the DSSC with graphene/SiNWs exhibits higher electro-catalytic activity and lower charge transfer resistance, suggesting that the 3D structure presents a potential way to fabricate low-cost, integrated and metal-free counter electrode for high-performance DSSCs. The DSSC based on graphene/SiNWs has an open-circuit voltage (Voc) of 738.11 mV, short-circuit current density (Jsc) of 15.48mA cm 2, fill factor (FF) of 0.67 and conversion efficiency (η) of 7.66%.

        Cerebrosides from Sea Cucumber Protect Against Oxidative Stress in SAMP8 Mice and PC12 Cells

        Alzheimer's disease (AD) is a neurodegenerative disorder. Emerging evidence implicates β-amyloid (Aβ) plays a critical role in the progression of AD. In this study, we investigated the protective effect of cerebrosides obtained from sea cucumber against senescence-accelerated mouse prone 8 (SAMP8) mice in vivo. We also studied the effect of cerebrosides on Aβ-induced cytotoxicity on the rat pheochromocytoma cell (PC12) and the underlying molecular mechanisms. Cerebrosides ameliorated learning and memory deficits and the Aβ accumulation in demented mice, decreased the content of malondialdehyde (MDA), 8-oxo-7,8-dihydro-2′-deoxyguanosine (8-OHdG), 8-hydroxy-2′-deoxyguanosine (8-oxo-G), and nitric oxide (NO), and enhanced the superoxide dismutase (SOD) activity significantly. The neuroprotective effect of sea cucumber cerebrosides (SCC) was also verified in vitro: the cerebrosides increased the survival rate of PC12 cells, recovered the cellular morphology, downregulated the protein levels of Caspase-9, cleaved Caspase-3, total Caspase-3, and Bax, and upregulated the protein level of Bcl-2, revealing that cerebrosides could inhibit Aβ-induced cell apoptosis. The results showed the protective effect of SCC was regulated by the mitochondria-dependent apoptotic pathway. Our results provide a new approach to developing the marine organisms as functional foods for neuroprotection.
