        A new whitefly of quarantine importance infesting a native and invasive plant, Rubus ellipticus Sm. (Rosaceae) in the Western Himalaya, India

        Puparia of Pealius kufriensis Dubey sp. nov. found infesting a native and highly invasive plant, Rubus ellipticus Sm. (Rosaceae) are described. The host shrub was found highly infested with the new whitefly on the underside of leaves. Puparial characteristics of the genus Pealius Quaintance and Baker shared with Aleyrodes Latreille and Bemisia Quaintance and Baker are discussed.

        Smart Phone Applications as a Source of Information on Stroke

        Background and Purpose: Smartphone applications have been increasingly identified as a novel platform for dissemination of healthcare related information. However, there have been no studies done to evaluate the availability and content of stroke related apps. Purpose: This study aims to identify and analyze stroke-related applications available on the Apple iTunes and Android Google Play Store. Methods: The Apple iTunes store and Android Google Play Store were searched for stroke-related applications on July 27, 2013 using keywords: stroke, brain attack, intracranial hem-orrhage, subarachnoid hemorrhage, cerebral infarction. The content of the applications was analyzed by two independent investigators. Results: A total of 93 relevant applications (46.2% android and 53.8% iPhone) were identified of which 47.3% were available free of cost. 92% of apps were identified as useful by users and over 60% had scientifically valid information. There is a significant participation of healthcare agencies in dissemination of stroke related information through apps with 47.3% apps being uploaded by them. Over half of all stroke related apps were aimed towards health care workers (51.6%), 75% of which could be utilized as bedside tools for patient care and remainder had information related to recent research advances. The difference in scientific validity between the apps aimed at general population versus healthcare professionals was statistically significant (P<0.01). There was no statistical association between cost of app and scientific validity or usefulness. Conclusions: Smartphone apps are a significant source of information related to stroke. An increasing participation of healthcare agencies should be encouraged to promote dissemination of scientifically valid information.

      • Genotypic and phenotypic characterization of <i>Edwardsiella</i> isolates from different fish species and geographical areas in Asia: Implications for vaccine development

        <P> The genus <I>Edwardsiella</I> is one of the major causes of fish diseases globally. Herein, we examined 37 isolates from ten different fish species from India, South Korea and Taiwan to gain insight into their phenotypic and genotypic properties, of which 30 were characterized as <I>E. tarda</I> with phenotypic homology estimated at 85.71% based on API-20E biochemical tests. Genotyping using 16S rRNA put all isolates together with <I>E. anguillarum, E. hoshinae, E. tarda, E. piscicida and E. ictaluri</I> reference strains in a monophyletic group. In contrast, the gyrB phylogenetic tree clearly separated <I>E. ictaluri, E. tarda and E. hoshinae</I> reference strains from our isolates and put our isolates into two groups with group I being homologous with the <I>E. anguillarum</I> reference strain while group II was homologous with the <I>E. piscicida</I> reference strain. Hence, our findings point to <I>E. piscicida</I> and <I>E. anguillarum</I> as species infecting different fish species in Asia. Homology of the ompW protein suggested that strains with broad protective coverage could be identified as vaccine candidates. This study underscores the importance of combining genotyping with phenotyping for valid species classification. In addition, it accentuates the importance of phylogenetic comparison of bacterial antigens for identification of potential vaccine candidates. </P>

        Microarray-Mediated Transcriptome Analysis of the Tributyltin (TBT)-Resistant Bacterium Pseudomonas aeruginosa 25W in the Presence of TBT

        The tributyltin (TBT)-resistant bacterium, Pseudomonas aeruginosa 25W, which was isolated in seawater from the Arabian Sea, was subjected to transcriptome analysis in the presence of high concentrations of TBT. Only slight effects were observed at TBT concentration of $50{\mu}M$, but exposure to $500{\mu}M$ resulted in the upregulation of 6 genes and the downregulation of 75. Among the 75 downregulated genes, 53% (40 out of 75) were of hypothetical function, followed by 14 transcriptional regulation- and translation-associated genes. The results of this study indicated that although the 25W strain was highly resistant to TBT, high concentrations of TBT result in toxic effect on the transcriptional and translational levels. The target genes likely belong to a specific category of transcription- and translation-associated genes rather than to other gene categories.

        Identification of Nandrolone and its Metabolite 5α-Estran-3β, 17α-Diol in Horse Urine after Chemical Derivatization by Liquid Chromatography Tandem Mass Spectrometry

        Androgenic anabolic steroids (AASs) are synthetic derivatives of testosterone with a common structure containing cyclopentanoperhydrophenanthrene nucleus. Their use enhances the muscle building capacity and is beneficial during performance. The AASs are one of the most abused group of substances in horse doping. Liquid chromatography tandem mass spectrometry ($LC/MS^n$) has been successfully applied to the detection of anabolic steroids in biological samples. However, the saturated hydroxysteroids viz: nandrolone, $5{\alpha}-estrane-3{\beta}$, $17{\alpha}-diol$ exhibit lower detection responses in electrospray ionisation (ESI) because of their poor ionisation efficiency. To overcome this limitation pre-column chemical derivatization has been introduced to enhance their detection responses in $LC-ESI-MS^n$ analysis. The aim of present study was to develop a sensitive method for identification and confirmation of nandrolone and its metabolite in horse urine incorporating pre-column derivatization using picolinic acid. The method consists of extraction of targeted steroid conjugates by solid phase extraction (SPE). The eluted steroid conjugates were hydrolysed by methanolysis and free steroids were recovered with liquid-liquid extraction. The resulting steroids were derivatized to form picolinoyl esters and identification was done using LC-ESI-MS/MS in positive ionization mode. The picolinated steroid adduct enhanced the detection levels in comparison to underivatized steroids.

        Self-assembly of new arene-ruthenium rectangles containing triptycene building block and their application in fluorescent detection of nitro aromatics

        A suite of two new tetraruthenium metallarectangles 5 and 6 have been obtained from [2+2] self-assemblies between dipyridylethynyltriptycene 2 and one of the two dinuclear arene ruthenium clips, [Ru<SUB>2</SUB> (μ-η<SUP>4</SUP>-OO@?OO) (η<SUP>6</SUP>-p-cymene)<SUB>2</SUB>][OTf]<SUB>2</SUB>; (OO@?OO=oxalate 3; 6,11-dihydroxy-5,12-naphthacenedionato (dotq) 4; OTf=triflate). These molecular rectangles are fully characterized by <SUP>1</SUP>H NMR spectroscopy, electrospray mass spectrometry. A single crystal of 6 was suitable for X-ray diffraction structural characterization. These new metallarectangles showed fluorescence behavior in solution, have been examined for emission quenching effects with various aromatic compounds, and show high quenching selectivity and sensitivity towards nitroaromatics, particularly picric acid and 1,3,5-trinitrobenzene. Excited-state charge transfer from the rectangles to nitro aromatic substrates can be used to develop selective fluorescent sensors for nitro aromatics.

        A NEUMANN TYPE PROBLEM ON AN UNBOUNDED DOMAIN IN THE HEISENBERG GROUP

        We discuss the wellposedness of the Neumann problem on a half-space for the Kohn-Laplacian in the Heisenberg group. We then construct the Neumann function and explicitly represent the solution of the associated inhomogeneous problem.

      • Agroinfiltration-based expression of hairpin RNA in soybean plants for RNA interference against <i>Tetranychus urticae</i>

        <P>The coatomer subunit alpha (COPA) and aquaporin 9 (AQ9) genes from the two-spotted spider mite, Tetranychus urticae, were previously determined to exhibit RNA interference (RNAi)-based lethality when their double stranded RNAs were systemically delivered via multi-unit chambers (Kwon et al., 2016 [8]). In current study, the hairpin RNAs of the COPA and AQ9 were transiently expressed in soybean plants by agroinfiltration. When T. urticae was fed with the soybean plants agroinfiltrated with the COPA and AQ9 hairpin RNA cassettes, the cumulative mortality increased significantly at 6 days post-infestation. Quantitative PCR analysis revealed that the transcript level of both COPA and AQ9 was significantly reduced in T. urticae after 2 days post-infestation, thereby confirming that the significant increases in mortality resulted from the knockdown of COPA and AQ9 transcripts. Our findings demonstrate the utility of COPA and AQ9 as potential genes for plant host-mediated RNAi control of T. urticae. In addition, we proved the usefulness of agroinfiltration as a rapid validation tool for confirming the RNAi-based lethality of target genes against arthropod pests before producing transgenic plants as agroinfiltration requires less time and skill to validate transgene function. Furthermore, these findings prove the concept that hairpin RNA expressed in plant hosts can also induce RNAi and eventually kill T. urticae, a sap-sucking pest. (C) 2017 Elsevier Inc.       • Low energy ultrasonic single beacon localization for testing of scaled model vehicle

        Tracking the location (position) of a surface or underwater marine vehicle is important as part of guidance and navigation. While the Global Positioning System (GPS) works well in an open sea environment but its use is limited whenever testing scaled-down models of such vehicles in the laboratory environment. This paper presents the design, development and implementation of a low energy ultrasonic augmented single beacon-based localization technique suitable for such requirements. The strategy consists of applying Extended Kalman Filter (EKF) to achieve location tracking from basic dynamic distance measurements of the moving model from a fixed beacon, while on-board motion sensor measures heading angle and velocity. Iterative application of the Extended Kalman Filter yields x and y co-ordinate positions of the moving model. Tests performed on a free-running ship model in a wave basin facility of dimension 30 m by 30 m by 3 m water depth validate the proposed model. The test results show quick convergence with an error of few centimeters in the estimated position of the ship model. The proposed technique has application in the real field scenario by replacing the ultrasonic sensor with industrial grade long range acoustic modem. As compared with the existing systems such as LBL, SBL, USBL and others localization techniques, the proposed technique can save deployment cost and also cut the cost on number of acoustic modems involved.
