Biological characterization of mulberry leaves bioconverted with Viscozyme L

Background Mulberry ( Morus alba L.) leaves is a Korean medicinal herb that shows bioactive compounds such as quercetin, anthocyanins and polysaccharides. Bioconversion with Viscozyme L process could be producing enriched physiologically active compounds of mulberry leaves. Objective Thus, this study focused on the purifi cation of the bioconverted compounds responsible for ethyl acetate extraction and column chromatography using normal-phase silica gel and Sephadex LH-20. Results In this study, we obtained the purifi ed compounds, MAV 1, MAV 2 and MAV 3, which were identifi ed as caff eic acid, quercetin, and kaempferol, by analysis of nuclear magnetic resonance spectroscopy. Also, we optimized the yield of the most active compound, quercetin in 20% substrate, 5% enzyme, at 37 °C and pH 4 for 24 h, 2.34 ± 0.03 mg of quercetin was produced from 1 g of dried mulberry leaves. Furthermore, we showed that bioconverted compounds have biological activities such as α-glucosidase, acetylcholinesterase inhibitory, cytotoxicity activity, tyrosinase, xanthine oxidase (XO) and pancreatic lipase inhibition. Conclusion These results indicate that the enhanced activities of α-glucosidase, tyrosinase, xanthine oxidase inhibitory activities, and the anti-oxidant properties bioconverted from mulberry leaves with Viscozyme L. One of them, the quercetin which is a potent α-glucosidase inhibitor bioconverted from mulberry leaves with Viscozyme L has an anti-diabetic eff ect. And it can be used for the development of health foods and pharmaceutical products.

We investigated the probiotic characteristics and anti-obesity effect of Lactiplantibacillus plantarum MGEL20154, a strain that possesses excellent intestinal adhesion and viability. The in vitro properties, e.g., gastrointestinal (GI) resistance, adhesion, and enzyme activity, demonstrated that MGEL20154 is a potential probiotic candidate. Oral administration of MGEL20154 to diet-induced obese C57BL/6J mice for 8 weeks resulted in a feed efficacy decrease by 44.7% compared to that of the high-fat diet (HFD) group. The reduction rate of weight gain was about 48.5% in the HFD+MGEL20154 group compared to that of the HFD group after 8 weeks, and the epididymal fat pad was also reduced in size by 25.2%. In addition, the upregulation of the zo-1, pparα, and erk2, and downregulation of the nf-κb and glut2 genes in Caco-2 cells by MGEL20154 were observed. Therefore, we propose that the anti-obesity effect of the strain is exerted by inhibiting carbohydrate absorption and regulating gene expression in the intestine.
학동전 정상 소아에서 연령별 혈청 총 IgE 집먼지진드기 특이 IgE 및 ECP 치에 관한 전국적 조사 연구

목 적 : 혈청 총 IgE와 알레르겐 특이 IgE는 알레르기 질환의 진단을 위한 초기검사 방법의 하나로서 널리 이용되고 있으며, 전 세계적으로 가장 주요한 알레르겐인 집먼지진드기(Dermatophagoides pteronyssinus : Dp, D.farinae :Df)는 조기 노출 및 감작에 의하여 천식, 비염 및 아토피 피부염의 발병 가능성을 높여 주고 있기 때문에 자주 검사의 대상이 되고 있다. 한편, 알레르기 염증에 관여하는 호산구의 생산물인 eosinophil cationic protein(ECP)는 기관지 상피세포 손상, 기관지 염종 및 기관지 과민반응을 유발하는 것으로 밝혀진 후 혈청내 ECP 측정의 임상 응용이 활발하다. 본 연구는 학동기전 정상 소아에서 혈청 총 IgE 및 집먼지진드기 특이 IgE 및 ECP의 국내 정상참고치를 제시함으로써 알레르기질환의 진단에 필요한 기본적인 자료를 설정하는데 도움이 되고자 하였다. 방 법 : 1996년 5월부터 1997년 8월까지 서울, 경기, 강원, 경북, 전남 등 진국 5개 지역에서 무작위로 선정되고 그 지역을 대표할 수 있는 9개 병원의 육아상담실에 내원한 그리고 보육기관에서 거주하면서 발열이나 상기도 감염 및 알레르기질환의 과거력과 가족력이 없는 7세 미만의 건강한 소아 449명을 대상으로 하였다. 이들로 부터 각각 말초 정맥혈을 채취 후, 원심분리하여 -20℃에서 냉동 보관 후 혈청 총 IgE, Dp-lgE, Df-IgE 및 ECP치를 fluoroimmunoassay(UniCAP, Pharmacia-Upjohn, Sweden)로 측정하고, 그 각각의 결과에서 기하평균과 신뢰구간을 구하여 분석 평가하였다. 결 과 : 1) 연령별 혈청 충 IgE는 출생 후 4새까지 서서히 증가하다가 이후에 완만한 커브를 나타내었다. 2) 혈청 집먼지진드기 특이 IgE(Dp-IgE와 Df-IgE) 치는 3세 이후 특히 남아에서 나이가 들면서 학동기까지 각각 서서히 의미있게 증가하였다. 3) 혈청 ECP치는 출생 후 4세까지 점차적으로 증가한 후 감소하는 추세를 보였다. 4) 혈청 총 IgE, Dp-IgE와 Df-IgE 및 ECP치는 남아와 여아 사이에 통계학적으로 의미있는 차이는 없었다. 5) 혈청 총 IgE, Dp-IgE와 Df-IgE 및 ECP치의 지역별 차이를 보면 Dp-IgE와 Df-IgE가 도시 지역에서 해안에 가까운 기타 도시 지역보다 다소 높았으나, 통계학적으로 유의한 차이는 없었다. 6) 혈청 총 IgE, Dp-IgE와 Df-IgE 및 ECP치 사이의 상관성을 보면 총 IgE와 Dp-IgE(r=0.321, P<0.001) 및 Df-IgE(r=0.324, P<0.001) 그리고 Dp-IgE와 Df-IgE(r=0.943, P<0.001) 사이에 각각 양의 상관관계가 있었으나, ECP치와는 유의한 상관성이 없었다. 결 론 : 혈청 총 IgE 및 ECP치는 4세 전후에 그리고 집먼지진드기 특이 IgE는 3세 이후에 증가를 시작하는 것으로 미루어 조기 감작의 예방이 필요하며, 본 연구 결과는 학동전 소아의 정상 참고치로서 이용 가능할 것으로 사료된다. Purpuse : Serum IgE have been shown to be related to allergic disease and used for an initial diagnosis of allergic diseases. House dust mite such as Dermatophagoides pteronyssinus(Dp) and D. farinae(Df) is very important for inhalant allergens, which may be a cause and/or triggering factor of atopic diseases. On the other hand, eosinophil cationic protein (ECP) causes epithelial damages of the airway, and bronchial hyperresponsiveness has been used as a useful indication of allergic inflammation. To date, there are few studies on the longitudinal patterns of total and inhalant allergen-specific IgE, and ECP in normal children in Korea. The present study was performed to determine the reference levels of serum total and Dp- and Df-specific IgE and ECP in healthy children under 7 years of age by the groups of different age and gender. Methods : Total, and Dp- and Df-specific IgE and ECP concentrations were measured by fluoroimmunoassay(UniCAP, Pharmacia-UpJohn, Sweden) in serum from 449 healthy children under 7 years of age by using a population-based cohort followed from 1996 through 1997. All subjects were enrolled from eight large health organizations in geographically defined area such as Seoul and four local cities. Results : Serum total IgE concentration showed steady increase with age from birth to 4 years, and a plateau. In contrast, Dp- and Df-IgE levels showed apparently continuous increments after 4 years of age. ECP showed variable increase until 4 years of age and slow decrease afterwards. There were no statistically significant differences in those results by gender and/or the living area, but much higher increments of Dp- and Df-IgE of boys from 3 years of age through 6 years were found. Conclusion : We found feasible results of total, Dp- and Df-specific IgE, and ECP in serum from healthy children under seven years of age, which may be useful for a reference.
A Case of Systemic Amyloidosis Found by Goiter in Crohn’s Disease

Secondary amyloidosis is characterized by accumulation of insoluble, fibrous amyloid proteins in various tissues and organs, accompanied by infectious or inflammatory diseases. Amyloidosis may involve the thyroid, gastrointestinal tract, kidneys, liver, or bone marrow. Amyloidosis as a complication of Crohn’s disease is rare but serious, and may worsen the prognosis. We have experienced a case of amyloid goiter and gastrointestinal amyloidosis secondary to Crohn’s disease. A 74-year-old female patient with Crohn’s disease was admitted to Soonchunhyang University Hospital with general weakness and poor oral intake. Anterior-neck diffuse goiter and tenderness around the navel were found. Amyloid goiter and gastrointestinal amyloidosis diagnosed by sonoguided needle biopsy of the thyroid and endoscopic biopsies of the stomach and duodenum.

Probiotics consumption has become a necessity in diet to metabolic syndrome. In this study, in vitro effects of anti-metabolic syndrome were confirmed on the probiotic strain isolated from feces. Each of the strain upregulated the mRNA expressions on zo-1, claudin-1 and occludin related to intestinal tight junction in human epithelial colorectal cell. In particular, the lactiplantibacillus treatment groups significantly upregulated the erk1/2 and map expression related to cell proliferation. In addition, the strain showed that upregulation of ppar-α related to fatty acid oxidation and downregulation of glut-2 related to glucose uptake. Based on the results of this study, we are conducting in vivo experiments to verify the efficacy of the probiotics for alleviating metabolic syndrome.
