        Synthesis of a Novel Near-Infrared Fluorescent Dye: Applications for Fluorescence Imaging in Living Cells and Animals

        Fluorescence imaging is considered as one of the most powerful techniques for monitoring biomolecule activities in living systems. Near-infrared (NIR) light is advantageous for minimum photodamage, deep tissue penetration, and minimum background autofluorescence interference. Herein, we have developed a new NIR fluorescent dye, namely, RB-1, based on the Rhodamine B scaffold. RB-1 exhibits excellent photophysical properties including large absorption extinction coefficients, high fluorescence quantum yields, and high photostability. In particular, RB-1 displays both absorption and emission in the NIR region of the "biological window" (650-900 nm) for imaging in biological samples. RB-1 shows absorption maximum at 614 nm (500-725 nm) and emission maximum at 712 nm (650-825 nm) in ethanol, which is superior to those of traditional rhodamine B in the selected spectral region. Furthermore, applications of RB-1 for fluorescence imaging in living cells and small animals were investigated using confocal fluorescence microscopy and in vivo imaging system with a high signal-to-noise ratio (SNR = 10.1).

        Structural configurations and dynamic performances of flexible riser with distributed buoyancy modules based on FEM simulations

        Flexible risers are usually used as conveying systems to bring ocean resources from sea bed up to onshore. Under ocean environments, risers need to bear complex loads and it is crucial to comprehensively examine riser's configurations and to analyze structural dynamic performances under excitation of bottom vehicle motions, to guarantee structural safe operation and required service lives. In this study, considering a saddle-shaped riser, the influences of some important design parameters, including installation position of buoyancy modules, buoyancy ratio and motion of mining vehicle, on riser's configuration and response are carefully examined. Through our FEM simulations, the spatial distributions of structural tensions and curvatures along of riser length, under different configurations, are compared. Then, the impacts of mining vehicle motion on riser dynamic response are discussed, and structural tolerance performance is assessed. The results show that modules installation position and buoyancy ratio have significant impacts on riser configurations. And, an appropriate riser configuration is obtained through comprehensive analysis on the modules positions and buoyancy ratios. Under this proposed configuration, the structural tension and curvature could moderately change with buoyancy modules and bottom-end conditions, in other words, the proposed saddle-shaped riser has a good tolerance performance to various load excitations.

        <P>The ability to tune material properties using gating by electric fields is at the heart of modern electronic technology. It is also a driving force behind recent advances in two-dimensional systems, such as the observation of gate electric-field-induced superconductivity and metal-insulator transitions. Here, we describe an ionic field-effect transistor (termed an iFET), in which gate-controlled Li ion intercalation modulates the material properties of layered crystals of 1T-TaS2. The strong charge doping induced by the tunable ion intercalation alters the energetics of various charge-ordered states in 1T-TaS2 and produces a series of phase transitions in thin-flake samples with reduced dimensionality. We find that the charge-density wave states in 1T-TaS2 collapse in the two-dimensional limit at critical thicknesses. Meanwhile, at low temperatures, the ionic gating induces multiple phase transitions from Mott-insulator to metal in 1T-TaS2 thin flakes, with five orders of magnitude modulation in resistance, and superconductivity emerges in a textured charge-density wave state induced by ionic gating. Our method of gate-controlled intercalation opens up possibilities in searching for novel states of matter in the extreme charge-carrier-concentration limit.</P>

        Overexpression of the ABC transporter gene TsABCG11 increases cuticle lipids and abiotic stress tolerance in Arabidopsis

        The cuticle, composed primarily of wax and cutin, covers most plant aerial surfaces and plays a vital role in interactions between plants and their environment. Some ATP-binding cassette G subfamily (ABCG) members are involved in cuticular lipid molecule exportation to outside in the plant surface. Thellungiella salsugineum, a relative of Arabidopsis thaliana with a heavy cuticle, has extreme stress tolerance. TsABCG11, an ABCG transporter was cloned (GenBank accession number JQ389853), and its structure was studied. qRT-PCR showed that TsABCG11 expression varied in different organs of T. salsugineum and was upregulated under ABA, NaCl, drought and cold conditions. The rosette leaves from 4-week-old TsABCG11 overexpressed (OE) Arabidopsis plants displayed lower rates of water loss and decreased chlorophyll-extracted rates compared to wild-type plants. TsABCG11-OE plants also exhibited significantly increased total cuticular wax and cutin monomer amounts, mainly due to prominent changes in the C29, C31, and C33 alkanes in the wax and C18:2 dioic in cutin monomers, respectively. TsABCG11-OE seedlings exhibit lower root growth inhibition under 100 mM of NaCl or 1 μM of ABA than the wild type. Four-week-old TsABCG11-OE plants exhibited higher photosynthetic rates and water-use efficiency under cold stress (4 °C) than control plants. These results indicate that TsABCG11 plays an important role in cuticle lipid exportation and is involved in abiotic stresses, probably having a close relationship with extreme stress tolerance in T. salsugineum.

        A Model Stacking Algorithm for Indoor Positioning System using WiFi Fingerprinting

        With the development of IoT and artificial intelligence, location-based services are getting more and more attention. For solving the current problem that indoor positioning error is large and generalization is poor, this paper proposes a Model Stacking Algorithm for Indoor Positioning System using WiFi fingerprinting. Firstly, we adopt a model stacking method based on Bayesian optimization to predict the location of indoor targets to improve indoor localization accuracy and model generalization. Secondly, Taking the predicted position based on model stacking as the observation value of particle filter, collaborative particle filter localization based on model stacking algorithm is realized. The experimental results show that the algorithm can control the position error within 2m, which is superior to KNN, GBDT, Xgboost, LightGBM, RF. The location accuracy of the fusion particle filter algorithm is improved by 31%, and the predicted trajectory is close to the real trajectory. The algorithm can also adapt to the application scenarios with fewer wireless access points.

        Exosome-mediated lnc-ABCA12-3 promotes proliferation and glycolysis but inhibits apoptosis by regulating the toll-like receptor 4/nuclear factor kappa-B signaling pathway in esophageal squamous cell carcinoma

        Esophageal squamous cell carcinoma (ESCC) is a kind of malignant tumor with high incidence and mortality in the digestive system. The aim of this study is to explore the function of lnc-ABCA12-3 in the development of ESCC and its unique mechanisms. RT-PCR was applied to detect gene transcription levels in tissues or cell lines like TE-1, EC9706, and HEEC cells. Western blot was conducted to identify protein expression levels of mitochondrial apoptosis and toll-like receptor 4 (TLR4)/nuclear factor kappa-B (NF-κB) signaling pathway. CCK-8 and EdU assays were carried out to measure cell proliferation, and cell apoptosis was examined by flow cytometry. ELISA was used for checking the changes in glycolysis-related indicators. Lnc-ABCA12-3 was highly expressed in ESCC tissues and cells, which preferred it to be a candidate target. The TE-1 and EC9706 cells proliferation and glycolysis were obviously inhibited with the downregulation of lnc-ABCA12-3, while apoptosis was promoted. TLR4 activator could largely reverse the apoptosis acceleration and relieved the proliferation and glycolysis suppression caused by lnc-ABCA12-3 downregulation. Moreover, the effect of lnc-ABCA12-3 on ESCC cells was actualized by activating the TLR4/NF-κB signaling pathway under the mediation of exosome. Taken together, the lnc-ABCA12-3 could promote the proliferation and glycolysis of ESCC, while repressing its apoptosis probably by regulating the TLR4/NF-κB signaling pathway under the mediation of exosome.

        OsCAS: a comprehensive web-based annotation platform for rice microarray data

        OsCAS (Oryza sativa Chip Annotation System) is a comprehensive web-based tool that analyzes the results of rice (Oryza sativa) microarray experiments and the potential relationships among genes. This platform is designed to facilitate the indepth exploration of microarray data with relatively high analysis efficiency. With a user-friendly web interface, OsCAS allows chip probe IDs as inputs to retrieve relevant information according to user’s designation. Public databases, such as GenBank, UniGene, Swiss-Prot, TIGR, KOME, KEGG, Gene Ontology, and miRBase, were integrated into our platform to cover gene information, protein features, metabolic pathways and regulatory factors in rice. Besides, OsCAS includes the reprocessed information from several analytical tools such as CSRDB and miRU to obtain deep insights into the primary annotations. OsCAS is time-cost-effective in annotating large sets of chip probe IDs, and can provide some precious hints for the biological experiment design. OsCAS runs on a Linux server and is accessible at http://bis.zju.edu.cn/oscas/.

        Enhancement mechanism of SO2 removal with calcium hydroxide in the presence of NO2

        The enhancement mechanism of SO2 removal by the presence of NO2 under low temperature and humid conditions was studied in a fixed bed reactor system. The presence of NO2 in the flue gas can enhance SO2 removal. The interaction between SO2 and NO2 in gas phase could not explain the effect of NO2 on SO2 removal under lowtemperature and humid conditions. When Ca(NO3)2 and Ca(NO2)2 as additive were added on the surface of sorbent,the desulfurization activity of sorbent decreased. However, the sorbent pretreated by NO2 for a moment has higher SO2removal. The oxidization of SO32− to SO42− and the evolution of sorbent surface structure in the presence of NO2 can explain the enhancement of SO2 removal by the presence of NO2. HSO3− and SO3− reacted with NO2 to form sulfate,which can accelerate the hydrolysis of SO2. The reaction between NO2 and Ca(OH)2 can make the unreacted sorbet under the SO2 removal product exposed to the reactant gas.
