Spherical crystallization of ezetimibe for improvement in physicochemical and micromeritic properties

        Ashwini Patil,Yogesh Pore,Yogesh Gavhane,Shitalkumar Patil,Sachinkumar Patil 한국약제학회 2014 Journal of Pharmaceutical Investigation Vol.44 No.3

Spherical agglomerates of ezetimibe (EZT)were prepared with hydrophilic polymers; polyvinyl pyrrolidoneK30 (PVP) and/or poloxamer 188 (poloxamer) atdrug to polymer ratios of 1:1 (w/w) by spherical crystallizationtechnique, in order to improve its physicochemicaland micromeritic properties. Three different bridging liquids;chloroform, dichloromethane and/or ethyl acetatealong with good solvent acetone and poor solvent waterwere used to form six batches of agglomerates. Initialcharacterization of all batches in terms of micromeritic andphysicochemical properties resulted in optimization of (A3,EZT:PVP:ethyl acetate) and (B3, EZT:poloxamer:ethylacetate) batches and hence further investigated for drug–polymer interaction, crystallinity and morphology usingFTIR, XRPD, DSC and SEM techniques. The resultsindicated presence of hydrogen bonding, crystallinity andspherical shape in agglomerates. Therefore, the optimizedagglomerates (B3) were directly compressed into tablet. Unfortunately, drug release from the tablet was not satisfactory, suggesting a need of disintegrant from dissolutionpoint of view. Therefore, these agglomerates were recompressedincorporating certain excipients and evaluated asper pharmacopoeia. The dissolution rate of prepared tabletwas similar to that of marketed tablet (p[0.05). It couldbe concluded that spherical crystallization could be one ofthe effective and alternative approaches for improvedperformance of EZT and its tablet formulation.

The Dietary Food Components Capric Acid and Caprylic Acid Inhibit Virulence Factors in Candida albicans Through Multitargeting

        Ashwini Jadhav,Supriya Mortale,Shivkrupa Halbandge,Priyanka Jangid,Rajendra Patil,Wasudev Gade,Kiran Kharat,Sankunny Mohan Karuppayil 한국식품영양과학회 2017 Journal of medicinal food Vol.20 No.11

Capric acid and caprylic acid are the dietary food components. They are found to inhibit the virulence factors like morphogenesis, adhesion, and biofilm formation in the human pathogenic yeast Candida albicans. Our study demonstrated that yeast-to-hyphal signal transduction pathways were affected by capric acid and caprylic acid. The expression profile of genes associated with serum-induced morphogenesis showed reduced expressions of Cdc35, Hwp1, Hst7, and Cph1 by the treatment with both the fatty acids. Cell elongation gene, Ece1, was surprisingly downregulated by 5208-fold by the treatment of caprylic acid. Nrg1 and Tup1, negative regulators of hyphal formation, were overexpressed in presence of capric or caprylic acid. Cell cycle studies revealed that capric and caprylic acids arrested cell cycle at G2/M and S phase. Targeting the virulence factors like yeast-to-hyphal transition is efficacious for treatment of opportunistic fungal infections. This research suggests that both capric and caprylic acid may be effective interventions for treating C. albicans yeast infections.

Loss of mitogenome-encoded respiratory genes complemented by nuclear transcripts in halophyte Pandanus odorifer (Forssk.) Kuntze

        Darshetkar Ashwini M.,Patil Swaranjali S.,Pable Anupama A.,Nadaf Altafhusain B.,Barvkar Vitthal T. 한국식물생명공학회 2024 Plant biotechnology reports Vol.18 No.1

The mitochondrial genomes possess genes of five complexes involved in respiration. Mitochondrial gene content greatly varies across eukaryotes. Frequent gene losses have been reported in the mitogenomes of halophytes. In this study, we report, assemble, and annotate complete mitochondrial genome of a halophyte Pandanus odorifer (Forssk.) Kuntze (Pandanaceae). It is a commercially important species for its essential oil. It grows naturally in coastal regions and can withstand winds as well as salt sprays. The mitogenome of P. odorifer is circular with length of 330,962 bp. The genome contains 33 protein coding genes, 3 rRNAs, and 6 tRNAs. The analysis revealed that the important respiratory genes ccmC, cox1, nad1, nad4, and nad9 were pseudogenized in mitogenome but nuclear encoded copies of cox1, nad1, and nad9 were recovered. The phylogenetic analysis placed P. odorifer sister to P. tectorius. Further, mitogenome was compared with the other monocot mitogenomes for their gene contents. Thus, the study provides first complete mitochondrial genome for the family Pandan- aceae and order Pandanales.

Eight prehilar branches of the right renal artery

        Satheesha B Nayak,Surekha D Shetty,Swamy Ravindra,Srinivasa Rao Sirasanagandla,Ashwini P Aithal,Jyothsna Patil,Naveen Kumar 대한해부학회 2014 Anatomy & Cell Biology Vol.47 No.3

Imaging technology with its advancement in the field of urology is the boon for the patients who require minimally invasive approaches for various kidney disorders. These approaches require a precise knowledge of the normal and variant anatomy of vessels at the hilum of the kidney. During routine dissections, a variation in the branching pattern of the right renal artery was noted in an adult male cadaver. The right renal artery divided into upper and lower divisions 6cm away from the hilum of the kidney. The upper division gave 4 branches, and the lower division gave two branches. These two branches further bifurcated and gave 2 branches each. Thus, there were 8 prehilar branches of renal artery. The multiple prehilar branches led to a congested atmosphere at the hilum of the kidney. This arterial congestion might result in hindering the blood flow at the renal hilum. Apart from this, it might cause difficulties in diagnostic and therapeutic invasive procedures. Knowledge of this variation is of importance to radiologists and urologists in particular.
