        Effect of Dietary Vitamin E Supplementation on Serum α-Tocopherol and Immune Status of Crossbred Calves

        An experiment was conducted with twenty crossbred male calves (7-15 days old) divided into 4 different experimental groups on the basis of body weights to study the effect of vitamin E supplementation on the serum ${\alpha}$-tocopherol concentration and immune response of the calves. All the calves were fed on milk and calf starter up to 13 weeks and afterwards, they were fed on concentrate mixture and oat hay up to 32 weeks of age. In addition, the calves in groups I, II, III and IV were supplemented with 0, 125, 250 and 500 IU feed grade DL-${\alpha}$-tocopheryl acetate, respectively. Blood samples were collected at 0 day and subsequently at 1, 2, 3, 4, 6 and 8 months of age to monitor the serum ${\alpha}$-tocopherol concentration in crossbred calves. After 24 weeks of experimental feeding, 4 animals from each group were intramuscularly inoculated with single dose (3 ml) of Haemorrhagic septiceaemia (Pasteurella multocida P52 strain) oil adjuvant vaccine. The cumulative group mean serum ${\alpha}$-tocopherol concentration (${\mu}g/100ml$) was 88.12, 210.11, 235.21 and 294.02 in-groups I, II, III and IV, respectively and differed significantly (p<0.001) among the four groups. Lymphocyte stimulation indices (LSI) did not differ among the groups significantly. The pooled mean ELISA antibody titer against Pasteurella multocida (P52 strain) was 788.02, 926.85, 1,214.00 and 1,109.51 for group I, II, III and IV, respectively, which indicated higher antibody titer in groups supplemented with vitamin E as compared to the control group. It may be concluded that vitamin E supplementation increased the ${\alpha}$-tocopherol concentration in serum and dietary supplementation of vitamin E at higher level has a humoral immune enhancing effect against killed bacterial antigen.

        Development and Validation of a Perfect KASP Marker for Fusarium Head Blight Resistance Gene Fhb1 in Wheat

        Fusarium head blight (FHB) is a devastating wheat disease with a significant economic impact. Fhb1 is the most important large effect and stable QTL for FHB resistance. A pore-forming toxin-like (PFT) gene was recently identified as an underlying gene for Fhb1 resistance. In this study, we developed and validated a PFTbased Kompetitive allele specific PCR (KASP) marker for Fhb1. The KASP marker, PFT_KASP, was used to screen 298 diverse wheat breeding lines and cultivars. The KASP clustering results were compared with gelbased gene specific markers and the widely used linked STS marker, UMN10. Eight disagreements were found between PFT_KASP and UMN10 assays among the tested lines. Based on the genotyping and sequencing of genes in the Fhb1 region, these genotypes were found to be common with a previously characterized susceptible haplotype. Therefore, our results indicate that PFT_ KASP is a perfect diagnostic marker for Fhb1 and would be a valuable tool for introgression and pyramiding of FHB resistance in wheat cultivars.

        Pharmacognostical Evaluation of Leucas aspera Link.

        Leucas aspera Link. (Lamiaceae) is an important medicinal plant in indigenous systems of medicinecomunities find diverse medicinal properties. It is used as antipyretic, stimulant, expectorant and diaphoreticdrug. The present communication deals with the detailed pharmacognostical evaluation of whole plant of L.aspera collected from five different geographical zones of the country-Uttar Pradesh, Orissa, Karnataka, WestBengal and Gujarat. The botanical characters and TLC fingerprint profile of all the samples were quite similar butsome variations were observed in physicochemical parameters. However, some microscopical characters and TLCprofile can be used as diagnostic characters for identification of , for example amphistomachic leavesand two types of trichomes-abundant, non glandular, uniseriate, 1-3 celled and few glandular, 2-5 celled stalk withrounded tip. Presence of some components at Rfs-0.56, 0.65 and 0.76 under UV 366 and at Rfs-0.31, 0.43, 0.60,0.76 and 0.82 under visible light after derivetization in TLC profile may also be used as diagnostic character.

        Standardization and Quality Evaluation of Banafsha

        `Banafsha` is an important herbal drug of indigenous systems of medicine. Flowers of Viola odorata L. (Violaceae) are considered to constitute the genuine drug `Banafsha`. However, due to limited distribution of this plant, another species of Viola, i.e. V. pilosa having almost similar medicinal properties, is mostly used under this vernacular instead. Ethnobotanically also different species of Viola i.e. V. odorata, V. pilosa and V. betonicifolia are used for various ailments at different places in our country. In the present study two species of Viola, namely V. pilosa and V. betonicifolia along with the commercial samples of `Banafsha` were studied and authenticated. It was observed that the market samples procured from Dehradun, Mumbai, Lucknow, Palampur, Ramnagar and Ranikhet showed close resemblance with Viola pilosa in having similar morphological characters like uniseriate hairs on the ovary, geniculate and clavate style, truncate stigma and almost similar TLC profiles. On the other hand the Almora sample was identified as Viola betonicifolia by the presence of large purple flowers, clavate style and convex stigma forming hump like structure. However, the market sample procured from Pathankot was found to be a mixture of two species of Viola namely, V. pilosa and V. odorata having dominance of the former species.

        Pharmacognostic Evaluation of the Flower of Alcea rosea L.

        The flowers of Alcea roses L., Malvaceae, sold in the Indian market under the trade name `Gulkhairo`, are well known for their expectorant, cooling and diuretic properties and used in many indigenous cough mixtures in India. The present paper deals with the detailed pharmacognosy of the floral parts including morphological, anatomical, phytochemical and fluorescence characters. Some of the diagnostic features of the drug are : pedicel characterized by multicellular appendages, stellate hairs, rosette crystals of Ca-oxalate. starch sheath and large sized mucilage canals; sepals having distinctive multicellular appendages arranged in a semilunar fashion present adaxialty at their base; monadelphous stamens, pollen grains pentaporate provided with dimorphic spines; placentation axile, ovules campylotropous; dark green fluorescence of the powder with nitrocellulose in amyl acetate and yellow fluorescence of trichomes under Fluorescence microscope.
