        전정신경초종과 청력손상

        Vestibular schwannomas (VSs) are the most common tumors of the cerebellopontine angle. Although VSs arise from the vestibular nerves, 95% of patients with VS present with sensorineural hearing loss (SNHL). The underlying pathophysiological mechanisms of this SNHL are currently unknown. Better understanding of the possible etiologies of hearing loss will help in guiding VS treatment choices based on tumor size and location. The mechanical effect is thought to involve compression or stretching of the cochlear nerve or compromise of the vascular supply to the cochlea and neurotoxic injury caused either by alterations in the biochemical properties of the inner ear fluid or through accumulation of tumor toxic metabolites may be responsible for hearing loss of VS. We review some of these mechanisms with special emphasis on hearing loss associated with VS.

        약물스크리닝을 통해 청신경종양 세포를 억제하는 유효물질의 발굴

        Objectives : Our knowledge of the molecular biology of vestibular schwannomas (VS) and the development of novel medical therapies for their treatment are increasing. Although several anticancer drugs have been tested in VS, new medical therapies without toxicity are needed. Therefore, we investigated to identify novel hit compounds for potential inhibitors of VS growth using a cell-based screening assay. Method : HEI-193 cells and Nf2 knockout SC4 cells were used to investigate the inhibitory effects on VS cells. We investigated 2,300 FDA-approved / investigational drugs and natural compounds using a cell-based screening assay. Briefly, HEI-193 cells were plated in 96-well plates at 30% confluence and then the test compounds were added to each well at final concentration of 10μM with fresh growth medium. After 24 hour incubation, MTS assays were done to assess cell proliferation. Results : In primary screening, twenty three hit compounds were identified as potential inhibitors of VS growth from 2,320 compounds of FDA approved/ investigational drugs at a concentration of 10μM. The hit compounds were defined by > 50% reduction in VS growth. Conclusion : We identified several schwannoma growth inhibitory compounds for VS from a cellbased screening with 2,320 compounds. These kinds of pharmacologically well-characterized molecules can be readily translated to further preclinical studies for VS.

        크기가 크거나 거대한 전정신경초종의 임상양상 및 청각전정소견

        Background : Large and giant vestibular schwannomas (VSs) are considered a challenging and dangerous disease entity owing to their surgical difficulty and following morbidity. The purpose of this study is aimed to identify the clinical symptoms and audio-vestibular findings in patients with large and giant VS. Methods : We retrospectively enrolled 19 patients who were diagnosed with large (≥3 cm and <4 cm) and giant (≥4 cm) VS from March 2005 to February 2018. We studied the clinical presentations and audio-vestibular findings in patients with large and giant VS. Results : There were 7 large and 12 giant VS in our series. Dizziness was the most frequent presenting symptom in 14 patients with large and giant VS. Gradual hearing loss was observed in nearly half of patients who had central symptoms. While 4 patients showed class A in AAO-HNS hearing classification, all patients showed abnormal canal paresis on caloric test. There was no difference about the clinical symptoms and audiovestibular findings between large and giant VS. Conclusions : If we pay attention to the auditory symptoms of large and giant VS, it would be possible to promote early diagnosis of VS.

        Age-related hearing loss (ARHL) or presbycusis is bilateral progressive sensorineural hearing due to the aging process. ARHL negatively affects interpersonal relationships, cognitive function, psychosocial problems and quality of life. In an aging society, ARHL is rapidly emerging as a public health problem. To improve the welfare and quality of life of the elderly, early diagnosis, prevention and rehabilitation should be carried out. In this review, the characteristics, classification, prevention and treatment of ARHL were investigated.

        한국어판 Penn Acoustic Neuroma Quality-of-Life Scale의 타당도 및 신뢰도 검증

        Background : Quality of life (QOL) has emerged as an increasingly important factor on vestibular schwannoma (VS) treatment. There have been some literatures that have investigated the QOL of VS patients. However, there is no Korean translation of disease-specific questionnaires for VS. The objective of this study was to determine the reliability and validity of a Korean version of the Penn Acoustic Neuroma Quality-of-Life Scale (PANQOL). Methods : The development of the Korean version of PANQOL included the processes of standard independent forward translation, backward translation and reconciliation. The finalized K-PANQOL was administered to 125 patients with VS visiting 3 referral hospitals in a prospective manner. Reliability and validity were evaluated by Cronbach’s alpha coefficient, confirmatory factor analysis and test-retest analysis. Results : K-PANQOL showed good reliability with high internal consistency (Cronbach’s α = 0.939). Construct validity was confirmed by the results of factor analysis and criterion validity demonstrated positive correlations between each part of K-PANQOL and the characteristics of the patients with VS. Conclusions : Reliability, and construct validity outcomes in the current study support the validity of the Korean version of the PANQOL. The K-PANQOL seems to be a valid disease-specific measure of QOL in Korean patients with VS.

        In modern society, looks play an important role in job competition which turns out to be very common. Due to this, female college students of today not only know how to do everyday make-up but also are capable of doing advanced-skilled makeup, which the “before” and “after” shows a vast difference, almost to the point that gives an illusion of plastic surgery. In this research, the majority of the female students recognized the importance of make up through the effect of lookism. And there also will be a further increase of the female college students who do the make-up. Through the online survey which enrolled the female college students who usually gained the makeup skill, information via TV and media, the cosmetics firms can efficiently take advantage of the product information when they seek to make good marketing strategies. Thus the color expression of the domestic brand cosmetics makeup and the technological developments need to be improved urgently. The positive market promotions are also necessary. Therefore, the results of this research can be used as the basis of cosmetics make-up developments and marketing strategies which would be a help in the developments of domestic cosmetics make-up.

        Vocal cord polyps are generally removed using a direct laryngoscope and surgical microscope. In some case of laryngeal microsurgery, laryngeal exposure with surgical microscope is difficult because of severalfactors. We experienced a case of vocal polyp with difficult laryngeal exposure in a 56 year old male patient. The vocal cord polyp was removed successfully through endoscopic approach.

        전기 자극을 이용한 CD271 양성 중간엽줄기세포의 연골세포 분화능

        Mesenchymal stem cells (MSCs) exhibit self-renewing capacity and multi-lineage differentiation potential. CD271 has been identified as a marker of the most homogeneous MSCs subset. In this study, we separated CD271 positive MSCs (CD271-MSCs) from canine adipose tissue-derived MSCs (AD-MSCs) using immunomagnetic selection method. Both CD271-MSCs and AD-MSCs showed similar morphology and expressed common immunophenotypic markers (CD34, CD44, and CD45). However, CD271-MSCs and AD-MSCs exhibited difference in chondrogenic differentiation potential. Specifically, CD271-MSCs exhibited increased expression levels of chondrocyte specific markers, collagen type II alpha 1 (COL2A1) and Aggrecan compared to AD-MSCs. Also, electrical stimulation (ES)-treated CD271-MSCs have a greater capacity for expressing heat shock protein 70 (HSP70), COL2A1, and Aggrecan compared to control without exogenous differentiation condition. This study might contribute to the differentiation potential of homogenous MSCs into cartilage cells by specific ES condition in the absence of exogenous differentiation agents.

        Effects of canine mesenchymal stem cells-derived exosomes in a mouse model of canine mammary tumor

        Canine mammary tumors account for ~30% of all tumors in the female dogs and approximately 50% of the tumors are malignant. Exosomes have been the focus of great interest, as they appear to be involved in numerous important cellular processes. In this study, we examined the anti-tumor effects of canine mesenchymal stem cells-derived exosomes (MSC-exosomes) in an experimental murine mammary tumor model using canine mammary carcinoma cells, REM134. The MSC-exosomes were injected tumor site and tail vein of REM134 xenografted mice. We found that tumor size of the MSC-exosomes-treated group decreased compared to those of the only tumor group in REM134-driven tumorigenic mouse model. In addition, the MSC-exosomes-treated tumor group showed meaningfully reduced expression levels of the MMP-3, IL-1β, IL-6, and TNF-α compared to those in the tumor group. Specifically, we confirmed that the expression level of the CD133, potent cancer stem cell (CSC) markers, decreased in the MSC-exosomes-treated tumor group compared to the tumor group. This study suggests that the MSC-exosomes exhibited anti-tumor effects through downregulating CSC-related markers in the canine mammary tumor murine model. Further study is needed in the future, and we are conducting research on the detailed anti-tumor mechanism of the MSC-exosomes.
