Effects of Cynaroside, Cynarin and Linarin on Secretion, Production and Gene Expression of Airway MUC5AC Mucin in NCI-H292 Cells

In this study, we investigated whether cynaroside, cynarin and linarin derived from Chrysanthemum indicum L. affect the secretion, production and gene expression of MUC5AC mucin in airway epithelial cells. Confluent NCI-H292 cells were pretreated with cynaroside, cynarin or linarin for 30 min and then stimulated with PMA (phorbol 12-myristate 13-acetate) for 24 h. The MUC5AC mucin gene expression, mucin protein production and secretion were measured by RT-PCR and ELISA, respectively. Effect of linarin on EGF (epidermal growth factor) - or TNF-a (tumor necrosis factor-a)-induced MUC5AC mucin gene expression and mucin protein production was also examined. The results were as follows: (1) Cynaroside and cynarin did not significantly affect PMA-induced MUC5AC mucin secretion from NCI-H292 cells. However, linarin decreased MUC5AC mucin secretion; (2) Cynaroside did not affect PMA-induced MUC5AC mucin production and gene expresion from NCI-H292 cells. However, cynarin and linarin inhibited the production and gene expression of MUC5AC mucin; (3) Linarin also inhibited the production and gene expression of MUC5AC mucin induced by EGF- or TNF-a from NCI-H292 cells. These results suggest that linarin can regulate the gene expression, production and secretion of mucin, by directly acting on airway epithelial cells.

Paying attention to the alleged demise of working­class novels, this paper situates Pat Barker’s two working­class novels, Union Street and Liza’s England in the historical context of 1980s England when Thatcher’s neoliberalism began to gain momentum, and probes how they represent the drastic changes and disruption of working­class communities, respectively in the 1970s and 1980s, which were caused in the main by deindustrialization and accelerated by the hostile labor policy of the Thatcher government. It will turn out that these novels, illuminating the lives of working­class women hitherto neglected in the literary representation, not only redress the main tradition of English literature, but make a timely critical intervention in the contemporary political and cultural milieu in which ‘class’ tends to be deliberately excluded from the agenda of public discussion; and that, thematizing the significance of historical memory, especially in Liza’s England, they also enact a gesture of resistance both to Thatcher’s cultural policy that subjects the cultural values with their deep historical implications to the market principles, and to the loss of historical consciousness characteristic of postmodernism.

Akin 절골술의 내측 횡 봉합사 고정: 술기 보고

The Akin osteotomy is a complimentary procedure in hallux valgus surgery. Surgical techniques may vary depending on the operators or fixation devices. Suture fixation, for which a removal procedure is not necessary, can often be recommended. However, there is a risk of failure due to the thin cortex of the phalanx. We describe a new technique using Ethibond suture fixation in Akin osteotomy, which can lower the risk of phalangeal cortical failure and articular cartilage irritation. First, the Akin osteotomy was performed on the proximal phalanx 5 to 6 mm distal to the first metatarsophalangeal joint. Then bone holes were drilled from dorsum to plantar parallel to osteotomy with the Kirschner wire. The final procedure involved passing the Ethibond sutures connected to a straight needle through the holes and tying it. This fixation method offers an effective and easy technique for performance of Akin osteotomy.

While vigorously searching for a new novel form to accommodate the contemporary theoretical insight into the nature of language and novel, John Fowles pays great, though not devoid of irony, homage to the tradition of realism novel. His novels are permeated with his firm belief in the moral, didactic power of art to bring changes to life. This essay tries to show how his didactic intention informs the whole narrative of The Magus, one of his most comprehensive and compelling works. The Magus is a peculiar sort of Bildungsroman, a novel of education and growth, replete with metafictional elements, which also serve to enhance the aesthetic education of the reader. Nicholas, the protagonist of the novel, whose logical and cautiously sceptical attitude invites the reader to identify with him, plays the role of reader as he is induced into an active participation in and interpretation of the metatheatrical ‘godgame’ of Conchis who stands for the metaphorical fictional world. Going through it under the enigmatic direction of Conchis, Nicholas gradually gets over his evasive attitude towards reality as well as his escapist aesthetics. Also his self-centered, life-denying cynicism gives way to a new perception of the mysterious reality of life itself, combined with his growing understanding of the existential human condition. The concluding section of this essay thinks over a few problems regarding the author's overt didacticism in this novel, casting a last short glance at the ironical fact that, over the whole period of his writing, he failed to make much progress in his own existential ideas in spite of his unremitting passion for education.

리얼리즘과 깨진 “말-거울”: J. M. 쿳시의 『엘리자베스 코스텔로』

The purpose of this paper is to examine J. M. Coetzee’s ideas on realism in Elizabeth Costello, with a focus on its three frame stories: Lesson 1, “Realism,” Lesson 8, “At the Gate,” and the Postscript. In Lesson 1, he detaches realism from the practice of detailed realistic description or the notion of verisimilitude and redefines it by the pivotal notion of ‘embodiment’ and ‘embeddedness (in life).’ Also, he presents realism as facing an unprecedented plight resulting from the language crisis introduced by Costello’s speech on the broken “word-mirror.” What this crisis points to is the irrevocable loss of correspondence between word and object, statement and meaning. In Lesson 8 and the Postscript, the motif of language crisis is more fully developed, and the allegorical dimension of language is explored, heightening the conflict and tension between the realistic impulse to be faithful to the reality of things and the representational limitations of language. The paper concludes that staying in this taut tension is essential to Coetzee’s new version of realism, which offers an alternative both to the untenable old realism and to the fall into the abyss of representational impossibility.

The Effect of Bone Morphogenic Protein-2 (BMP-2)-Immobilizing Heparinized-Chitosan Scaffolds for Enhanced Osteoblast Activity

The aim of orthopedic and dental tissue engineering is to generate synthetic bone-graft tissue substitutes. It is generally comprised through the combination of viable cells, a scaffolding material, and sometimes the inclusion of bone morphogenic proteins. The object of this study is to develop novel bone-grafting scaffolds that enhance osteoblast activity. We were fabricated the chitosan scaffolds with channel-shaped and spherically shaped pore morphologies. Also, Bone morphogenic protein-2 (BMP-2) was sequentially immobilized to the heparinized-chitosan (Hep-chitosan)scaffolds. Osteoblast activities of all chitosan scaffolds were investigated by a cell proliferation assay, alkaline phosphatase (ALP) activity, calcium deposition, and the expression of osteogenic markers. The results showed that BMP-2-immobilizing heparinized-chitosan (BMP-2/Hep-chitosan) scaffolds significantly enhanced ALP activity and calcium deposition of the osteoblast cells when compared with chitosan scaffolds only. Also, mRNA expressions of osteocalcin and osteopontin of osteoblast cells cultured on BMP-2 (100 ng)/Hep-chitosan scaffolds were increased versus chitosan scaffolds only. Taken together, BMP-2 (100 ng)/Hep-chitosan scaffolds could achieve the functions of excellent osteoblast promotion. Therefore, osteoinductive protein-functionalizing scaffold substrates such as BMP-2/Hepchitosan scaffolds are a promising material for the enhanced osteoblast activity in orthopedic and dental fields

무지외반증 환자에서 제1중족골두의 연골 미란: 수술 중 실측한 연골 미란과 수술 전 측정지표의 연관성

Purpose: To analyze relation between age or parameters measured before operation and cartilage erosion of the first metatarsal head measured during operation. Materials and Methods: The study was targeted at 56 patients and 79 feet, who underwent Scarf osteotomy or Scarf and Akin osteotomy from November 2009 through November 2010, and whose cartilage lesion of the first metatarsal head referred to the cartilage grade III or IV of the International Cartilage Repair Society. The measurement parameters were age, hallux valgus angle, intermetatarsal angle (1~2), tibial sesamoid position, proximal articular set angle and distal articular set angle. The cartilage erosion of the first metatarsal head was measured by one surgeon using cellophane. Occupancy rate and frequent involved sites of the cartilage erosion were recorded using Auto$CAD^{(R)}$ and adobe Illustrator CS4 program. SPSS correlation test and T-test were used for statistical analysis of the parameters and the cartilage erosion. Results: The cartilage erosion was incurred frequently in the sagittal groove and the site where subluxation or dislocation of the tibial sesamoild bone occurred but frequent involved sites had no statistical significance with cartilage erosion. The age showed a statistical significance with the cartilage erosion in the correlation test (p=0.003). Especially, the group of over 51 year old patients was turned out to have association with the cartilage erosion, compared to the group of below 51 (p=0.007). But, hallux valgus angle, intermetatarsal angle (1~2), tibial sesamoid position, proximal articular set angle and distal articular set angle were no statistical significance with the cartilage erosion. Conclusion: We found the more the age of patients increased (especially above 51), the more cartilage erosion increased. And it is thought that we pay attention to reduce tibial sesamoid bone.

D. H. 로렌스의 「말장수의 딸」 : 온전성 탐구로의 초대

이 논문은 로렌스의 대표적 단편의 하나인 「말장수의 딸」에 존재의 온전성을 탐구하는 작가의 문제의식이 어떻게 담겨 있는지를 살펴보는 한편 작품이 미묘 하면서도 지속적으로 독자의 감정적, 지적 감수성을 요구한다는 점에 주목한다. 지금까지 비평 흐름을 보면, 이 작품의 주제가 ‘생의 승리’ 또는 죽음으로부터 삶 으로의 재탄생이라는 데는 대체로 일치하나 작품의 세부 장면이나 인물들의 성 격을 두고는 다양한 상이한 해석이 있어 왔다. 이는 작품 자체에 의도적으로 모 호성이 심어져 있어서 발생하는 문제라기보다 인물들 내면의 무의식적 층위가 미묘하게 제시되고 행위의 동기가 불분명하게 나타나는 작품의 특성에서 비롯된 다. 따라서 작품을 사실적인 층위에서 해석하기 까다로운 점 때문에 신화적, 상 징적 또는 심리학적 해석에 치우치는 경향이 있었다. 본고는 두 중심인물이 깊이 연루된, 작품의 가장 핵심적이면서도 당혹스러운 장면에 분석의 초점을 맞추는 가운데 작품의 존재론적인 의미를 규명하고, 그러한 의미는 독자의 적극적인 감 정적, 지적 참여를 통해 형성되는 것임을 환기한다. This paper aimes to examine D. H. Lawrence’s “The Horse-Dealer’s Daughter”, one of his best-known short stories, as a quest for the wholeness of being, trying to show how it makes consistent and subtle demands on the reader’s emotional and intellectual sensibility. Though most of critics agreed to see this story as a ‘triumph of life’ or a passage from death to rebirth, there has been a wide difference in interpretations on key scenes or traits of characters. It is not because of ambiguities planted in it, but of subtle presentation of the inner workings of characters’ psyche and apparent lack of motivation. The difficulty in understanding this story at a realistic level led to the prevalence of mythological, symbolic or psychological interpretations. Focusing on its most crucial and puzzling scene, in which two main characters are deeply involved, this paper will tease out its ontological meaning which could not be reached without reader’s active emotional and intellectual engagement.

몸을 사유하기: D. H. 로렌스와 장-뤽 낭시(Jean Luc-Nancy)

In the midst of overflowing discourses on the body, this paper aims to compare Lawrence’s thinking of the body with that of Jean-Luc Nancy and gain a new understanding of the nature of the body. Pointing out that the body is not an object of knowledge, but an experience in itself, both of them emphasize that the body is open to itself and to other bodies. For Nancy, the body, an open space where existence takes place, is not a substance. It always comes to its presence; it is an aperture, a self in departure, repeating its movements of arriving and withdrawing. Also a body cannot exist alone; it co-exists; it is exposed to other bodies, always already in touch with them. Lawrence, with his relational thinking and his firm belief in the ever-flowing, living present of being, shares these ideas and explores how man can achieve a harmonious relationship with his circumambient universe, avoiding the disastrous damages brought about by the death of the body in the modern society. There is, however, an unbridgeable gap between their thoughts. While Lawrence lays stress on the integrity of embodied self, Nancy argues that the body, constituted by fragmentation of parts and division of senses, is never completely whole nor proper. This discord reminds us of the elusiveness of the body that demands continual efforts to keep our thinking in contact with it.

Enhanced Effects of Osteoclastogenesis Inhibition by Curcumin-Delivering Heparin Nanoparticles

Curcumin has various pharmacological activities such as anti-inflammatory, antioxidant, antimicrobialand antitumor activity. However, its use has been limited due to its poor solubility in water and minimal systemicbioavailability. Thus, we developed curcumin-delivering heparin-based nanoparticles and evaluated the inhibitioneffects of osteoclastogenesis by curcumin-delivering heparin nanoparticles (Cur-HD NPs). Cur-HD NPs were dispersedwell in aqueous solution by forming self-assembled nanoparticles and showed sustained release of curcumin. In vitro studies, HD NPs facilitated intracellular delivery of curcumin into macrophages and osteoclasts, and thus,Cur-HD NPs effectively inhibited osteoclastogenesis in a dose-dependent manner by suppressing tartrate-resistantacid phosphatase (TRAP) activity and TRAP-positive multinucleated cells as well as by reducing the expression ofosteoclast marker genes (i.e., TRAP and nuclear factor of activated T cells cytoplasmic 1 (NFATc1)). Furthermore, Cur-HD NPs markedly stimulated apoptosis of osteoclasts. Therefore, we hope that Cur-HD NPs will be useful nanodrugs forthe treatment of bone-related diseases.
