Hexagonally packed microwell plates for hypoxic microenvironment induction in tumorspheres

The growth of a solid tumor in a confined microenvironment leads to increased compression, hypoxic/nutrition gradient within the tumor. However, Tumorspheres (TSs) cultured in free of spatialconfinement do not exhibit physiological compressive stress. Here, we designed a microwell-basedsphero-plate that allowed TSs to experience spatial confinement. On this platform, TSs underwentgrowth-mediated compressive stress and formed hypoxic cores. Moreover, drug resistance was enhancedby the hypoxia-inducible signaling and decreased by HIF-1 targeted drug within compact TSs. Thesefindings highlight the impact of the design of the TSs-based drug testing platform.

Rosin-alum sizing has been very important in papermaking since it was developed by Morits lllig in 1807. But effect of alum is varied by pH condition very significantly. Therefore rosin sizing has limited in acid condition. So cellulose reactive sizing agents were developed and used papermaking industry. But they have so many problems and concern for rosin sizing in neutral pH condition is increased. In this study, we attempt to confirm the possibility of rosin sizing in neutral pH condition with cupric ion and C-PAM as mordant.
