        Centrifugal analyer를 이용한 혈청중 Complement C3 C4 측정법의 검토

        The complement acts as a part of immune system to protect the individual from microbial infection by mediating a variety biologic reaction; opsonization, chemotaxis of leukocytes, increased vascular permeability, and cytolysis of target organisms. The complement system is consisted about 18 proteins, among them, The C3 and C4 are the highest concentrates in serum. These proteins are said to be in "classic pathway" or "alternative pathway" names. Activation of the complement proteins result in loss of their precursors, native activities and, or their accelerated clearence from p1asma. Progressing hypercatabolism without compensation by increased synthesis of complements, the determination of depressed levels of them may provide the evidence of the comp1ement system activation. For the evaluation of the patient with autoimmune disease such as autoimmune hemolytic anemia, SLE, rheumatoid arihritis and poststreptococcal glomerulonephritis, the determination of C3 and C4 concentration are both of the most informative method. Low concentration of C4 indicated that classic pathway activation has occurred, since C4 is extremely sensitive to C1 Depressed level of C3 suggests that rather intense activation of ether pathway. Decreased C3 associated with normal C4 levels indicate exclusive activation of alternative pathway. During the past decade, a number of invastigators have developed the techniques for measurement of complement. Since 1929, Michael Heidleberger has developed immunoprecipitation analysis of complement. And also Boyden et al (1947) demonstrated turbidimetric absorbance using the basis of antigen quantitation. The radial immunodiffusion (RID) technique has proved to be one of the most useful methods in clinical laboratories during the past few years, but it require rather long processes from 18 to 48 hrs in order to complete precipitation ring formation. So we attempt to introduce and establish more rapid completion of complement quantitation based on turbidimetric technique using a centrifugal analyzer.
