The present study aimed to screen unrecorded wild yeasts from Jeju lsland and Jangsado on the southern coast of Korea, and to investigate their microbiological characteristics. To date, Coniozyma leucospermi JJD37-2, Hanseniaspora thailandica JJD44-1, Kluyveromyces nonfermentans JJD15-1, Kockovaella fuzhouensis JJD47-3, Vishniacozyma heimaeyensis JJD8-4, Candida carpophila JSDH24-1, Wickerhamomyces strasburgensis JSDH34-2, Candida savonica HJD6-4, and Candida karawaiewii YP23-3 have not been previously recorded in Korea. In the present study, we investigated the microbiological characteristics of these previously unrecorded yeasts. Except for W. strasburgensis JSDH34-2 strain, none of the strains formed spores, and only the C. leucospermi JJD37-2 strain formed pseudomycelia. Almost all strains grew well in yeast extract-peptone-dextrose (YPD) medium, potato dextrose (PD) medium and yeast extract-malt extract (YM) media. C. carpophila JSDH24-1 and W. strasburgensis JSDH34-2 also grew well in YPD medium containing 10% NaCl. H. thailandica JJD44-1 is fermented to produce glucose, fructose and mannose.
‘코넬리우스 반틸의 삼위일체론’에 관한 비평적 고찰 : ‘전통적 삼위일체론’과의 ‘공통성’ 및 ‘차이성’을 중심으로

본 연구는 전통적 삼위일체론과의 공통성 및 차이성에 기초하여 코넬리우스 반틸의 삼위일체론을 비평적으로 고찰하는 것을 목적으로 한다. 전통적 삼위일체론의 핵심논점은 ‘니케아-콘스탄티노플 공의회’를 통해 ‘한 본질-세 위격들’로 정식화되었다. 일반적으로, “전통적 삼위일체론에서 본질과 위격은 서로 다른 논의 지평에 속하기 때문에 전승되어온 삼위일체론에는 아무런 모순이나 비합리적인 요소도 없다”라고 말해진다. 그러나 반틸은, “본질과 위격이 개념적으로 서로 구별되는 것이기 때문에, 전통적 삼위일체론에는 이제 그 어떠한 형태의 한계성도 존재하지 않는다”라고 주장하는 것만으로는 “아직 충분치 않다”고 역설한다. 이를 입증하기 위해, 반틸은 먼저 자함적인 삼위일체 하나님을 자기 존재의 근거적 의미를 자기 안에 담지하는 절대적 합리성의 개념을 통해 사유한다. 그리고 반틸은 이러한 절대적 합리성을 가진 삼위일체 하나님은 개별성과 보편성이 모두 궁극적인 것으로 제시되는 ‘절대적 인격체’, 곧 ‘한 인격-세 인격들’로 계신 하나님이시라고 강조한다. 이와 같은 반틸의 삼위일체론은 전통적 삼위일체론, 특히 서방교회적 삼위일체론과의 발전적 연속성 아래에서 이해된 공통성과 차이성을 동시에 가진다. 전통적 삼위일체론보다도 더 정통적인 삼위일체론을 추구했던 반틸은, 서방교회가 ‘한 본질-세 위격들’로 정식화된 전통적 삼위일체론에서 ‘한 본질’의 존재론적 성격을 유비적 사유에 기초하여 ‘비인격적인 것’으로 이해함으로써 근본적인 한계성을 가지게 된다고 파악한다. 반틸은 삼위일체 하나님의 ‘한 본질’이 ‘비인격적인 본질’에만 머무르는 것이 아니라 ‘한 인격’임을 계시적으로 밝혀냄으로써, 개혁주의 삼위일체론에 독창적인 공헌을 남겼다. 그럼에도, 논자는 그 ‘한 인격’을 계시된 말씀에 근거하여 ‘긍정적’으로 이해할 수 있는 방향으로 추가적인 보충연구가 이루어져야 한다고 본다. The purpose of this study is to critically examine the Trinitarianism of Cornelius Van Til based on the Commonality and Difference with the Traditional Trinitarianism. The Traditional Trinitarianism was formulated as ‘one Essence-three Persons’ through ‘the Council of Nicaea-Constantinople’. Generally, it is said that there is no contradiction or irrational element in the Traditional Trinitarianism because Essence and Person belong to different horizons of discussion. However Van Til insists that it is not yet enough to argue that there is no limit to the Traditional Trinitarianism because Essence and Person are conceptually distinguished. To prove this, Van Til first thinks of the Trinity God through the concept of Absolute Rationality which contains the grounded meaning of self-being in Himself. And Van Til emphasizes that the Trinity God with such Absolute Rationality is God who is Absolute Personality, in which particularity and university are both as the Ultimate that is ‘one Person-three Persons’. Such Trinitarianism of Van Til has the Commonality and Difference understood under developmental continuity with the Traditional Trinitarianism, especially Western Church Trinitarianism. Here, Van Til seek after more Orthodox Trinitarianism than Traditional Trinitarianism. Van Til who pursued more orthodox Trinitarianism than the Traditional Trinitarianism grasps that Western Church has fundamental limit by understanding the ontological nature of one essence as non-person based on analogical thinking in the Traditional Trinitarianism formalized as one Essence-three Persons. Van Til made a creative contribution to the Reformed Trinitarianism by revealing that the One Essence of the Trinity God is not only non-personal essence but one Person. Nevertheless, the author thinks that additional supplementary research should be conducted in a direction that can be understood positively the one Person based on the Bible’s written revelation.
제조공정에 따른 강종별 잔류응력 특성에 관한 연구; AISI 1536V, AISI A387

This study analyzes the residual stress of AISI 1536V for an engine shaft of the shipbuilding industry and AISI A387 for a reactor shell of the chemical refining industry by the hole drilling method with a strain gauge rosette, which transforms fine mechanical changes into electrical signals. Tensile residual stress is generated in the forging and heat treatment process because specimens are affected by thermal stress and metal transformation stress. In the heat treatment process, the residual stress of AISI A387 is almost 170% the yield strength at 402 MPa. Since during the machining process, variable physical loads are applied to the material, compressive residual stress is generated. Under the same condition, the mechanical properties greatly affect the residual stress during the machining process. After the stress-relieving heat treatment process, the residual stress of AISI A387 is reduced below the yield strength at 182 MPa. Therefore, it is necessary to control the temperature, avoid rapid heat change, and select machining conditions depending on the mechanical properties of materials during manufacturing processes. In addition, to sufficiently reduce the residual stress, it is necessary to study the optimum condition of the stress-relieving heat treatment process for each material.
충남 논산천과 삽교호로부터 야생효모의 분리 및 국내 미기록 효모들의 균학적 특성

This study aimed to isolate wild yeasts from water and soil sample of the Nonsan stream and Sapgyoho (lake) in Chungcheonnam-do, Korea, and to further characterize previously unrecorded wild yeast strains. In total, 102 strains, representing 55 different species of wild yeast were isolated from 95 samples collected from the Jangseoncheon and Ipchoncheon of Nonsan stream in Jellabuk-do and Chungcheonnam-do. Among these, 33 strains were isolated from alkalophilic yeast extract-peptone-dextrose (YPD) medium (pH 9.0), and 9 strains were isolated concurrently on general YPD medium (pH 6.5) and alkalophilic medium. Seventeen strains of Cryptococcus laurentii were predominantly isolated. Additionally, 65 strains, representing 27 different species of wild yeast were isolated from 58 samples obtained from Sapgyoho (lake). Among the 82 isolated wild yeast strains, 8 strains, including Candida fructus JSC 72-1(JSL-GGU 015), had not previously been recorded. All 8 previously unrecorded yeasts were oval in shape except C. fructus JSC72-1(JSL-GGU-015), and only the Filobasidium chernovii JSC39-1(JSL-GGU-013) strain formed spores. All strains except Pseudosydowia eucalypti JSC23-6(JSL-GGU-012) grew well in yeast extract-peptone-dextrose (YPD) and yeast extract-malt extract media and grew in vitamin-free medium. Four strains, including P.eucalypti JSC23-6(JSL-GGU-012) grew well in 15% NaCl-containing YPD medium. F.chernovii JSC39-1(JSL-GGU-013) and Sirobasidium intermedium JSC7-3(JSL-GGU-014) assimilated lactose, and five strains, including F. chernovii JSC39-1(JSL-GGU-013) also assimilated starch. All strains were resistant to 800 ppm of Ca, Cu, Li, and Mg ions.
비병원성 야생효모들로부터 생산된 항산화물질과 항통풍성 Xanthin 산화효소 저해물질의 부분정제 및 특성

비병원성 Rhodosporidium paludigenum HHGG35-1와 Rhodosporidium diobovatum NMD18-1 균주들이 생산하는 항산화물질과 항통풍성 물질을 산업적으로 응용하기 위하여 먼저 이들 각각의 YPD 배양 상등액을 한외여과를 실시하여 3kDa 이하의 활성물질을 얻은 후 Sephadex G-50으로 여과하여 부분 정제된 항산화물질과 xanthin 산화효소 저해물질을 얻었다. 부분 정제한 항산화물질의 작용 최적 온도와 pH는 각각 30℃, pH 7.0 이었고, xanthin 산화효소 저해물질의 작용 최적 온도와 pH는 각각 15℃, pH 5.0 이었다. The antioxidants of Rhodosporidium paludigenum HHGG35-1 was partially purified by ultrafiltration and Sephadex G-50 filtration and investigated its optimal reaction condition. Optimal reaction temperature and pH of the partial purified antioxidants were 30℃ and pH 7.0. The xanthin oxidase inhibitor from Rhodosporidium diobovatum NMD18-1 was purified partially by enzyme treatment, ultrafiltration and Sephadex G-50 filtration. Optimal reaction temperature and pH of the partial purified xanthin oxidase inhibitor were 15℃ and pH 5.0. Some heavy metal ions and sugars did not affect inhibitory activity of the parial purified xanthin oxidase inhibitor.

항진균 바이오의약의 소재인 killer toxin을 비병원성 야생효모로부터 생산하기 위하여 25개 비병원성 색소 생성 효모들의 killer toxin 활성을 측정하여 우수균주로 Rhodosporidium toruloides HGG35-2와 Wickerhamomyces anomalus HO9-2을 선발하였다. 이들 killer toxin 생성균주들의 효모 감수성 균주들에 대하여 killer 활성을 측정한 결과 시판 포도주등의 알콜발효로 이용되고 있는 Saccharomyces cerevisiae (La Parisienne®)에 대하여 우수한 항진균활성을 보였다. Killer toxin production of non-pathogenic wild yeasts, Rhodosporidium toruloides HGG35-2 and Wickerhamomyces anomalus HO9-2 were investigated. Rhodosporidium toruloides HGG35-2 showed strong killer activity against pathogenic yeast, Candida guilliermondii AHP28-1 and commercial alcohol fermentation yeast, Saccharomyces cerevisiae(La Parisienne). Wickerhamomyces anomalus HO9-2 also showed killer activity against Rhodotorula nothofagi JSC10-1 and commercial alcohol fermentation yeast, Saccharomyces cerevisiae(La Parisienne®).
항치매성 acetylcholineaterase 함유 검은 비늘버섯 추출물의 PC12 신경세포사 저해 효과

본 연구에서는 부분 정제된 acetylcholinesterase 저해물질을 함유한 검은비늘버섯 추출물의 신경세포 PC12에 대한 세포독성을 조사하고 PC12 신경세포 저해효과를 조사하였다. 검은 비늘버섯 추출물은 세포 생존에 영향을 미치지 않았고 무처리군의 85.6%에 비해 19.3%로 세포사 저해효과가 우수하였다. Dementia disease has been increased because of the significant increase in life expectancy over sixty-five years of age in Korea. This studies were carried out to develop new anti-dementia compounds such as acetylcholinesterase(AChE) inhibitor from edible mushrooms.. AChE inhibitor from water extract of finally selected Pholiota adiposa were partially purified by Sephadex G-50 gel permeation chromatography and RP-HPLC. The partial purified acetylcholinesterase inhibitor-containing Pholiota adiposa extracts showed high cell viability and also showed apoptotic inhibitory effects against PC12 neuron-like cells.
야생효모, Wickerhamomyces anomalus HO9-2로부터 급성 폐손상 억제물질인 호중구 elastase 저해물질 생산 및 생산균주의 균학적 특성

This study was performed to produce acute lung injury-preventing neutrophil elastase inhibitor from non-pathogenic wild yeast and further determine enzyme activity and physiological functionality of the potent wild yeast. Culture supernatants and cell-free extracts of one hundred twenty five non-pathogenic wild yeast strains were prepared and their neutrophil elastase inhibitory activities were measured. The neutrophil elastase inhibitory activity of the cell-free extract from Wickerhamomyces anomalus HO9-2 showed the highest inhibitory activity of 38.4% and its elastase inhibitor was maximally produced(IC50,3.2㎎) when Wickerhamomyces anomalus HO 9-2 was cultured in pH 7.0 of malt extract medium at 20℃ for 24h. Wickerhamomyces anomalus HO9-2 was oval shape and did not form ascospore and pseudomycelium. The HO9-2 strain grew well in YPD and YM media and was halophilic yeast which grew well in YPD medium containing 15% NaCl. Cell-free extract of the HO9-2 strain were showed high 17.2U of glucoamylase activity and anti-diabetic α-glucosidase inhibitory activity of 87.5%. 본 연구는 급성 폐손상 예방(또는 치료)의약 소재를 개발하기 위하여 비병원성 야생효모들의 elastase 저해활성을 측정하여 우수 균주를 선발하였다. 또한 선발균주의 elastase 저해제 생산 조건을 최적화 하였으며 선발균주의 미생물학적 특성 등을 조사하였다. 비병원성 야생효모인 Wickerhamomyces anomalus HO9-2의 무세포추출물이 38.4%의 호중구 elastase 저해활성을 보여 우수균주로 선발하였고 이 균을 pH 7.0 의 malt extract 배지에서 20℃로 24시간 배양하였을 때 최고의 elastase 저해활성(IC50, 3.2mg)을 보였다. 또한 이 균은 난형으로 포자와 의균사를 형성하지 않았고 YPD와 YM배지에서 생육이 양호하였고 NaCl을 15% 함유한 배지에서도 생육하는 호염성 효모이었다.
검은 비늘버섯 Pholiota adiposa로부터 항치매성 Butyrylcholinesterase 저해물질의 생산

본 연구에서는 식용버섯 유래의 새로운 항치매성 butyrylcholinesterase(BChE) 저해물질을 생산할 목적으로 식용버섯 추출물들의 BChE 저해활성을 측정하였다. 식용버섯들 중에서는 검은비늘버섯의 물 추출물이 42.7 %의 BChE 저해활성을 보여 최종 선발하였다. 또한 검은비늘버섯 자실체를 30 ℃에서 24시간 물 추출하였을 때 BChE 저해활성이 가장 높았다. n this paper, screening potent edible mushroom with high anti-dementia butyrylcholinesterase(BChE) inhibitory activity and optimal extraction condition of BChE inhibitors were described. Among various edible mushroom, the water extract from Pholiota adiposa fruiting body had high BChE inhibitory activity of 42.7 %. BChE inhibitor from P.adiposa fruiting body was maximally extracted when its fruiting body was trated with distilled water for 24 hr at 30 ℃.
