        소, 돼지 미성숙 난포란의 유리화 동결. 융해후 FDA 처리가 체외수정과 배 발육에 미치는 영향

        This experiment was carried out to study the determination of survival of vitrified and thawed mammal follicular oocytes by FDA-test. Oocytes were divided into 3 groups according to attachment of cumulus cell. Group A oocytes were tightly surrounded by cumulus cell, group B oocytes were partially surrounded by cumulus cell, and group C oocytes were poorly surrounded by cumulus cell. Vitrification solution developed by our previous study (Kim et al, 1992) which consisted of permeable agent (20 % glycerol + 10 % ethylene glycol) and nonpermeable agent (30 % Ficoll + 10 % sucrose). Oocytes (7~10) loaded into 0.25 ml straw after 10 min equilibration were plunged into liquid nitrogen (- 196) directly. The FDA-score of vitrified and thawed group A oocytes was higher in rat (4.2) than in rabbit (3.9), cow (3.8), mouse (3.4) and porcine (2.4), however that of cumulus cell was higher in rabbit (4.7) than in rat (4.1), cow (2.9), porcine (2.6) and mouse (1.4). The FDA-score of vitrified and thawed group B oocytes were 3.1 (cow), 2.9 (rabbit), 2.9 (mouse), 2.6 (rat) and 2.5 (porcine), respectively. However that of cumulus cell was higher in rabbit (3.7) than in porcine (2.6), rat (2.3), cow (1.7) and mouse (0.3). The FDA-score of vitrified and thawed group C oocytes was higher in mouse (4.1) than in cow (2.9), rabbit (2.6), rat (1.3) and porcine (1.1). As shown in the above results, The survival rates of oocytes were higher in group A than in group B and C except in mouse and cow. These results suggest that the survival of cumulus cell as well as follicular oocytes can be reliably judged by their fluorescence with FDA-test.

        제주도 한우의 번식장해 발생원인과 대책에 관한 연구 제2보 , 제주한우의 번식장해 발생상황 조사연구

        The objective of this study was the status of reproductive disorder for Korean cattle in Jeju-do area and provide the fundamental data for the suitable control of this problem. Four hundred and fifty seven cows were used in this survey according to the coast area, mountain area and enterprise properties in Jeju-do. The results are summarized as follows; 1. The total occurrence rate of the reproductive disorder due to genital discase(12.9%) and. distocia (3.3%) was 16.2%, but the figure did not include the delayed puberty and postpartum Estrus. 2. In total 457 cases, the cows with ovarian disease were 39 cows(8.5%), 6cows (1.3%), in uterine disease, 3 cows (0.7%) in vaginal disease and 11 cows (2.4%) in repeat breeder. 3. In 43 sterile cows, the animals with overian disease were over 65% ; in which ovarian atrophy appeared 30.5%, ovarian hypoplasia 15.2%, delayed ovulation 10.2% and ovarian cyst and persistent corpora lutes were both 5.1%. 4. The delivery disorders in total investigated cows were 1.5% with abortion, 0.9% stillbirth, 0.22 dystocia and 0.65% retained plecenta, and these results demonstrated that native cattle showed less delivery disorders than that of dairy cattle. 5. On the investigation of the delayed puberty 123 heads (40.9%) among 301 cows showed delayed puberty at 26-30 months old and 45 heads (14.9%) showed it at 31 months old. 6. Among 284 cows 103 heads (36.3%) were the alternate year calving cows which showed alternation more than once and in 760 total calving frequencies, the alternate year calving was 118 frequencies (15.5%).

        제주도 한우의 번식장해 발생원인과 대책에 관한 연구 제1보 , 제주한우의 번식상황에 관한 연구

        This study was conducted to investigate the reproductive status in Jeju native cattle. Total 800 cows from 10 districts and 2 government faciities in 4 main area of Jeju-do were employed in this study. The results obtained regionally were summarized as follows; 1. The occurrence of the age at first estrus was average 26.3 months, but, regional differences in the first estrus were observed shortest in southern area (20.6 months) and the longest in eastern area (28.3 months). 2. The age of first insemination averaged 30.2 months but showed a few regional differences as that of sexual maturity. 3. The length of estrus cycle and gestation period of cows were average 21.0 and 284.3 nays respectively and no regional differences were found to except for calving interval;420 days south area) 444.8 days (north area.) 447.7 days (west area) and 453.2 days (east area), respectively. 4. The average age at first calving was 1127 days and those of cows in south and west area showed one month earlier than that of east and north area. 5. The first estrus after calving appeared on 70.7 days in south area and 77.3 days in northern area. Longest period was observed in as tern region and government fecilit.

      • FDA-Test 생사판정법이 초급속 동결된 Mouse 수정란의 배양과 이식후 착상에 미치는 영향 I. FDA 첨가 수준이 초급속 동결된 생쥐상실배의 배양과 이식에 미치는 영향

        This study was carried out to test the effects of fluorescein diacetate(FDA 0${mu}ell$/ml, 0.5${mu}ell$/ml, 5${mu}ell$/ml, 10${mu}ell$/ml or 0${mu}ell$/ml, 5${mu}ell$/ml in PBS) treated before culture on the survival of vitrified mouse morulae in vitrification solution(20% glycerol+glycerol+10% ethylene glycol+30% ficoll+10% sucrose). The results were summarized as follows; 1. The survival rate of FDA-tested fresh mouse morulae after 24 hours culture was over 96%((P4.8) in the control or treatment groups with various levels of FDA. Because the rate of mouse morulae which developed to hatched blastocysts was higher with the various levels of FDA treatment(67%) than control(50%), it was considered that toxicity of FDA did not affect the survival of mouse morulae. 2. When mouse morulae were FDA-tested in FDA 0(control), 0.5, 5, and 10${mu}ell$/ml treatment after vitrification, the development rate to expanded blastocyst were 66, 82, 64 and 76%, and FDA scores were P4.2(84%), P4.7(94%), P4.2(84%) and P3.9(78%), respectively. There were no significant differences between control and FDA treatments, but there were significant difference between 0.5${mu}ell$/ml)94%) and 10${mu}ell$/ml(78%) treatment(P<0.01). 3. The survival rates of cultured mouse morulae according to FDA-scores(P0=non-fluorescence; P1~P5=according to their fluorescence) after vitrification were P5;92%, P4;67%, P3;42% and P2.P0;0%, respectively. 4. Implantation rates of morulae stage embryos cultured into early blastocysts and implanted into uterine hornes vitrification were 14 and 11% embryos treated control(0${mu}ell$/ml) and FDA 5${mu}ell$/ml and the normal fetus development was 2% embryos for both treatments. Results of this percent study indicated that toxicity of FDA does not affect not only the survival of fresh and vitrified mouse morulae but also the development rate and implantation of fetus after transfer as well. The development rates of mouse morulae with the FDA score of P5, P4 and P3 were 92, 67 and 42%, respectively, it was considered that FDA-test was fit for the judge of survival.

        제주도 한우의 번식장해 발생원인과 대책에 관한 연구 제3보 . 난소질환 제주한우에 있어서 hormone 처리가 수태에 미치는 영향

        According to the investigation of reproductive status and disorders in Jeju native cattle, the delayed puberty, fist calving age in heifer, and the alternate year calving cow due to anestrus following calving were observed more frequently on this island. This study, therefore, was carried out to investigate the effect of PMS as a method of treatment for the cows with ovarian hypoplosia and atrophy. For the 15 cows with ovarian hypoplasia, the blood was tested and 40 cows of anestrus due to ovarian hypoplasia and atrophy were selected to treat with PMS. The results obtained were an follows. 1. The blood test indicated that cows of the ovarian hypoplasia appeared to bave lower serum protein and calcium level than that of normal cow. Especially obnormality in A/G and Ca/P ratio in Holstein was found. 2. Treatment of PMS hormone into 40 cows with the ovarian atrophy and hypoplasia showed emus on average 6.7 days after the final hormone injection. 3. Total 38 cows were conceived by PMS (1000 IU) hormone treatment; 26 cows (68.4%) at the first estrus, 11 cows (30.6%) at the second estrus and the remainder at the third estrus. 4. The above results inducated that treatment of PMS hormone into cows with ovarion hypoplasia and atrophy was effective for inducing estrus and conception.

        제주마의 번식상황과 번식장해 원인에 관한 연구 2 . 제주마의 번식장해상황과 원인에 관한 연구

        In order to determine ways of improving reproductive efficiency 428 heads of Cheju ponies were investigated. The results are as follows; 1. The total occurrence rate of reproductive disorders for Cheju mares is 41.2%. 24.5% of those investigated showed every other year foaling, 16.7% showed other types of reproductive disorders. 2. The total reproductive disorders of ovarian and uterine diseases were 34 heads(10.9%), and 2 heads(0.6%) respectively. 3. Reproductive disorders of ovarian diseases was 89.5%. Those subdivided into ovarian dysfunction(34.2%), persistent corpus luteum(23.7%), ovarian atrophy(15.8%) and ovarian cyst, delayed ovulation(0.7%). respectively. 4. The dilivery disorders of abortion, dystocia and still-birth were 2.9%. 0.3% and 0.6%. respectively. 5. There were no significant differences in R.B.C, W.B.C, Hb. globulin between pregnant and non-pregnant mares, but albumin phosphorus, glucose in pregnant mares were of higher levels than non-pregnant mares. Generally albumin, Ca, glucose were of lower levels than those reported in other research.

        제주마의 번식상황과 번식장해 원인에 관한 연구 1 . 제주마의 번식상황에 대한 조사 연구

        The reproductive status of 428 heads of Cheju pony mares raised on Cheju island was investigated to improve the reproductive efficiency and to get fundamental data for Cheju pony mares. 1. The average ages at the first estrus and the first foaling were 15.1±1.59 and 38.9±5.48 months, respectively, the differences among the feeding farms were great. 2. The average durations of estrus, estrus cycle, gestation period and foaling interval of mares were 5.7±1.81. 20.8±2.42, 333.9±5.41 and 432.6±134.2 days, respectively. 3. The average parturition rate was 60.9% and the differences among the feeding farms were great: The highest parturition rate was 71.4% for Cheju national monumental pony mares. 4. The reccurrence of estrus after foaling excluding and including alternate year foaling were 21.5±19.0, 114.3±133.84 days, respectively. 5. Seasonal variations of estrous mares were 49.4% goes to summer, 45.2% spring, 3.1%, winter and 2.3% autumn.

        한우 다태분만 ( 多胎分娩 ) 유도를 위한 호르몬 처리수준 결정시험

        This study was carried out to determine the dosage of PMS for increasing the rate of multiple births in Korean native cow. Animals were injected subcutaneously with 50G, 750, 1,000IU PMS, respectively on day 17 of the oestrus cycle followed intravenously by 1000IU HCG at the onset of the next oestrus (Only 1-2 Graffian follicles). The results obtained were as follows; 1. Most. of the cows treated witty 1000IU PMS exhibited normal oestrus behaviour(87.5%) and those treated with 500ICT revealed the poorest oestrus rate (56.6%) among the treatments. 2. Duration from the injection time with PMS to the onset of estrus was 4.8 days and was no significant difference among the treatments of various dosages of PMS. 3. The mean number of matured follicles per cow treated with 1000IU PMS was 1.3.

        호르몬 처리에 의한 한우의 다지분만에 관한 연구

        This study was conducted to investigate the rate of multiple births in Korean native cow treated with subcutaneous injection of 1000IU PMS on day 17 of estrus cycle followed by an intravenous injection of 1000IU HCG on the onset day of estrus. The results obtained were as follows: 1. The rate of normal estrus exhibited was 78.9% in the lst m 2nd parity cows and increased as the parity increased. 2. The mean duration from PMS Treatment to the onset of estrus was 4.3 days grad was no significant difference among tire parities. 3. The mean number of matured follicles per cow treated was 1.2 and was no significant difference among the parities. 4. The rates of multiple birts were 4.8%, 40.0% and 23.9% for 1st to 2nd parity, over 3rd parity and over all, respectively, and were increased as the parities increased.
