        Quercetin extracted from Sophora japonica flower improves growth performance, nutrient digestibility, cecal microbiota, organ indexes, and breast quality in broiler chicks

        Objective: The objective of this study was to evaluate the effects of supplementing quercetin extracted from Sophora japonica flower (QS) to the diet of broiler chicks on their growth performance, apparent nutrient digestibility, cecal microbiota, serum lipid profiles, relative organ weight, and breast muscle quality.Methods: A total of 1,088 1-day-old broiler chicks (mixed sex) were randomly assigned to four groups based on the initial body weight (43.00±0.29 g). The experimental period was 35 days (starter, days 0 to 7; grower, days 7 to 21; finisher, days 21 to 35). There were 17 replicate cages per treatment and 16 birds per cage. Dietary treatments consisted of birds receiving basal diet without quercetin as the control group and treatment groups consisted of birds fed basal diet supplemented with 0.2, 0.4, or 0.6 g/kg QS.Results: With the increase of the QS dosage, body weight gain during days 0 to 7 (p = 0.021), 7 to 21 (p = 0.010), and 1 to 35 (p = 0.045), feed intake during days 0 to 7 (p = 0.037) and 1 to 35 (p = 0.025), apparent dry matter digestibility (p = 0.008), apparent energy retention (p = 0.004), cecal lactic acid bacteria counts (p = 0.023), the relative weight of breast muscle (p = 0.014), pH value from breast muscle (p<0.001), and the water holding capacity of breast muscle (p = 0.012) increased linearly, whereas the drip loss from breast muscle (p = 0.001) decreased linearly.Conclusion: The addition of QS in the diet of broiler chicks had positive effects on the breast muscle yield and breast muscle quality, and improved the dry matter digestibility and energy retention by increasing cecal beneficial bacteria counts, thus improving growth performance.       • Parylene-C-Coated Indium Tin Oxide Electrodes for the Optical- and Electrical-Impedance Characterization of Cells

        <P>The demand for the label-free and real-time monitoring of biological cells for the diagnosis of diseases and the discovery of new drugs is increasing. The electrical-impedance characterization of cells has been used to quantify in real time the behaviours of biological cells adhered on the gold or platinum microelectrodes. In this article, a parylene-C-coated indium tin oxide (ITO) electrode, which has better transparency than the conventional electrodes, and high biocompatibility, was investigated for the optical- and electrical-impedance characterization of the cell growth. The living cells on the fabricated parylene-C-coated ITO electrode were observed via phase contrast microscopy, which produces high-contrast images of the transparent cells without staining. The electrical properties of the cellular parameters were extrapolated through fitting analysis with the designed equivalent circuit to the measured impedance spectra of the cell-covered electrode. From the experimental results, it was proven that the fabricated parylene-C-coated ITO electrode is feasible for the optical- and electrical-impedance characterization of cell growth.</P>

      • Impedance spectroscopy of highly ordered nano-porous electrodes based on Au-AAO (anodic aluminum oxide) structure.

        <P>Electrochemical measurements using the microelectrodes are increasingly utilized for the label-free detection of the small amount of biological materials such as DNA, protein, and cells. However, the interfacial electrode impedance increases and may hinder the detection of weak signals as the size of electrode decreases. To enhance the measurement sensitivity while reducing the electrode size, in this study, microelectrodes employing a nanoporous structure were fabricated and characterized by using electrical impedance spectroscopy. We made the highly ordered honeycomb nanoporous structure of Anodic Aluminum Oxide (AAO) by electrochemical anodizing and formed Au layer on the surface of AAO (Au/AAO) by electroless Au plating method. The electrical characteristics of the fabricated Au/AAO electrodes were evaluated by using de Levie's model derived for the pore electrodes. As a result, the interfacial electrode impedance of the fabricated Au/AAO electrodes was 2-3 order lower than the value of the planar electrodes at frequencies below 1 kHz. It implies this nanoporous electrode could be directly applied to label free detection of biomaterials.</P>

        Silymarin seed extract supplementation enhances the growth performance, meat quality, and nutrients digestibility, and reduces gas emission in broilers

        Objective: A feeding trial was carried out to determine the effect of dietary inclusion of silymarin seed extract on growth performance, nutrient digestibility, excreta microbiota, excreta gas emission, blood profiles, and meat quality in broilers. Methods: A total of 1,088 one-day-old Ross 308 broiler chicks (mixed-sex) with an initial body weight of 42.34±0.82 g, were randomly allocated into 1 of 4 dietary treatments with 17 replicates of 16 chicks per cage and fed a basal diet supplemented with 0%, 0.02%, 0.04%, and 0.06% of silymarin. Results: The inclusion of silymarin supplementation linearly increased the body weight of broilers during days 7 to 21 and 1 to 35 days. On day 35, broilers fed a diet containing graded levels of silymarin supplementation linearly increased the nutrient digestibility of dry matter, gross energy, and nitrogen and cecal Lactobacillus counts (p = 0.038). While silymarin supplement linearly reduced the methyl mercaptans (p = 0.039) and acetic acid (p = 0.007) emission in broilers. No significant effects were observed on the blood profile. Relative weights of organs such as breast muscle, bursa of fabricius were increased (linear effect, p<0.05), water holding capacity was enhanced by increasing the silymarin level from 0% to 0.06%. A linear reduction (p>0.05) in drip loss from meat samples during days 1, 3, 5, and 7 by the addition of graded level of silymarin to the diet. Conclusion: An increasing level of silymarin supplementation to the diet of broiler would be beneficial to enhance growth performance, nutrient digestibility, excreta microflora, blood profile, and meat quality traits.         Fetal Bovine Serum을 포함한 세포 배양액에 담근 Indium Tin Oxide 전극 계면의 전기화학적 특성

        For the biocompatibility test of implantable devices or for the sensitivity evaluation of biomedical sensors, it is required to understand the mechanism of the protein adsorption and the interaction between the adsorbed proteins and cells. In this study, the adsorption of proteins in a cell culture medium with fetal bovine serum onto an indium tin-oxide electrode was characterized by using linear sweep voltammetry and impedance spectroscopy. We immersed the fabricated ITO electrodes in the culture medium for 30, 60, or 90 min, and then measured the electrochemical properties of electrodes with 10 mM $Fe(CN){_6}^{3-/4-}$ and 0.1 M KCl electrolyte. With an increase of contacting time, the anodic peak current was decreased and the charge transfer resistance was increased. However, both parameters were recovered to the values before contact with the medium after the treatment of Trypsin/Ethylenediaminetetraacetic acid hydrolyzing proteins.

        <P>Microneedles have emerged for transdermal monitoring of biomarkers, which are a miniaturized replica of hypodermic needles with length-scales of hundreds of micrometers, with a goal to achieve time-sensitive clinical information for routine point-of-care health monitoring. Transdermal biosensing via microneedles offers remarkable opportunities for moving biosensing technologies from research laboratories to real-field applications and enables development of easy-to-use point-of-care microdevices, minimally invasive, and with minimal-training features that are very attractive for both developed and emerging countries. This would eliminate the need for blood extraction using hypodermic needles and in turn, reduce the related problems such as infections in the patients, sample contaminations, and analysis artifacts. In this review, we provide a general overview of recent progress in microneedle-based sensing research, including: (a) in-vivo microneedle diagnostic systems for glucose monitoring with an emphasis on sensor construction and general health monitoring (b) in-vitro use of microneedle sensors. The main objective of the review is to provide a thorough and critical analysis of recent advances and developments in microneedle research field and to bridge the gap between microneedles and biosensors.
