Fig(Ficus carica L.) which belongs to Moraceae family has been used as digestion promoter, cure for ulcerative inflammation and eruption in Korea. Figs have very short preservation period and can be kept only 1 week even in the refrigeration system. The possibility of wine-making from Korean figs was tested by analyzing the fermentation characteristics, proximate analysis and free sugar contents of wine made of slices with 20% sugar addition. Ethanol fermentation of the figs by Saccharomyces cerevisiae was done rapidly. The ethanol contents, pH, total acidity and sugar content were 12.1%, 3.91, 0.42% and 9.9 °Brix, respectively. The results from the sensory evaluation were as follows. The wine made from the sliced figs were better than that from fig juices in their sensory scores. The wine made with frozen figs was not different from that made with fresh ones. To improve the quality of wine, the addition of dried leaf powder in the concentration of 100 ppm were tried and found it was superior to the one made without leaf.

This study was performed to investigate the effects of peking-duck extracts added with medicinal herbs(DJ) on the intoxication of heavy metals (Hg, Pb, Cd, As) in rats. Sprague-Dawley rats weighing 150±15g, were randomly assigned to 5 groups: basal diet only in normal control group(NCG); basal diet and heavy metals without DJ injection in heavy metal control group(HMC); basal diet, heavy metals and DJ(3㎎/㎖) injection in heavy metal low duck juice group(HMLD), basal diet, heavy metals and DJ(30㎎/㎖) injection in heavy metal middle duck juice group(HMMlD); basal diet, heavy metal and DJ(300㎎/㎖) injection in heavy metal high duck juice group(HMHD). Hg and As was injected by 50ppm and Cd and Pb by 25ppm for 17days. Also DJ oral feeding was conducted for 28days. The result of this study were as follows: Food intake and body weight gain in heavy metal administered groups were lower than those of NCG. Liver, kidney and testis weights were not significantly different among 5 groups. GOT, GPT and BUN activities were significantly reduced in DJ treated groups as compared to HMC. DJ showed the suppressing effect on the accumulation of Hg, Pb and Cd in serum, liver and kidney. Fecal Hg and Cd excretions increased with DJ feeding. The results suggested that DJ may have some protective effects on Hg, Cd and Pb intoxication by reducing the accumulation m tissues and increasing excretion. This study also showed the effective way of using duck-extract and its application to the oriental medicine.
Lipopolysaccharide alleviates liver injury induced by thioacetamide in rats

Lipopolysaccharide (LPS) is the major of outer membrane of gram-negative bacteria and one of the most potent microbial initiators of inflammation. From the previous study showed that exposure to a low dose of LPS renders animals tolerant to a lethal dose of LPS, and protects against the toxicity of various chemicals. However, the effects of LPS treatment in thioacetamide (TA) - induced liver injury remain largely unknown. Liver injury caused by various toxic chemicals such as carbon tetrachloride, alcohol, dimethylnitrosamine. Here, we induced rat liver injury by intraperitoneal injection of TA, a representative hepatic fibrosis inducer. In this study, we investigated the effects of LPS in TA group, LPS group, LPS/TA group and vehicle control group on Sprague-Dawely rats (five rats for each group). All rats at the end of the experiment were sacrificed, and liver and serum were obtained. Serological analysis and hematoxylin and eosin staining showed that LPS/TA co-treatment was associated with decrease of aspartate aminotransferase (AST), alanine transaminase (ALT) and totalbilirubin and fibrosis than in TA-treated rats. RT-PCR showed that the levels of IL-6 and Cox2 mRNA were lower in the liver of LPS/TA-cotreated rats than in TA-treated rats. There were no significant differences ALT, ALP, AST, total-bilirubin, IL-6 and Cox2 between vehicle control and LPS-treated rats. These results imply that LPS/TA cotreatment partially alleviates the TA-induced liver injury of rats by reducing inflammatory response.
Kidney Toxicity Induced by 13 Weeks Exposure to the Fruiting Body of Paecilomyces sinclairii in Rats

Paecilomyces sinclairiis (PS) is known as a functional food or human health supplement. However concerns have been raised about its kidney toxicity. This study was performed to investigate the kidney toxicity of PS by 13 week-oral administration to rats. Blood urea nitrogen (BUN), serum creatinine, and kidney damage biomarkers including beta-2-microglobulin (${\beta}2m$), glutathione S-transferase alpha (GST-${\alpha}$), kidney injury molecule 1 (KIM-1), tissue inhibitor of matrix metalloproteinase 1 (TIMP-1), vascular endothelial growth factor (VEGF), calbindin, clusterin, cystatin C, neutrophil gelatinase-associated lipocalin (NGAL) and osteopontin were measured during or after the treatment of PS. BUN, creatinine and kidney damage biomarkers in serum were not changed by PS. However, kidney cell karyomegaly and tubular hypertrophy were observed dose-dependently with higher severity in males. KIM-1, TIMP-1 and osteopontin in kidney and urine were increased dose dependently in male or at the highest dose in female rats. Increased urinary osteopontin by PS was not recovered at 2 weeks of post-exposure in both genders. Cystatin C in kidney was decreased at all treatment groups but inversely increased in urine. The changes in kidney damage biomarkers were more remarkable in male than female rats. These data indicate that the PS may provoke renal cell damage and glomerular filtration dysfunction in rats with histopathological lesions and change of kidney damage biomarkers in kidney or urine. Kidney and urinary KIM-1 and cystatin C were the most marked indicators, while kidney weight, BUN and creatinine and kidney damage biomarkers in serum were not influenced.

<P>Previously, we found that adiponectin (APN) suppresses IL-2–induced NK cell activation by downregulating the expression of the IFN-γ–inducible TNF-related apoptosis-inducing ligand and Fas ligand. Although the antitumor function of APN has been reported in several types of solid tumors, with few controversial results, no lymphoma studies have been conducted. In this study, we assessed the role of APN in immune cell function, including NK cells, CTLs, and myeloid-derived suppressor cells, in EL4 and B16F10 tumor-bearing APN knockout (KO) mice. We observed attenuated EL4 growth in the APNKO mice. Increased numbers of splenic NK cells and splenic CTLs were identified under naive conditions and EL4-challenged conditions, respectively. In APNKO mice, splenic NK cells showed enhanced cytotoxicity with and without IL-2 stimulation. Additionally, there were decreased levels of myeloid-derived suppressor cell accumulation in the EL4-bearing APNKO mice. Enforced MHC class I expression on B16F10 cells led to attenuated growth of these tumors in APNKO mice. Thus, our results suggest that EL4 regression in APNKO mice is not only due to an enhanced antitumor immune response but also to a high level of MHC class I expression.</P>
차나무 미숙 접합자배 배양에 의한 체세포배 형성 및 식물체 재분화

차나무 종자 및 배의 성숙도, 적합 절편체 및 식물생장조절물질이 배발생적 캘러스 및 체세포배 형성에 미치는 효과를 조사하기 위해 8월부터 10월까지 종자를 채취한 후배를 다양한 식물생장조절물질이 첨가된 MS 배지에 배양하였다. 배발생적 배양은 차나무 미숙접합자 배배양에 의해 시작되었으며 상배축, 하배축 및 전배 등 다양한 절편체를 미숙종자에서 분리하여 0.1 ㎎/L 2,4-D와 1.0 ㎎/L BA, 0.5 ㎎/L NAA 와 1.0 ㎎/L BA가 혼용처리된 MS 배지에 20일간 배양한 다음 동일한 배지에 30일 간격으로 계대배양 하였다. 접합자 배의 발육단계는 배발생적 캘러스 유도 및 배발생에 커다란 영향을 미쳤는데 10월에 채취된 종자내 미숙배 크기가 10-15㎜로 유백색 또는 담황색을 띤 어린 자엽기로 조직배양에 적합하였다. 어린자엽기의 미숙배를 전배, 상배축 및 하배축으로 구분한 결과 전배 및 상배축을 0.1 ㎎/L 2,4-D와 1.0 ㎎/L BA가 첨가된 MS배지에서 배양하여 고빈도(85-89%)의 배발생적 캘러스가 유도되었으며, 자엽기의 미숙배를 0.5 ㎎/L NAA와 1.0 mg/L BA가 첨가된 MS 배지에 배양하여 다량의 체세포배가 직접 형성되었다. In order to investigate the effect of maturity of seed and embryos, optimal explants and plant growth regulators on embryogenic callus induction and somatic embryogenesis, embryos of Camellia sinensis L. sampled from August to October, 2005 were cultured on MS medium containing various plant growth regulators. Embryogenic cultures were initiated from Camellia sinensis L. immature zygotic embryos. Using a variety of different explants, including whole embryo axis, epicotyl and hypocotyl from immature seed were isolated and cultured on modified MS medium with various combination of plant growth regulators.Developmental stage of zygotic embryos had a large influence on embryogenic callus induction and embryogenesis. Seeds sampled at October, whitish yellow or slight yellow color of embryos (10-15㎜ size) optimized for tissue culture. Cotyledonary embryo stage optimized for embryogenic callus induction and somatic embryosgenesis.The greatest frequency (85-89%) of embryogenic callus were induced from whole embryo and epicotyl callus cultures on MS medium with 0.1 ㎎/L 2,4-D plus 1.0 mg/L BA. On the other hand, plenty of somatic embryos were obtained directly by culturing cotyledonary stage of immature embryos on MS media with 0.5 ㎎/L NAA plus 1.0 ㎎/L BA.

In order to investigate the effect of the scion and plant growth regulators on in vitro propagation of Camellia sinensis L., young shoots from in vitro seedlings were used. Explants selected from the first node with apical meristem to the fifth node were cultured on MS medium with BA, 2iP, TDZ, zeatin, or GA3 alone. Scion explant affected shoot differentiation and elongation, and the second or third node with a shoot tip was the optimal size of explant for in vitro cuttings. Shoot and multiple shoot initiation were initiated from the node with axillary bud explant on MS medium supplemented with 1.0 mg. L-1 2iP. The combination of NAA and BA, IBA and BA, and 2,4-D and BA were the best to increase callus formation. Shoot and multiple shoots were produced on a combination of MS medium with 2,4-D and 2iP, IBA and 2iP, and IBA and BA. Especially, 32.9-40.2% of shoot and multiple shoots were formed on MS medium with 0.5 mg. L-1 IBA and 2.0 mg. L-1 2iP. White and rigid roots were produced on MS medium with 3.0-5.0 mg. L-1 IBA alone. During the culture, shoot regeneration was not observed through organogenesis or embryogenesis from callus. This result indicates that the second or third nodal region with an axillary bud can be used for mass propagation of Camellia sinensis L. The MS medium with 1.0 to 2.0 mg. L-1 2iP alone or the combination with 0.5 mg. L-1 2,4-D, NAA, or IBA demonstrated the possibility of rapid propagation.
공동주거에 영향을 미칠 수 있는 초기노년기 생리적 습관에 관한 연구

This study has been performed to investigate lifestyles of the 212 younger elderly aged 50 to 75, sampled in Choong-Nam province in 2000. In this study there are two areas of the life styles that may influence congregate residence in later life, questionnaired among the subjects; extrovertive life style and physiological habits. The conclusions are drawn as follows; 1) Among the early elderly, urban residents are more likely to out well -dressed and encounter visitors than the rural counterparts. 2) The younger elderly in 50`s are more like to order designed costumes, purchase ordinary clothes, receive intimate assistance for clothing, and go to hair shops, particularly with higher chance of hair styling efforts among the women and costume advice among the urban subjects. 3) As for physiological habits, the younger urban women tend to have higher frequencies of sanitary activities such as bathing, tooth brushing, and showering. 4) The older female groups are more likely to have meals alone, but there are some younger subjects who fast breakfast. 5) Younger and naval groups are more regularly scheduled for eating lunches, while younger urban groups tend to have supper and go to bed late. Overall in this study, it is proposed that when managing congregate residences, residence control and schedule designs be separately guided among the subgroup of age, sex, and residence.
