        안전한 화장품 개발을 위한 동물실험대체법 연구 동향

        The AMOREPACIFIC Corp., a leading cosmetic company in South Korea, has been making an effort to use variety of alternative methods for many years. We have been making use of alternative methods on assessing phototoxicity of fragrances, corrosivity of hair care products, and ocular irritancy of eye creams and eye makeup products. In addition to existing protocols, there are also several trials on improving and applying current alternative methods. We have intended to apply the HET-CAM (Hen`s egg test on the chorioallantoic membrane) test to various studies on safety assessment of cosmetic products prior to ophthalmology test or anti-irritant material screening. A battery test was introduced for the purpose of improving the accuracy of in vitro phototoxicity method. We also make effort to get suitable alternative methods for predicting sensitizing potency of cosmetic ingredients and products. The one of the strong candidates is h-CLAT (human cell line activation study) and which is now under examination of its accuracy and utility.

        연구논문 : 안점막자극시험의 대체시험법으로서의 CAM-TBS assay 소개

        The hen`s egg test-chorioallantoic membrane (HET-CAM) assay, which employs the vascular membrane of a fertile hen`s egg, has been actively studied by several countries as an alternative method to the Draize rabbit eye irritation test. However, the scoring system of the original HET-CAM test is not satisfactory because of its lack of objectivity and quantitativeness. Moreover, the test required skill when the effects of test substance are to be regarded. To overcome these disadvantages, Hagino et al. developed and objective evaluation technique using the amount of trypan blue absorbed at the site of treatment as an indicator of injury to the CAM (Chorioallantoic membrane-trypan blue staining, CAM-TBS). In this study, we evaluate the CAM-TBS methods and used three materials (1%, 2%, 5% SLS, tween 20 and PBS) to evaluate the correlation of CAMTBS with HET-CAM test. The amounts of trypan blue adsorbed with the CAM showed a good correlation with the HET-CAM test score(r=0.958). According to these results, we suppose that the trypan blue staining assay using CAM-TBS may be a good method to replace the HET-CAM test as an alternative to Draize rabbit eye irritation test. Despite our results, additional studies with more materials are necessary to validate this method.

      • Validation of HET-CAM assay as alternative methods to the Draize rabbit eye test

        One of the most important biological properties of cosmetics, and also of many raw materials, is the local compatibility to mucous membrane. First of all, the assessment of eye irritation is very important in cosmetics industries. Until now standardization in vivo tests are accepted by public health authorities as valid to estimate the irritation potential of chemicals and suitable for the risk assessment. However, the controversial discussion on animal tests, and particularly on the Draize rabbit eye test, is increasing in the public and scientific domain. Efforts have been made to validate proper and suitable in vitro tests in international cosmetics industries during the last decade. One of the most important in vitro tests is the HET-CAM assay. HET-CAM assays have become widely used to assess the irritation potential of test substances. In this paper, hen`s egg-chorioallantoic membranes were used to screen for and assess eye irritation properties of surfactants, chemicals and cosmetics, in connection with Draize rabbit eye test. The purposes of this paper are validation and verification of HET-CAM assay as alternative methods to the Draize rabbit eye test. We decided to use endpoint assessment (EPA) for prediction model and found out saline, squalane, 10% ethanol and eutanol G as suitable vehicles for both HET-CAM assay and Draize eye test. The Pearson correlation of HET-CAM assay and Draize eye test with 5 chemicals, 8 surfactants and 9 cosmetics was 0.894(p<0.001). The correlation was higher (0.992, p<0.001) in cosmetics than others. After considering all the results on Pearson correlation, we suppose that the HET-CAM assay is a useful tool as alternative method to the Draize eye test for safety assessment of cosmetics.

        Customers have used various skin care and functional cosmetics to prevent their skin aging as well as improve current condition of the skin. It is important to analyzing and understanding their skin to select optimized skin care solutions. The aim of this study is to quantitatively analyze the effect of environment, lifestyle, and innate genes on the current skin condition. For this purpose, degree of hydration, sebum, wrinkles, melanin, dullness and redness in a highly controlled condition were collected in conjunction with a questionnaire survey analyzing their lifestyles and genetic data from about 3000 women. We classified participants based on the types of skin, using 6 kinds of index representing skin properties, questionnaire survey on lifestyle and genetic data with Gaussian Mixture Model and Decision Tree for classification model. Through this study, it was possible to divide the skin of Korean women into 12 clusters according to wrinkles, melanin, redness, dullness and oil/moisture balance with Gaussian Mixture Model. Also, we were able to identify a pattern in which each factor was correlated. Next, we tried to discover factors for predicting skin condition changes through correlation analysis with lifestyle, climate/environment, and innate genes that affect the current skin condition. Ultimate purpose of this study is to predict future skin condition using big data and AI technology. This knowledge will enable us to provide more proactive and personalized solutions not only for cosmetics but also for life care such as lifestyle, eating habits, and environmental response.

        토끼 안점막자극 대체시험법으로서의 HET-CAM assay 소개

        The hen`s egg test-chorioallantoic membrane(HET-CAM) assay, which employs the vascular membrane of a fertile hen`s egg, has been actively studied by several countries as an alternative method to the Draize rabbit eye irritation test. There are two methods to evaluate the eye irritation potential, reaction time method(RTM) and end point assessment(EPA), between which can be chosen as the apt method depending on the characteristics of test material. In this study, we employed the EPA method to compare with the Draize rabbit eye irritation test method and then calculated the Pearson correlation coefficient value. The Result of Pearson correlation was high(0.986, p=0.002) with 5 test materials: 3 raw materials(IPMS 03, VCP-IS-2Na, Seletinoid: Amorepacific R&D center, Korea), 1 commercial cleansing form(Amorepacific R&D center, Korea) and 1 surfactant (Amisoft LS-11, Ajinomoto, Japan). According to these results, we suppose that the HET-CAM assay is a good method to replace the Draize eye irritation test as the tests of 5 materials have proven. Despite our results, additional studies with more materials are necessary to validate this method as an alternative to the Draize rabbit eye irritation test method.
