      국문 초록 (Abstract) kakao i 다국어 번역

      배경 : 우리나라 고령화 사회 지표는 2060년에는 세계 2위 수준으로 높아질
      것으로 예측되고 있다. 노인성 질환 가운데 뇌혈관 질환은 2019년 한국의
      사망원인통계에서 4위를 차지하고 있으며 성인에서 발생하는 후천적 장애의
      가장 흔한 원인 질환이 된다. 大黃黃連瀉心湯은 大黃과 黃連 두 가지 한약
      으로 구성된 한방 처방으로 항혈전, 항지질, 항염증, 항산화, 항암효과가 보
      고되었으며 알츠하이머, 혈관성 치매, 뇌혈관질환에 사용되고 있다. 본 연구
      는 허혈성 뇌손상에서 大黃黃連瀉心湯 후처치의 항염증 기전을 확인하며,
      인지기능 개선 효능을 확인하기 위하여 이루어졌다.
      재료 및 방법 : C57BL/6 수컷 생쥐를 5마리씩 무작위로 4개의 그룹(Sham
      군, 양측 총경동맥 협착(BCAS) 수술군, 大黃黃連瀉心湯을 각각 5, 50 ml/kg
      농도로 투여한 실험군)으로 나눈다. 양측 총경동맥 협착에 의해 만성적인 뇌
      혈류 저하 상태를 유발하였으며 1주 후부터 大黃黃連瀉心湯 추출물을 저농
      도, 고농도로 구별하여 4주간, 1주에 5일 경구 투여한 다음 학습 및 기억의
      행동측정을 실시하였다. 또한 해마 CA1 영역에서 신경 세포의 형태적 변화
      를 관찰하고, western blot을 통해 염증 관련 인자들을 관찰하였다.
      결과 : 고농도의 大黃黃連瀉心湯을 후처치한 군에서 apoptosis 관련 단백질
      의 발현이 감소하였으며 해마 CA1 영역에서 신경 세포의 손상이 억제되었
      다. 또한 인지기능 개선 효과가 확인되었다.
      결론 : 저농도의 大黃黃連瀉心湯을 후처치한 군에서는 대조군과 비교하였을
      때 유의미한 효과 없었으나, 고농도의 大黃黃連瀉心湯을 후처치한 군에서는
      인지기능개선 효과를 관찰할 수 있었으며, 위와 같은 효과에는 항염증 및
      항세포자멸 경로와 관련이 있는 것으로 확인되었다.

      Background : Korea's aging society index is predicted to rise to the
      second highest level in the world by 2060. Among geriatric diseases,
      cerebrovascular disease ranks 4th in Korea's 2019 cause of death
      statistics and is the most common cause of acquired disorders in adults.
      DHST is a herbal prescription consisting of two herbal medicines, Rhei
      Rhizoma and Coptidis Rhizoma, and has been reported to have
      antithrombotic, antilipid, anti-inflammatory, anti-oxidant, and anticancer
      effects. This study was conducted to confirm the anti-inflammatory
      mechanism of post-treatment of DHST in ischemic brain injury and to
      confirm the cognitive function improvement effect.
      Materials and Methods : C57BL/6 male mice were randomly divided
      into 4 groups (Sham operation group, bilateral common carotid artery
      stenosis (BCAS) control group, and experimental group administered with
      DHST at concentrations of 5 and 50 ml/kg) and each group consisted of
      5 mice. BCAS caused chronic cerebral blood flow decrease. From 1 week
      later, DHST was divided into low and high concentrations and
      administered orally for 4 weeks, 5 days a week, and then behavioral
      measurement of learning and memory was carried out. In addition,
      morphological changes of neurons in the CA1 region of the hippocampus
      were observed, and inflammation-related factors were observed through
      western blot.
      Results : In the H-DHST group, the expression of apoptosis-related
      proteins was reduced, and neuronal damage was suppressed in the
      hippocampal CA1 region. In addition, the effect of improving cognitive
      function was confirmed.
      Conclusion : In the L-DHST group, there was no significant effect
      compared to the control group. but in the H-DHST group, a cognitive
      improvement effect was observed, and it was confirmed that the above
      effect was related to the anti-inflammatory and anti-apoptotic pathways.
