    Transient receptor potential canonical (TRPC) channels are nonselective cation channels that shape excitability and signaling across neuronal, cardiovascular, and visceral tissues. In native systems, TRPC1/4/5 family members are frequently expressed as heteromeric assemblies rather than isolated homomers, suggesting that assembly-dependent gating rules—not single-subunit properties—contribute to physiological channel identity. We therefore hypothesized that incorporation of TRPC1 into TRPC4/5-rich tetramers reconfigures intracellular ion coupling and GPCR/G-protein control through an intrinsically asymmetric heteromeric architecture.
    We established expression conditions that favor heteromer formation and used concatemeric constructs with defined subunit arrangements to compare TRPC5 homomers with TRPC1 containing channels. Whole cell recordings showed a clear shift in ion dependence: TRPC5 homomers were strongly potentiated by intracellular Ca²⁺ and showed little response to intracellular Na⁺, whereas TRPC1–5 assemblies were activated by intracellular Na⁺ in the tens-of-millimolar range and exhibited weak Ca²⁺ dependent potentiation. A TRPC1–5/TRPC4–4 assembly retained Na⁺ dependent gating, but exhibited altered current–voltage relationships and modified sensitivity to pharmacological modulators, indicating that TRPC4 quantitatively tunes, rather than eliminates, TRPC1 dependent Na⁺ responsiveness.
    To mechanistically interpret this assembly-dependent ion coupling, we interpreted our electrophysiological data in the context of available cryo-EM structures and our structure-informed mutational analyses. Rather than invoking a discrete intersubunit Na⁺ binding pocket, our results are most consistent with an allosteric model in which TRPC1 incorporation remodels intracellular coupling centered on the S2–S3 linker. This TRPC1-dependent coupling architecture differs from TRPC5 in a way that supports low affinity Na⁺ dependent modulation of gating while attenuating the Ca²⁺ dependent potentiation characteristic of TRPC5 homomers. Consistent with separable contributions of coupling versus permeation, pore-domain substitutions in TRPC1 altered conductance and activation profiles toward more TRPC5-like behavior without abolishing heteromeric expression.
    Finally, we show that regulation of TRPC1 containing assemblies is strongly G protein subtype dependent. Constitutively active Gαi2 enhanced TRPC1–5 currents, whereas Gαq suppressed them, supporting a model in which heteromeric TRPC assemblies can act as direct G protein effectors in a manner determined by subunit composition. Together, these findings identify TRPC1 as an assembly dependent tuning subunit that converts TRPC4/5 channel logic from Ca²⁺ biased potentiation to Na⁺ responsive gating while imposing subtype selective G protein modulation. This dissertation provides a mechanistic framework for how TRPC heteromers integrate ionic homeostasis with receptor-coupled signaling, with implications for excitable cell physiology and therapeutic targeting of TRPC1/4/5 assemblies.
