    Small RNA 는 대표적인 non-coding RNA 의 한 종류이며, 식물에서는 hairpin RNA (hpRNA) 와 double-stranded RNA (dsRNA)로부터 20-30개의 염기로 이루어진 micro RNA (miRNA) 와 small interfering RNA (siRNA) 가 형성되어 염기서열 상보성에 의한 유전자 발현 억제 현상인 RNA 간섭 현상 (RNA interference; RNAi) 을 일으킨다. 한 세포에서 발생된 small RNA 는 세포 및 조직간 연결망을 통해 광범위하게 이동하며, 이러한 특성을 통해 식물의 개화, 발달, 다양한 스트레스에서 식물의 반응을 조절하는 신호물질로써 중요한 역할을 수행한다. 오늘날 숙주유래 유전자발현억제 (host-induced gene silencing; HIGS) 현상으로 규정되는 식물 small RNA 이동과 병원체의 RNAi 현상은 기존 저항성 연구의 주요 한계점인 저항성 개체의 높은 출현빈도와 저항성 유전자원의 도입 및 고정에 드는 시간과 노동력을 획기적으로 절감시킬 새로운 저항성 개체 개발 전략으로 주목 받고 있다. 이미 미국과 유럽, 중국등의 주요 농업 생산국에서는 주요 식량 및 환금작물들을 대상으로 그 제작방법들과, 환경위해성 평가에 관한 연구들도 활발히 이루어지고 있다. 따라서 본 연구는 벼와 벼 도열병 곰팡이의 HIGS 현상을 규명하고 이를 응용하여 벼의 도열병 곰팡이 저항성을 개선할 수 있는지를 평가하고자 하였다. 첫번째로, 벼와 도열병 곰팡이 간의 HIGS 현상을 규명하였다. small RNA는 소포체 (vesicle) 의 내·외식세포 작용 (endo- and exocytosis) 를 통해 이동한다는 기 연구결과에 근거하여, 벼의 small RNA도 도열병 곰팡이와의 상호작용이 가능한 포자의 부착과 발아 (1 단계), 세포 침투체 형성 (2 단계), 균사체 형성 (3 단계) 과정에서 이동할 수 있다는 가설을 수립하였다. 다음으로 유전자 발현 검증용 형광 단백질인 enhanced Green Fluorescent Protein (eGFP)를 발현하는 형질전환 벼 도열병 곰팡이 계통인 KJ201::eGFP 와 eGFP의 siRNA를 발현하는 형질전환 벼인 35S::dsRNAi_eGFP 벼와의 병원성 상호작용을 관찰하였다. 그 결과, 벼 세포의 표면과 내부에 형성된 대부분의 곰팡이 병원체 구조들이 대조군에 비해 현저히 낮은 eGFP 발현을 나타냄을 정량적, 정성적으로 확인하였다. 이를 통해 HIGS가 벼와 도열병 곰팡이의 상호작용에서도 일반적인 현상임을 확인하였다. 다음으로, 도열병 곰팡이의 병원체 발달 단계를 조절하는 RGS1 (포자의 부착과 발아, 1 단계), MgAPT2 (세포 침투체 형성, 2 단계), LHS1 (균사체 형성, 3 단계)의 HIGS가 벼와 도열병 곰팡이의 상호작용을 어떻게 조절하는지 검증하였다. 먼저, 비 특이적 유전자 발현 억제 효과 (off-target effect)가 가장 적은 cDNA 염기서열 부분을 in silico 방식으로 추출하였고, 각 유전자들의 35S::dsRNAi 벡터를 제작하여 각 곰팡이 병원성 유전자에 특이적인 siRNA를 발현하는 형질전환 벼를 제작하였다. 각 형질전환 벼의 잎과 뿌리, 전신감염 (systemic infection)에 대한 결과는 HIGS 현상이 병 진행의 후반부 조절자일수록 더 강하며, 1단계 조절 유전자인 RGS1의 발현감소는 오히려 저항성을 감소시켰고, 2, 3 단계 조절 유전자인 MgAPT2 와 LHS1 의 발현 감소는 감소량에 비례하는 뚜렷한 저항성 증가가 확인되었다. 마지막으로, 곰팡이 병원성 유전자의 HIGS에 의한 벼와 도열병 곰팡이의 상호작용 변화를 구체적으로 검증하였다. 35S::dsRNAi_MgAPT2 및35S::dsRNAi_ LHS1 벼의 감염된 세포들을 현미경으로 관찰한 결과들은 곰팡이의 발달이 결손 돌연변이체(deletion mutant) 곰팡이(ΔMgapt2, Δlhs1)와 유사했지만, 저항성 granule의 전신적 형성 (systemic accumulation)과 벼의 주요 저항성유전자인 OsPR1a와 OsPR10의 독특한 발현 변화는 HIGS를 통해 벼가 새로운 방식의 저항성을 획득할 수 있음을 보여 주었다. 또한 35S::dsRNAi_RGS1 벼의 저항성 감소는 벼의 저항성 발현이 억제되고, 곰팡이 균사체의 (Invasive hyphae; IH) 발달 및 세포침투(penetration) 능력의 증가가 원인임을 알 수 있었으며, 기존의 ΔMorgs1 곰팡이 연구에서 제시된 병원성 증가 가능성을 HIGS를 통해 증명하였다. 본 연구를 통하여 HIGS는 벼에서도 일반적인 현상이며, 벼의 도열병 저항성 개선을 위한 HIGS의 응용방법을 유전자원의 선별법, 벡터 제작법 및 저항성 검증법으로 나누어 구체적으로 제시하였다. 또한, HIGS는 기존의 보고된 방법과 다른 방식으로 식물-곰팡이 상호작용을 조절함을 구체적으로 증명하여 HIGS기반의 새로운 곰팡이 유전자 기능 연구법을 제시하였다. 본 연구결과들의 지속적인 응용을 통해 향후 HIGS의 농업적, 생물학적 활용을 위한 제반기술들을 선도적으로 확보할 수 있을 것으로 사료된다.

    In plants, small RNAs including microRNAs (miRNAs) and small interfering RNAs (siRNAs) play crucial roles in RNA interference (RNAi) in a cell-autonomous and non-cell-autonomous manner. Recently, host-induced gene silencing (HIGS) has been applied for the successful control of fungal pathogens in various crops. Although, the study of HIGS between rice and rice blast fungus, which is the main biological causal agent of global rice yield, is still elusive in major producing countries of Asia. Encouraged by this inactivity, in this study, HIGS of rice blast fungus, Magnaporthe oryzae, and novel functions of fungal pathogenic genes were suggested. First, using eGFP gene and 35S::dsRNAi vector system, a cytological system to verify HIGS between rice and rice blast fungus was developed and the HIGS of M.oryzae was successfully established. Next, to apply HIGS to the enhancement of rice resistance against M.oryzae, fungal pathogenic genes specifically regulating the main infection stages, such as RGS1, MgAPT2, and LHS1 were selected. HIGS of MgAPT2, and LHS1, which have critical roles in appressorial penetration and hyphal development, respectively, successfully changed compatible interaction between rice plants and M. oryzae to incompatible, resulting in the enhanced tolerance of rice plants to rice blast fungus. In contrast, HIGS of RGS1, regulating conidium germination and appressorium initiation enhanced fungal pathogenicity, which was antagonistic to a previous study conducted with ΔMorgs1 mutant fungus. Moreover, the molecular mechanisms underlying the effects of HIGS of RGS1, MgAPT2, and LHS1 genes on interaction between rice plants and M. oryzae was also studied. Different patterns of granulation, callose deposition, and ROS generation in 35S::dsRNAi rice plants during blast infection suggest novel roles of the three pathogenic genes related to modulation of host defense system. In sum, this study suggests not only the optimal target genes for efficient HIGS of the rice blast fungus to protect whole rice plants from rice blast infection, but also a novel aspect in studying functions of fungal pathogenic genes during interaction between rice and rice blast fungus.
