    Humanized mice are expected to be a useful tool for studying human immune system and human diseases. Since NOD/SCID/c-/- (NSG) mouse has been developed, more powerful host mouse than NOD/SCID mouse with the reconstitution of human cells, many groups has been published their works and applied their humanized mice to various human diseases. In many groups, the humanized mice were designed by the method of transplantation of human umbilical cord blood-derived CD34+ hematopoietic stem cells. Commonly in such cases, the amounts of human T cells among engrafted human cells in humanized mice were so small, while the amounts of human B cells were abundant.
    To improve this limitation and make advanced humanized mice model, we transplanted human fetal liver, thymus tissue under kidney capsule and human CD34+ stem cells intravenously into NSG mice. Our humanized mice were reconstituted with sufficient number of human T cells. With this humanized mouse, we investigated whether the entire process of human T cell developments were arose similarly as reported human data in chapter 1. Our humanized mice transplanted with fetal liver and thymus tissue and fetal liver-derived CD34+ stem cells (FLT+FLCD34 mice) showed higher levels of human cells and T cells in their periphery and various organs more than those of humanized mice transplanted fetal liver-derived CD34+ stem cells only (FLCD34 mice). In enlarged transplanted thymus tissue under kidney of FLT+FLCD34 mice, human populations of thymus seeding progenitors (TSPs), early thymic progenitors (ETPs), and pre-T cells were identified in CD4 and CD8 double negative (DN) thymocytes, and CD3-CD4+ immature single positive (ISP), CD3-CD4+CD8+ early double positive (EDP), CD3+CD4+CD8+ double positive (DP), CD3+ CD4+ single positive (SP) and CD3+ CD8+ SP populations were identified in thymocytes excluded DN population. In the human T cells of periphery, a notable difference was founded in their character. FLT+FLCD34 mice have much CD45RA+ naïve T cells than CD45RO+ memory T cells. On the contrary to this, FLCD34 mice have much more CD45RO+ memory T cells rather than CD45RA+ naïve T cells. These memory T cells from spleen of FLCD34 mice were proliferated rapidly ex vivo under stimulation of anti-CD3 and anti-CD28 and showed drastic induced cell death. We found out that this difference was resulted from the thymic environments, because the human T cells of FLCD34 mice were educated by mouse MHC class II in mouse thymus. These T cells showed the properties of innate T cells expressing PLZF (promyelocytic leukemia zinc finger protein) in their thymocytes and expressed intracellular interferon- (IFN-) within a short time.
    In chapter 2, the chimerism and the tolerance induction in the humanized mice reconstituted with fetal liver and thymus tissue and HLA-mismatched umbilical cord blood-derived CD34+ stem cells (FLT+UCB-CD34 mice) was confirmed. Both HLA-A2+ and HLA-A24+ T cells were developed from implanted human fetal thymus tissue and released to periphery. Both mature T cells didn’t show cytotoxic responses against cord blood originated target cells ex vivo. Also, the tolerance was confirmed against the fetal origin by skin graft assay. Fetal donor originated skin graft did not cause graft rejection in FLT+FLCD34 mice while identical skin grafts caused rejection in UCB-CD34 mice as expected. This method could be useful establishing a patient-specific humanized mice model using co-transplantation of allogeneic human fetal tissues with patient’s stem cells.
    In these finding of chapter 1 and chapter 2, an enhanced human T cell reconstitution occurs normally in our humanized mice model compared to other humanized models without the implantation of human fetal tissue. Also, these developed human T cells educated by human MHC class II in the implanted thymus showed more human like features in their function and phenotypes in the periphery without showing any feature of innate T cells. Therefore, our humanized mice model is expected to be a useful model for human T cell research, development of patient’s specific cell therapy and applied to preclinical diagnostic screening system.

Since NOD/SCID/c-/- (NSG) mouse has been developed, more powerful host mouse than NOD/SCID mouse with the reconstitution of human cells, many groups has been published their works and applied their humanized mice to various human diseases.     인간화 마우스는 인간의 면역체계와 질병연구를 위한 유용한 수단으로 대두되고 있다. 특히 인간세포의 재건에 있어서 주로 쓰이던 종전 동물모델인 NOD/SCID보다 훨씬 강력한 NOD/SCID/c-/- (NSG) 마우스가 개발된 이후로 많은 연구자들이 그들의 결과를 앞다투어 발표하고 여러가지 인간의 질병에 적용하고 있다. 대부분의 많은 연구자들이 인간의 제대혈에서 분리한 CD34+ 조혈모세포를 이식하는 방법으로 인간화 마우스를 모델링 하고 있는데, 그런 경우에 대부분의 인간화 마우스는 인간의 B 세포가 재건된 인간세포의 대부분을 차지하며 T세포는 매우 작은 양만이 재건되는 공통적인 결과들을 보였다.
    이 결점을 보완한 인간화 마우스를 만들기 위해, 인간 태아의 간과 흉선조직을 NSG 마우스의 신장막에 이식하고 미정맥에 CD34+ 조혈모세포를 이식하였다. Chapter 1에서는 이렇게 제작한 인간화 마우스는 T세포가 충분히 재건되고 이 세포들이 인간의 T세포 발달의 전 과정을 거치는지 보고된 데이터와 비교하여 확인하였다. 인간 태아의 간과 흉선조직과 태아 간에서 분리한 CD34+ 조혈모세포가 동시에 이식된 인간화 마우스(FLT+FLCD34)는 오직 간에서 분리한 CD34+ 조혈모세포만 이식된 마우스(FLCD34)에 비하여 말초혈액과 여러 기관 내에 많은 양의 인간세포와 T세포를 지니고 있었다. 일정시간이 지난 후 이식했던 태아의 흉선조직이 매우 비대해졌는데 그 안에서 흉선세포를 분리하여 가장 미성숙한 세포인 thymus seeding progenitor (TSP), early thymic progenitor (ETP), Pre-T 세포를 CD4와 CD8이 음성인 DN 흉선세포에서 확인하고, CD3-CD4+ immature single positive (ISP), CD3-CD4+CD8+ early double positive (EDP), CD3+CD4+CD8+ double positive (DP), CD3+ CD4+ single positive (SP), CD3+ CD8+ SP 세포군을 DN을 제외한 나머지 세포들에서 확인하였다. 말초혈액에 생성된 T세포에서 FLT+FLCD34 마우스와 FLCD34 마우스 간의 눈에 띄는 차이점이 발견되었는데, FLT+CD34 마우스의 T세포에는 많은 양의 CD45RA+인 naïve T세포들이 존재하고 소량의 CD45RO+인 memory T세포들이 존재하는 반면, FLCD34 마우스에는 대부분의 T세포들이 CD45RO+인 memory 타입으로 존재하였다. FLCD34 마우스의 비장세포를 체외에서 anti-CD3와 anti-CD28로 자극하여 배양하였더니 빠르게 분열하는 것을 관찰할 수 있었으며 동시에 급격한 세포사멸에 빠지는 것이 관찰되었다. 우리는 FLCD34 마우스의 T세포는 마우스 자신의 흉선 밖에 없기 때문에 마우스의 MHC class II에 의해 교육받게 되므로 앞에서 나타난 차이점은 두 마우스의 흉선환경에서 비롯된다는 것을 알게 되었다. 그러한 T세포들은 PLZF (promyelocytic leukemia zinc finger protein)를 발현하며 빠르게 IFN-를 분비하여 innate T 세포의 특성들을 지닌다는 것을 보여주었다.
