    인체의 기관 중 가장 바깥에서 다양한 외부요인으로부터 내부를 보호하는 첫 번째 관문으로 알려져 있는 피부는, 외부로부터의 변화에 항상 동일한 상태, 즉 항상성을 유지해야만 하는 기관으로 잘 알려져 있다. 피부는 매우 다양한 세포들로 구성되어 있는데, 가장 외부를 담당하는 각질형성세포(keratinocyte)부터, 피부의 구조를 담당하는 섬유아세포(fibroblast), 멜라닌을 합성 후 최외각 각질형성 세포로 전달하는 미백세포(mealnocyte), 그리고 지방세포로 구성된 피하지방층(subcutaneous fat) 등이 존재하는 것으로 보고되어 있다. 결국, 피부는 각각의 개별 기능을 가진 다양한 세포들이 적절한 구성비와 특별한 조건을 가지고 인체의 가장 외각에서 보호자 역할을 담당하고 있다.

    앞선 두 가지 연구 결과를 고려한 결과, 피부 항상성을 유지하기 위한 전략은 피부를 구성하는 각 세포의 특성을 파악함과 더불어 다양한 내,외부 요인들간의 관계를 명확히 규명하는 것이 선행되어야 한다고 판단된다. 따라서, 본 연구에서 언급하는 인체 피부항상성 유지를 위한 두 가지 전략은 임상학적 접근의 매력이 있음에도 불구하고, 아직까지는 관련된 더 많은 연구가 선행되어야만 한다. 특히, 안전성 및 정확도가 확보되고 이를 적용하기 위한 기준을 밝혀야만 본 연구는 보다 가치가 있을 것으로 판단된다.

    The skin is the first line of defense protecting our body from various external factors (e.g., pathogens, excessive water loss) and it has a crucial role in maintaining homeostasis to prevent fluctuations caused by environmental changes outside of the body. The skin consists of various layers, which include many types of cells such as keratinocytes (KCs), fibroblasts (FBs), melanocytes (MCs) and subcutaneous fats (SCFs). In spite of their different functions, these cells can communicate and orchestrate complementarily to regulate homeostasis. For example, melanin is first produced by endo/-exogenous factors and then transported from MCs to KCs, resulting in the prevention of DNA damage in KCs through the absorption of ultraviolet (UV). However, excess amounts of melanin, which is mainly associated with most pigment disorders (e.g., hyperpigmentation) is aesthetically unpleasant and causes melanoma, skin cancer. In particular, excess amounts of melanin derived from endogenous factors can impair the functions of MCs according to previous literatures. However, fundamental causes have not been elucidated.
    The first chapter focuses on hyperpigmentation and –related mechanisms mainly induced by endogenous factors such as substance P (SP) in MCs. SP is well-known for triggering the initial stage of wound healing and it is also a causative substance for psychological stress. It has been reported that abnormal pigmentation (e.g., hyper- or hypopigmentation) often occurs in injury lesions or areas surrounding the human skin during the wound healing process. However, the mechanisms underlying imbalanced pigmentations have not been elucidated. Therefore, I first attempted to investigate the mechanism of SP-related pigmentation including hyperpigmentation and concomitant signaling pathways in MCs and devised a strategy to maintain homeostasis of the skin. To do so, MCs were treated with SP and the underlying mechanisms of SP related pigmentation were investigated. Based on our results, I found that SP receptors (neurokinin 1 receptor, NK1R) were expressed on MCs and verified that melanin synthesis was promoted and regulated by NK1R receptor-dependent down-stream signaling pathways. These results showed that SP-NK1R-endothelin 1 (EDN1)-endothelin receptor type B (EDNRB) signaling can induce skin darkening through promoting melanin biosynthesis, which is much faster than the normal signal delivery system (e.g., stress conditions) by inducing SP.
    In the second chapter, we discussed about controlling the lipolysis process using visible radiation in SCFs, which exists under the dermis layer of the skin. Until recently, studies on fat have focused on visceral fat research, and limited studies were conducted on SCFs. However, recent reports on SCFs highlighted the importance of maintenance of bulge stem cells during the hair growth process in human scalp, while stem cell research took place in the spotlight concurrently. Additionally, further investigative studies revealed that there is different interaction between the internal mechanism and its function from further SCFs investigation studies. The SCFs are not only responsible for the formation of fat with age, but also causes dyslipidemia accumulation/formation of cellulite, which is well known causative factor for asymmetry in the shape and related dysfunction of the skin. Although a variety of physical procedures, including laser therapy to eliminate the cellulite has been developed, new approaches are required because significant limitations and adverse side effects were reported. In this chapter, I confirmed that a specific wavelength in the visible radiation (VR) can induce specific light receptor, rhodopsin, and -related downstream signals can promote cyclic adenosine monophosphate (cAMP)-dependent lipolysis pathway via activation of 3-adrenergic receptor in SCFs under the skin. As a result, we propose a second strategy to maintain the skin's homeostasis through a new method of lipid regulation via the series of relevant mechanisms mentioned above. Based on two previous studies, strategies for maintaining skin homeostasis should be designed by understanding the characteristics of each cell type that constitutes the skin, as well as establishing a clear relationship between various internal and external factors. Therefore, additional studies should still be preceded by a more relevant study, although two strategies for maintaining the homeostasis of human skin refer to the attractive results of the clinical approach. Especially, these studies are deemed to be more valuable whenever safety and accuracy has been confirmed and the criterion for its application is established. Therefore, I expect to pursue a deeper and more efficient skin homeostasis strategy based on these prior studies
