Pain is defined as an unpleasant sensory and emotional experience associated with actual or potential tissue damage. Excessive pain can delay patient recovery, hinder return to society, lower quality of life, and cause economic loss. Especially after surgical procedures, abnormal neural activity and neuroinflammation can lead to persistent pain that further delays recovery. Various strategies including pharmacological, behavioral, and psychological therapies have been suggested to address these issues. Among these, noninvasive repetitive magnetic stimulation has attracted attention as an efficient and promising approach for pain management and functional recovery.
In this study, we established a reliable animal model of neuropathic pain and nerve regeneration by employing the chronic constriction injury (CCI) model of the sciatic nerve, widely used in experimental research. We evaluated the therapeutic effects of repetitive magnetic stimulation on pain modulation and nerve recovery. Magnetic stimulation was applied for 8 weeks, beginning 2 weeks post-sciatic nerve ligation. Sensory and motor recovery were assessed over 10 weeks using the von Frey test and the sciatic functional index (SFI).
Results demonstrated that the magnetic stimulation group exhibited significant improvements in mechanical pain threshold and gait function compared to the non-stimulated group, indicating effective pain relief and functional recovery. Histological examination (H&E and Masson’s Trichrome staining) revealed better-preserved nerve structures and reduced muscle fibrosis in the stimulation group. Immunofluorescence for Neurofilament and Iba1 further confirmed enhanced axonal organization, promoted nerve regeneration, and decreased microglial activation, indicating alleviated inflammation.
Collectively, these findings indicate that repetitive magnetic stimulation promotes nerve regeneration and functional recovery by modulating neuroinflammation and improving nerve and muscle structure. This approach suggests clinical value as a noninvasive neuromodulatory therapy for improving function and relieving pain after chronic nerve injury.
